Circulating angiogenic T cells as potential immune-vascular biomarkers associated with disease activity in axial spondyloarthritis.

OBJECTIVE: Axial spondyloarthritis (axSpA) is a chronic inflammatory autoimmune disease associated with persistent systemic inflammation and endothelial dysfunction. Angiogenic T cells (Tang cells), defined as CD3+CD31+CD184+ T cells, are specialized immune cells involved in vascular repair and endothelial homeostasis, but their clinical relevance in axSpA remains unclear. This study aimed to characterize circulating Tang cells in axSpA and evaluate their potential as immune-vascular biomarkers of disease activity. METHODS: Peripheral blood samples were obtained from patients with axSpA strat

In vivo CRISPR screens identify dual functions of DNMT3A in mediating tumor immune evasion.

Cancer immunotherapy has achieved remarkable clinical efficacy across multiple cancers but remains ineffective for most patients, and the mechanisms underlying resistance remain incompletely understood. Here, we perform in vivo CRISPR-Cas9 screens in female mice and identify DNA methyltransferase 3 A (DNMT3A) as a key driver of tumor immune evasion through both methyltransferase-dependent and -independent mechanisms. DNMT3A represses TNFRSF1A and TNFRSF1B expression, enabling tumor cells to evade TNFα-induced killing. Independently of its methyltransferase activity, DNMT3A interacts with NFYA

Zbtb44-mediated transcriptional repression critically regulates colonic inflammation.

The regulatory mechanisms that maintain colonic immune homeostasis remain incompletely understood. Here we show that genetic deletion of Zbtb44, a previously uncharacterized member of the ZBTB transcription factor family, leads to spontaneous colitis and rectal prolapse. Zbtb44-/- T cells expressed increased IFN-γ, a pro-inflammatory cytokine strongly associated with human inflammatory bowel disease. Mechanistically, Zbtb44 undergoes SUMOylation, which facilitates its interaction with a conserved SUMO-interacting motif in T-bet, enabling binding to the IFN-γ promoter and enhancer to restrain c

Extracellular RNA clearance by non-canonical cell-surface MDA5 protects against lung fibrosis.

The role of innate immune receptors, specifically RNA sensors, in non-pathogen-driven lung fibrosis remains unclear. Here, we demonstrate that the cytosolic RNA sensor MDA5 could translocate to the cell surface upon bleomycin-induced damage. MDA5-deficient mice exhibited exacerbated lung fibrosis with increased IL-17A⁺ γδ T cells and neutrophils, which is independent of the canonical MAVS pathway and type I interferon signaling. Phenotypically, MDA5-deficient lung CD11b⁺ macrophages, which resemble human SPP1⁺ macrophages, showed elevated expression of CCR2 ligands. Mechanistically, cell-surfa

Single-cell transcriptomics reveals transcriptional reprogramming and microenvironmental remodeling in ectopic ACTH/CRH-secreting pheochromocytomas.

BACKGROUND: Ectopic ACTH- and/or CRH-secreting pheochromocytomas (ACTH+CRH+PCCs) are rare tumors that can mimic Cushing's syndrome and complicate the diagnosis and treatment clinically. However, the tumor-intrinsic and microenvironmental features of ACTH+CRH+ PCCs remains poorly understood. METHODS: Fresh tumor specimens from three ACTH+CRH+ and three ACTH- CRH- PCCs were profiled by single-cell RNA sequencing. Developmental trajectories and transcriptional regulatory networks were analyzed to investigate tumor-intrinsic programs, while CellChat and NicheNet were used to characterize cell-cel

Dauricine induces programmed cell death protein 1 lysosomal degradation in T cells to enhance cytotoxicity against melanoma.

Programmed cell death protein 1 (PD-1) is an inhibitory receptor expressed on the surface of activated T cells and suppresses immune responses upon binding to programmed cell death ligand 1 (PD-L1). Blockade of the PD-1/PD-L1 axis has emerged as a groundbreaking immunotherapeutic strategy for the clinical treatment of various cancers, including melanoma. Dauricine is a natural alkaloid with tumor-suppressing properties, although its underlying mechanisms remain incompletely understood. In this study, biochemical and immunofluorescence analyses demonstrated that dauricine effectively reduced PD

Targeting the Tumor Microenvironment Network to Overcome Immunotherapy Resistance in Non-Small Cell Lung Cancer: Mechanisms to Clinical Translation.

