IGF-I bioavailability in congenital isolated growth hormone deficiency.

BACKGROUND: The Itabaianinha cohort in Brazil carries a homozygous growth hormone-releasing hormone (GHRH) receptor (GHRH-R) gene mutation, causing congenital isolated GH deficiency (GHD). Affected individuals present with severe short stature, central obesity, hypercholesterolemia, and marked reductions in serum GH, IGF-I, and IGFBP 3 concentrations yet show no premature atherosclerosis and maintain a normal lifespan. IGF-I mostly circulates bound to IGFBPs and requires proteolytic cleavage for IGF-I receptor activation. Pregnancy-associated plasma protein A (PAPP-A) is an important IGF-depen

Structural instability impairs function of the UDP-xylose synthase 1 Ile181Asn variant associated with short-stature genetic syndrome in humans.

Glycosaminoglycan assembly on proteoglycans involves a common tetrasaccharide linker that starts with xylose attached to a serine on the protein. Defective linker biosynthesis caused by a missense mutation of human UDP-xylose synthase (hUXS1) is associated with connective tissue disorders characterized by skeletal abnormality and short stature. The Ile181Asn variant of hUXS1 was reported as inactive in releasing UDP-xylose from UDP-glucuronic acid. Here, we show that Ile181Asn-hUXS1 exhibited catalytic properties similar to the wild-type enzyme but featured a significant decrease in stability,

Near Final Height in Males treated with Aromatase Inhibitors.

OBJECTIVE: Data on the impact of aromatase inhibitor (AI) therapy on final or near-final adult height (FNFH) in males with short stature is limited. This study investigated whether AI therapy improved FNFH in males with advanced or rapidly advancing bone age (ABA) and compromised predicted adult height. METHODS: Data were collected through retrospective chart review. Descriptive statistics were used to characterize the study cohort. Fisher’s exact test and the Wilcoxon rank-sum test were used to compare outcomes. RESULTS: Of 72 patients reviewed, 59 (82%) received anastrozole, 11 (15%) recei

International guideline on genetic testing of children with short stature.

Short stature may be caused by a multitude of conditions, including genetic and non-genetic causes. Over the last decade, advances in genetic sequencing technologies have revolutionized our understanding of the underlying physiology of growth and greatly increased our ability to identify genetic etiologies of short stature. The current guideline provides a general overview of the approach to the evaluation of a child with short stature, followed by recommendations identifying factors in the medical and family history, physical examination, radiographic, and laboratory work up which increase th

Activation of OsOxO2 by T-DNA Insertion Affects Plant Height and Leaf Angle in Rice.

Inclination of leaf and plant height are important agronomic traits that are closely related to grain yields. Proteins involved in regulating free auxin (IAA) levels have a central role in modulating rice leaf angle and plant height. In this study, a rice mutant (named lc4) was found to have enlarged leaf angles and exhibit dwarfism compared to wild-type (WT) plants. Flanking analysis revealed that the mutation was caused by a T-DNA insertion at 270 bp upstream of the oxalate oxidase 2 (OsOxO2) ATG start codon. The insertion significantly increased the transcript levels of indole-3-acetic acid

Ectopic, but not endogenous, expression of the transcription factor gene MpSETA disrupts epidermal tissue formation in Marchantia polymorpha.

Basic helix-loop-helix (bHLH) transcription factors form dimers that regulate the expression of their downstream genes and drive morphogenesis in multicellular eukaryotes. We recently revealed that a bHLH heterodimer consisting of MpSETA (also known as MpSMF) and MpICE2 (also known as MpSCRM2) coordinately controls the development of the seta, a diploid sporophytic tissue, in the liverwort Marchantia polymorpha. Ectopic expression of MpSETA in the haploid thallus causes abnormalities in epidermal tissue formation, but whether native MpSETA has any role in haploid tissue formation was unclear.

The Iberian fossil record of †Otodus megalodon rejects Mediterranean dwarfism and supports nursery use.

