OBJECTIVE: Autoimmune thyroiditis (AIT) has been associated with an increased cardiovascular risk. Whether adipokines, inflammation and auto-immunity contribute to this increased risk is uncertain. Our aim was to evaluate the interrelationships of adipokines, inflammation, auto-immunity, and thyroid function with cardiovascular risk in autoimmune thyroiditis.
METHODS: Sixty-two patients with AIT, with normal free T4 (FT4) and free T3 (FT3), without other comorbidities were studied. Patients were divided into two groups: AIT with thyroid-stimulating hormone (TSH) ≤2.5 mU/L and AIT with TSH >2.

Elevated thyroid hormone concentrations with nonsuppressed thyroid-stimulating hormone (TSH) present a challenging diagnostic problem. Resistance to thyroid hormone beta (RTHβ), analytical interference, and a TSH-secreting pituitary adenoma (TSHoma) are important considerations. We describe a woman with persistent elevation of free thyroxine (FT4) and nonsuppressed or elevated TSH for more than five years, autoimmune thyroiditis, a heterozygous THRB c.284G>C (p.Gly95Ala) variant classified as a variant of uncertain significance (VUS), and a 4.9 × 4.4 mm pituitary microadenoma. Van Wyk-Grumbach syndrome (VWGS) is a rare complication of longstanding, untreated primary hypothyroidism in children. It is characterized by isosexual precocious puberty, delayed bone age, and multicystic ovarian enlargement despite the presence of severe hypothyroidism. Although thyroid enlargement is common, a discrete thyroid nodule has not previously been documented in this syndrome. This study presents a 6-year-old Saudi girl who exhibited a 2-day history of vaginal hemorrhage and a history of breast development for 6 months. The physical examination demonstrated a palpable right thyroid

BACKGROUND: The role of alemtuzumab (ALEM) after natalizumab (NTZ) discontinuation in relapsing-remitting multiple sclerosis (RRMS) remains to be better defined in real-world settings.
METHODS: We conducted a retrospective, longitudinal, single-centre cohort study including RRMS patients treated with ALEM. Clinical and MRI evaluations were performed every 6 months. Annualized relapse rate (ARR), and radiological activity were assessed longitudinally. NEDA-3 was evaluated both as an annual interval-specific outcome and as sustained NEDA-3 using Kaplan-Meier analysis.
RESULTS: Eighty-one patie

BACKGROUND: Hashimoto's thyroiditis (HT) features persistent immune intolerance and progressive thyrocyte injury, yet the mechanistic linkage between regulatory T cell (Treg) instability and inflammasome-driven inflammatory damage in thyroid follicular cells remains unclear. We hypothesized that a Treg-intrinsic proteostasis module restrains inflammatory effectors to preserve immune tolerance and limit thyrocyte pyroptotic signaling.
METHODS: We integrated peripheral-blood Treg transcriptomics from HT patients and healthy controls with functional perturbation, proteomic interaction screening,

Sweet syndrome, also known as acute febrile neutrophilic dermatosis, is a rare dermatological condition characterized by the abrupt onset of tender, erythematous skin lesions, often presenting with fever and systemic inflammation. It is commonly associated with infection, malignancy, and medication exposure. Isolated case reports have described the co-occurrence of Sweet syndrome and thyroid disease, but these reports have not previously been collected or appraised together. A case-based narrative review was conducted using published case reports and case series describing patients with both c

Thyroid diseases have become the most common endocrine disorders globally, including hyperthyroidism, hypothyroidism, thyroid nodules (TN), and thyroid cancer (TC). Traditionally, the pathogenesis was attributed to autoimmune dysfunction, iodine metabolism imbalance, and genetic-environmental interactions. Recently, research suggests that vitamin D deficiency and gut microbiota dysbiosis are prevalent in patients with thyroid disorders, particularly in autoimmune thyroid disease (AITD). Vitamin D, as well as gut microbiota and its metabolites, such as short-chain fatty acids and bile acids, jo