BACKGROUND: Immunotherapy has transformed the treatment landscape for advanced non-small cell lung cancer (NSCLC), yet resistance mediated by the tumor microenvironment (TME) limits its efficacy in most patients. This review aims to synthesize the current understanding of the TME as an integrated network driving immunotherapy resistance and to evaluate emerging therapeutic strategies aimed at overcoming it. METHODS: We conducted a comprehensive narrative review of key literature in the field, sourced from PubMed and major conference proceedings. The focus was on mechanistic studies elucidatin

Trogocytosis: Emerging insights into cellular communication, immunity, and pathogenesis.

Trogocytosis is a unique intercellular process that can occur when two cells become in contact to each other and one cell "nibbles" portions of another cell's membrane and surface molecules. This phenomenon is widely observed among immune cells such as B cells, T cells, NK cells, and antigen-presenting cells (APCs). Through trogocytosis, molecules including major histocompatibility complexes (MHC), costimulatory receptors, and antigens are transferred, influencing cellular communication and immune regulation. While trogocytosis supports processes like antigen presentation, immune activation, a

Cardiovascular Functional Cell Injury and Cardiovascular Diseases: Mechanistic Insights and Exercise Interventions.

Cardiovascular diseases (CVDs) are a leading cause of mortality worldwide and substantially impair physical health, psychological well-being, and quality of life. Cardiovascular functional cells, including cardiomyocytes, cardiac fibroblasts (CFs), vascular smooth muscle cells (VSMCs), vascular endothelial cells (ECs), and immune cells, play essential roles in maintaining cardiovascular homeostasis. Accumulating evidence demonstrates that injury to cardiovascular functional cells drives the initiation and progression of CVDs. Specifically, cardiomyocyte injury is associated with mitochondrial

Elucidating the Gene Expression Signature of Rosai-Dorfman Disease: A Study from the Atlas of Blood Cancer Genomes Initiative.

Rosai-Dorfman disease (RDD) is a rare histiocytic disorder characterized by accumulation of large histiocytes in nodal and/or extranodal sites. The underlying biology of RDD and factors that distinguish it from the other histiocyte disorders are poorly understood. To better understand the disease biology, we employed whole transcriptome RNA sequencing to study the gene expression profiles of RDD as part of the Atlas of Blood Cancer Genomes (ABCG) initiative. Gene expression profiles of RDD (N=23) were then compared to two other histiocytoses (Langerhans cell histiocytosis (LCH, N=15) and Erdhe

B cells regulate T cell transfer colitis by partially reconstituting luminal IgA and modulating the colonic microbiota.

Loss of immune tolerance to commensal microbial antigens is a central driver of inflammatory bowel disease. While regulatory T cells are well recognized in maintaining intestinal homeostasis, the contribution of B cells-particularly IgA-secreting cells-remains incompletely defined. In the T cell transfer colitis model, cotransfer of splenic B cells attenuates disease, an effect largely attributed to IL-10-producing regulatory B cells (Bregs). However, alternative B cell-dependent mechanisms have not been fully explored. Here, we investigated whether splenic B cells regulate T cell transfer col

Construction of a soluble mediator landscape and analysis of its clinical diagnostic potential in idiopathic pulmonary fibrosis.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal interstitial lung disease. Current diagnosis primarily relies on imaging and histology, lacking sensitive and specific early biomarkers. Although soluble mediators play pivotal roles in IPF pathogenesis, their systematic characterization and clinical utility remain unclear. This study aimed to construct a soluble mediator-based marker atlas for IPF and evaluate its diagnostic potential. METHODS: Multiple IPF-related transcriptomic datasets (GSE10667, GSE24206, GSE53845) were integrated from the Gene Expression Omnibus

Toward the next generation of CAR T cell therapies for cancer.

