Pharmacological blockade of CRHR1 prevents early-onset puberty and enhanced anxiety-like behavior induced by early-life adversity in female Sprague Dawley rats.

Pubertal development is regulated by hypothalamic neuroendocrine signaling and is sensitive to interactions between stress- and reproductive-related pathways. In particular, corticotropin-releasing hormone (CRH) signaling via corticotropin-releasing hormone receptor 1 (CRHR1) represents a key point of crosstalk between stress and pubertal maturation. Early-life adversity, such as the maternal separation (MS) model in rodents, has been shown to increase CRH expression and therefore has the potential to dysregulate CRHR1 signaling. We previously reported that MS-exposed female rats display early

Diagnosis and treatment analysis of a case of idiopathic hypogonadotropic hypogonadism caused by NSMF gene mutation.

Idiopathic hypogonadotropic hypogonadism (IHH) is a rare endocrine disorder characterized by deficiency or dysfunction of endogenous gonadotropin-releasing hormone (GnRH), leading primarily to hypogonadism. Based on the presence or absence of olfactory dysfunction, IHH is classified into Kallmann syndrome (KS) and normosmic IHH (nIHH). In this study, we reported a 15-year-old male with nIHH, presenting with delayed puberty, hypogonadotropic hypogonadism and normal olfaction. High-throughput gene sequencing identified a novel NSMF:c.79C>G (p.Arg27Gly) variant which has not been reported in the

Central precocious puberty in partial androgen insensitivity syndrome: a 9-year follow-up.

Androgen insensitivity syndrome (AIS) is a group of complex clinical symptoms caused by X-linked genetic defects. It results from inactivating mutations in the androgen receptor (AR) gene, leading to hormone resistance in individuals with an XY karyotype despite normal androgen synthesis. We present a case of a 2-year-1-month-old boy who initially showed signs of incomplete masculinization of the external genitalia and underwent transverse preputial island flap urethroplasty during childhood. At the age of 7, he presented with gynecomastia, accompanied by accelerated growth and advanced bone a

Bone densitometry findings and prevalence of osteoporosis at initial scanning in girls with anorexia nervosa.

BACKGROUND AND AIMS: Low bone mineral density (BMD) in children and young people (CYP) with anorexia nervosa (AN) is well recognized, yet the prevalence of osteoporosis remains unknown. This study investigates bone densitometry findings using dual energy X-ray absorptiometry (DXA) scan and peripheral quantitative computed tomography (pQCT) in girls with AN at initial referral, comparing outcomes by menstrual status and assessing osteoporosis prevalence. METHODS: A retrospective review included 264 girls with AN, aged 10.2-19.8 years, referred for initial DXA and pQCT scans at a tertiary pedia

A de novo SOX11 mutation causing hypogonadotropic hypogonadism: a case report and literature review.

BACKGROUND: SOX11 variants have been associated with Coffin-Siris syndrome and a broader neurodevelopmental spectrum (IDDMOH), but their role in isolated or syndromic hypogonadotropic hypogonadism (HH) remains under-recognized. CASE PRESENTATION: We report a 13-year-old Chinese girl who presented with delayed puberty (Tanner B1, PH1), infantile uterus and low basal gonadotropins. Trio whole-exome sequencing identified a de-novo heterozygous SOX11 c.346_348del (p.Tyr116del) variant. Olfactory and pituitary MRI revealed bilateral olfactory-bulb/nerve hypoplasia and a pituitary height of 3.4 mm,

Childhood-onset systemic lupus erythematosus presenting with growth retardation and delayed puberty: a case report.

Childhood-onset systemic lupus erythematosus (cSLE) is a chronic, multisystem autoimmune disease characterized by marked clinical heterogeneity and substantial morbidity. Although rash, arthritis, and renal involvement are common manifestations, atypical presentations may delay diagnosis. In rare cases, growth retardation and delayed puberty may precede typical systemic features, posing a diagnostic challenge. We report the case of a 12-year-old Chinese girl who presented to the endocrinology clinic with progressive growth retardation and delayed pubertal development. Comprehensive growth asse

Intergenerational disruptions to reproductive, physical, and neurobehavioral development in rats after prenatal exposure to the emergent pollutant betamethasone.