OBJECTIVE: Postoperative hypoparathyroidism (hypoPT) remains a significant complication after total thyroidectomy. Elevated thyroid autoantibodies are commonly considered surrogate markers of autoimmune thyroiditis and are presumed to increase surgical difficulty, potentially leading to parathyroid injury; however, their independent impact on postoperative parathyroid outcomes remain uncertain. This study aimed to evaluate the association between elevated preoperative thyroid antibody levels and postoperative parathyroid dysfunction in patients undergoing total thyroidectomy.
METHODS: This re

OBJECTIVE: Autoimmune thyroiditis (AIT) is one of the most common autoimmune diseases and often causes hypothyroidism in patients. As a traditional formulation in my country, Buzhong Yiqi decoction (BZYQD) has significant effects in improving clinical symptoms of AIT and reducing autoantibody titers, but its specific mechanism of action needs to be further explored. The purpose of this study was to explore the effective targets and related mechanisms of Buzhong Yiqi decoction in AIT mice based on transcriptome sequencing technology.

BACKGROUND: Autoimmune thyroiditis, such as Graves' disease (GD) and Hashimoto's thyroiditis (HT), is a complex, genetically linked condition. IL-8 and TNF-α play a crucial role in these pathologies.
METHODS: This research was conducted on Tunisian patients with autoimmune thyroiditis (AITD) to examine the link between genetic variations in the IL-8 and TNF-α genes, their protein expression, and the likelihood of developing GD or HT. A total of 319 healthy controls, 230 patients with HT, and 75 patients with GD underwent genotyping via PCR-RFLP for the TNF-α-238 G/A and IL-8+781C>T polymorphi

Background Lichen sclerosus (LS) is a chronic inflammatory dermatosis characterised by ivory-white patches to plaques. Evidence suggests an autoimmune aetiology for LS, given its association with various autoimmune conditions, such as autoimmune thyroiditis, alopecia areata (AA), and vitiligo. However, the association between LS and thyroid diseases remained controversial. Aim We aimed to investigate the association between LS and thyroid diseases. Methods We performed a systematic review of studies on the association between LS and thyroid diseases. We searched CENTRAL, MEDLINE, and Embase fr

INTRODUCTION: Autoimmune thyroiditis (AIT), a prevalent autoimmune disorder, can severely impair patients' quality of life when progressing to hypothyroidism. This investigation seeks to systematically evaluate the therapeutic efficacy of Buzhong Yiqi Decoction (BZYQ) against iodine-induced AIT while elucidating its underlying molecular mechanisms.
METHODS: NOD.H-2h4 mice received daily 0.05% sodium iodide gavage for 8 weeks to induce AIT. Thyroid histopathology was evaluated by HE staining. Serum autoantibody levels and lactate concentrations were measured. Western blotting analyzed PTEN, p-

Although well-differentiated thyroid carcinoma (WDTC) is characterized by a robust treatment response, aggressive subtypes, such as anaplastic thyroid carcinoma (ATC), remain highly lethal. To understand thyroid cancer evolution in both children and adults, we analyzed single-cell transcriptomes of 423,733 cells from 81 samples and spatially resolved key tumor and microenvironment populations across 28 tumors with spatial transcriptomics, including rare and unique composite WDTC/ATC tumors and pediatric diffuse sclerosing thyroid carcinomas. Additionally, we identified gene signatures of strom

INTRODUCTION: Type 1 diabetes mellitus (T1DM) is an autoimmune disease that damages insulin-producing pancreatic cells, often appearing in childhood. Global incidence is rising at 2%-3% yearly. Its exact cause is unclear. Prenatal exposures and maternal autoimmune disorders have been reported as potential risk factors. This study aimed to explore how maternal autoimmune conditions might correlate with the onset of T1DM, employing a population-focused approach.
METHODS: This is a retrospective population-based cohort study with a nested case-control analysis. Primary data were derived from the