With seven products approved by the US Food and Drug Administration (FDA) for 15 indications and more than 2,600 active clinical trials, chimeric antigen receptor (CAR) T cell therapy is firmly established as an effective therapeutic modality for cancer treatment, and promising early data demonstrate its potential in autoimmune diseases. Key challenges facing the field include extending efficacy to solid tumors and reducing the cost and complexity of manufacturing to improve patient access. In this review, we summarize the clinical lessons learned from the now substantial experience with comme

Skin-resident CXCR6+ memory CD4+ T cells and dendritic cells form dermal perivascular hubs that drive relapse in allergic dermatitis.

CD4+ tissue-resident memory T (TRM) cells are crucial for relapse in allergic skin conditions. However, the mechanisms underlying their maintenance and pathogenic potential remain poorly defined. Here, across multiple models of allergic dermatitis, we identified a C-X-C chemokine receptor 6 (CXCR6+) subpopulation within CD4+ TRM cells as a distinct pathogenic subset that rapidly induces relapse. This subset is supported by C-X-C chemokine ligand 16 (CXCL16)-dependent interactions with CD301b+ conventional type 2 dendritic cells (cDC2s) for in situ maintenance. Functional blockade of the CXCL16

Dynamics and functional heterogeneity of FOXP3+ regulatory T cells in Trypanosoma cruzi infection: a systematic review.

BACKGROUND: The role of regulatory T cells (Tregs) in Trypanosoma cruzi infection remains unresolved, with conflicting evidence suggesting either protection from immunopathology or impairment of parasite control. METHODS: We performed the first systematic review integrating data on Treg phenotype and function in human and experimental Chagas disease. A comprehensive search of PubMed, Embase, Scopus, and LILACS (2003-2025) identified studies assessing Tregs during T. cruzi infection. Study selection, data extraction, and risk-of-bias assessment followed PRISMA 2020 and Cochrane guidelines. RE

UBE2C-TRIM21-driven NOS2 degradation stabilizes Tregs and suppresses pyroptotic signaling in thyrocytes in Hashimoto's thyroiditis.

BACKGROUND: Hashimoto's thyroiditis (HT) features persistent immune intolerance and progressive thyrocyte injury, yet the mechanistic linkage between regulatory T cell (Treg) instability and inflammasome-driven inflammatory damage in thyroid follicular cells remains unclear. We hypothesized that a Treg-intrinsic proteostasis module restrains inflammatory effectors to preserve immune tolerance and limit thyrocyte pyroptotic signaling. METHODS: We integrated peripheral-blood Treg transcriptomics from HT patients and healthy controls with functional perturbation, proteomic interaction screening,

Predictors of immunological non-response despite virological suppression in people living with HIV on antiretroviral therapy: A systematic review and meta-analysis.

Immunological Nonresponse (INR) in people living with HIV (PLWH) is characterized by inadequate CD4+ T-cell recovery despite effective viral suppression through antiretroviral therapy (ART). This systematic review and meta-analysis identify key clinical, demographic, and molecular predictors of INR to optimize immune recovery strategies. A comprehensive literature search was conducted in accordance with PRISMA 2020 guidelines, yielding 59 eligible studies, with a total of 19,511 individuals with INR. Study quality was assessed using the Newcastle-Ottawa Scale (NOS), and pooled effect estimates

Human MAITregs possess diverse TCR repertoires and are functionally supported by glycolysis.

Mucosal-associated invariant T (MAIT) regulatory cells (MAITregs) represent a specialized subset with immunosuppressive functions, yet their properties and molecular basis are largely unknown. We demonstrate that MAITregs, while sharing T cell receptor (TCR) repertoires with conventional MAIT cells, undergo selective clonal expansion during in vitro generation, leading to biased V(D)J profiles and restricted CDR3 diversity. Moreover, integrated transcriptomics revealed that MAITregs preferred glycolysis, which was supported by chromatin remodeling at glycolytic gene loci. Functionally, glycoly