Betamethasone (BM) is the standard clinical treatment for accelerating fetal lung maturation in women at risk of preterm delivery. However, emerging evidence identifies BM as posing a significant "One Health" challenge. While antenatal corticosteroid therapy provides immediate neonatal benefits, early overexposure to glucocorticoids is linked to fetal programming and the Developmental Origins of Health and Disease (DOHaD), with potential consequences spanning multiple generations. The aim of this work was to evaluate the impacts of prenatal BM exposure on physical, neurobehavioral, and reprodu

An Unexpected Result in a Case of Gonadal Dysgenesis: Noonan Syndrome Caused by RIT1 Mutation.

Noonan syndrome occurs in approximately 1/1,000-1/2,500 live births and is caused by defects in the Ras/mitogen-activated protein kinase pathway. Pubertal development includes syndrome-specific differences which may manifest as delayed puberty in both sexes, as well as cryptorchidism and impaired gonadal function, especially in the males. However gonadal dysgenesis and disorders of sex development have not been previously reported in the literature before. Our patient presented with ambiguous genitalia at two days of age. There was no consanguinity between the parents. On physical examination,

Testosterone therapy in boys with constitutional delay of growth and puberty: a PubMed-based systematic review and exploratory meta-analysis.

Constitutional delay of growth and puberty (CDGP), the most common cause of delayed puberty in boys, accounts for the majority of pediatric endocrinology referrals. Although traditionally viewed as a variant of normal development, affected adolescents frequently experience short stature relative to their peers, delayed secondary sexual characteristics, and clinically meaningful psychosocial distress. Short-course low-dose testosterone has been used for more than 5 decades to accelerate pubertal progression and linear growth; however, the evidence base is heterogeneous and has not been synthesi

Pubertal development and hypothalamic-pituitary-gonadal axis are altered in male mice lacking Mecp2.

Mutations in the MECP2 gene, encoding the epigenetic reader Methyl-CpG binding protein 2, are the main cause of Rett syndrome, a rare neurodevelopmental disorder. Besides severe symptoms such as profound intellectual disability, loss of speech and motor skills, and epilepsy, loss of function of MECP2 has been associated with pubertal dysregulation, but the biological mechanisms leading to this remain unclear. Using a mouse model of Rett, in which males are hemizygous and females heterozygous for Mecp2 loss of function mutation, we assessed the onset and progression of puberty, together with in

Identification and management of unrecognized femoral head epiphysiolysis: a case report.

BACKGROUND: Slipped capital femoral epiphysis (SCFE) is the slippage of the proximal femoral epiphysis on the femoral neck through the epiphyseal plate. This condition is generally observed in male, obese patients during pubertal growth spurts, and it can sometimes be associated with endocrine disorders. Despite its significant incidence in the paediatric population, the diagnosis is frequently delayed which leads to significant worsening of the prognosis. We present a case of SCFE in which the diagnosis was delayed due to a series of confounding factors. CASE PRESENTATION: We present a case

MCM8 variants in two patients with primary ovarian insufficiency: clinical findings and in vitro defective DNA repair revealed by an MCM8 double-mutant construct.

BACKGROUNDS: Pediatric-onset primary ovarian insufficiency (POI) presents distinct clinical challenges, including delayed puberty or growth retardation. Previously, we reported a Chinese family with POI harboring compound heterozygous variants (p.C242R and p.S445*) in MCM8 gene, a critical factor for DNA repair and gonadal homeostasis. OBJECTIVE: This study describes clinical findings of pediatric-onset POI patients with compound heterozygous MCM8 variants and investigates the in vitro functional consequences of an artificial double-mutant MCM8 construct harboring both p.C242R and p.S445*. Fifteen-minute consultation: Delayed puberty in girls.

Delayed puberty in girls most commonly presents as absent breast development by age 13 years and occurs less frequently than delayed puberty in boys. Constitutional delay of growth and puberty (CDGP) is the most common individual diagnosis but pathological causes collectively account for a substantial proportion of cases. For the general paediatrician, a practical assessment framework should include focused history, auxology, Tanner staging, first-line investigations, recognition of red flags and timely referral to paediatric endocrinology. Girls with absent thelarche by age 13 years, absent m

Comparison Between Chronological and Bone Age at Menarche in Girls with Laron Syndrome.

Background: In the literature, the occurrence of menarche, the index of female sexual maturation, is related to chronological age (CA), but in conditions of abnormal growth it was proposed that determination of bone age (BA) was a better index of pubertal maturation than CA. Aim: To compare CA with BA at menarche in girls with Laron Syndrome (LS), a model of genetic IGF-I deficiency, which causes severe dwarfism and delayed puberty. Subjects: Data were retrieved retrospectively from our medical records. Nine untreated and four IGF-I-treated girls with LS were included in the study. Eleven GH-t