†Otodus megalodon, the largest known macropredatory shark, was globally distributed from the Miocene to Pliocene, yet most ecological and palaeobiological inferences rely heavily on American collections. This geographical bias limits understanding of its population structure and life-history strategies elsewhere. Two explanations have been proposed for its body-size patterns: a Bergmann-type cline, with larger individuals at higher, cooler latitudes and a related hypothesis of Mediterranean dwarfism; and the influence of localized nursery areas that generate assemblages dominated by small juve

Case Report: Dual pathogenic mechanism of a PRKG2 missense variant underlies an attenuated phenotype of acromesomelic dysplasia.

Acromesomelic dysplasia comprises a group of rare skeletal disorders characterized by dwarfism with anomalies predominantly affecting the middle and distal segments of the limbs. Based on genetic variants, five types are recognized, with significant phenotypic variability even within a single type. Here, we describe a girl presenting with borderline short stature and mild disproportion due to acro- and mesomelic shortening of the limbs. Radiographic examination revealed shortening of the radius and ulna, mild brachydactyly, absence of iliac flaring and metaphyseal alterations of the long bones

Submicroscopic 16q24.2-q24.3 deletion in a family with nonsyndromic short stature.

Array-based comparative genomic hybridization for a boy, his mother and his half-sister with etiology-unknown nonsyndromic short stature identified a hitherto unreported heterozygous ~1.5-Mb deletion at 16q24.2-q24.3. Whole-exome sequencing detected no pathogenic variants. Our results, in conjunction with previous reports of cases with similar deletions, indicate that the 16q24.2-q24.3 region provides a platform for submicroscopic deletions and possibly contains a gene(s) or regulatory elements involved in skeletal growth.

Predicting congenital hypothyroidism in newborns with complex risk profiles by using thyroid-stimulating hormone variations across serial dried blood spots.

OBJECTIVES: Newborn screening for congenital hypothyroidism (CH) relies on thyroid-stimulating hormone (TSH) levels from dried blood spots (DBS). We investigated whether incorporating TSH variation across serial DBS could improve prediction of CH in term and preterm infants with complex risk profiles. METHODS: Among 207,895 newborns screened, 272 (0.13 %) were diagnosed with CH. TSH variations across 3 serial DBS were analyzed in 6,146 healthy infants (2.96 %). Predictive algorithms were developed using linear mixed-effects models in 1,968 term and 1,387 preterm infants with ≥2 and ≥3 DBS, re

BSK family kinases are essential for brassinosteroid signaling and suppression of adventitious rooting by repressing the expression of LBD16.

While BRASSINOSTEROID (BR) SIGNALING KINASEs (BSKs) are known to interact with the BR receptor BR-INSENSITIVE 1 (BRI1), the extent of their genetic redundancy and essential contribution to BR signaling remain unclear. This study aimed to systematically investigate the function of BSK family members and uncover novel developmental outputs controlled by BR signaling via BSKs mediated pathway in Arabidopsis. We generated an undecuple bsk knockout/knockdown mutant (bsk-u) in Arabidopsis. Comprehensive phenotypic observation, molecular analyses, and genetic complementation were performed to charact

Immunometabolic Reprogramming in Hepatocellular Carcinoma Mechanisms, Biomarkers, and Therapeutic Implications.

Hepatocellular carcinoma (HCC) develops in a chronically inflamed and dysregulated liver metabolism, in which tumor progression and resistance to treatment are orchestrated by the changes in cellular metabolism and immune control. Growing evidence recognizes immunometabolic reprogramming as the two-way interaction of metabolic processes and immune cell capabilities as one of the major determinants of immune evasion and heterogeneity of treatment response in HCC. The review aims to comprehensively evaluate immunometabolic reprogramming in hepatocellular carcinoma, with a focus on its role in tu

The challenges of optimising the treatment of children diagnosed with idiopathic short stature.

Physicians who treat children with idiopathic short stature (ISS) are challenged in deciding whether the intervention to improve final height is truly efficacious, given the heterogeneity of the diagnosis of ISS. There are numerous studies and reviews that have attempted to assess and demonstrate the benefit of various interventions, such as growth hormone therapy and aromatase inhibitors. Recently, Zhang et al. published a well-written meta-analysis entitled "Comparative effects of growth hormone, testosterone, and aromatase inhibitors on height gain in children and adolescents with idiopathi
