Melanocortin 5 receptor signaling protects against podocyte injury in proteinuric glomerulopathies.

Melanocortin therapeutics, exemplified by adrenocorticotropic hormone, have a proven steroidogenic-independent anti-proteinuric and glomerular protective effect. The biological functions of melanocortins are mediated by melanocortin receptors (MCR), including MC1R, which recent studies have shown to protect against glomerular disease. However, the role of other MCRs like MC5R is unknown. Here, Mc5r knockout exacerbated glomerulopathy in mice injured by adriamycin (ADR) or nephrotoxic serum (NTS), as demonstrated by increased albuminuria and podocyte injury. Conversely, selective MC5R agonism u

TRα1 mutant suppresses KLF9 to cause endometrial metaplasia with ectopic IL-33 expression leading to uterine fibrosis and infertility.

Thyroid hormone receptors (TRs) mediate the genomic actions of thyroid hormone. Mutations of THRA gene cause a human disease known as resistance to thyroid hormone (RTHα). We created a mouse model expressing a dominant negative mutated TRα1 (Thra1PV/+ mice) that exhibits growth retardation, bone abnormalities, constipation, and anemia, as found in RTHα patients. In addition, female Thra1PV/+ mice exhibit decreased fertility. In the present study, we aimed to characterize the molecular events leading to infertility. Histologically, there was progressive uterine atrophy in Thra1PV/+ mutant mice,

Mechanisms of GLP-1 Receptor Agonist-Induced Weight Loss: A Review of Central and Peripheral Pathways in Appetite and Energy Regulation.

Glucagon-like peptide-1 (GLP-1) receptor agonists (RAs) have become central in managing obesity and type 2 diabetes, primarily through appetite suppression and metabolic regulation. This review explores the mechanisms underlying GLP-1 RA-induced weight loss, focusing on central and peripheral pathways. Centrally, GLP-1 RAs modulate brain regions controlling appetite, influencing neurotransmitter and peptide release to regulate hunger and energy expenditure. Peripherally, GLP-1 RAs improve glycemic control by enhancing insulin secretion, reducing glucagon release, delaying gastric emptying, and

Hypoparathyroidism: diagnosis, management and emerging therapies.

Hypoparathyroidism is characterized by inadequate parathyroid hormone (PTH) secretion or action and results in hypocalcaemia, and can lead to hyperphosphataemia and hypercalciuria. Most cases of hypoparathyroidism occur as a complication of surgery, with the remainder due to causes including autoimmune disease, genetic causes, infiltrative diseases, mineral deposition or due to abnormalities in serum levels of magnesium. Hypoparathyroidism can cause multisystem disease, with long-term complications resulting from ectopic calcification as well as renal complications with nephrocalcinosis, nephr

The MADS-box transcription factor CmFYF promotes the production of male flowers and inhibits the fruit development in melon (Cucumis melo L.).

The FOREVER YOUNG FLOWER (FYF/AGL42) gene plays an important role in regulating the flower development especially the flowering time, and senescence and abscission of floral organs. Melon is an important horticultural crop, and the flower development has an important influence on pollination, fertilization and final fruit yield. However, the function of FYF homolog is still unknown in melon. In this study, the characteristic of melon CmFYF was analyzed combining with molecular biology, developmental biology and biochemical tools. CmFYF was present in all detected tissues of melon, but its expr

Pheochromocytoma associated with ectopic ACTH-producing tumor and hyper-interleukin-6emia: a case report with review of literature.

Pheochromocytomas occur in the adrenal medulla and present with various symptoms associated with excessive catecholamine production. Although pheochromocytomas associated with ectopic ACTH-producing tumors and hyper-interleukin-6emia (hyper-IL-6emia) have been reported, those associated with both diseases simultaneously have not been reported. In pheochromocytomas with ectopic ACTH-producing tumors and hyper-IL-6emia, the disease characteristics and relationship between each hormone and cytokine are unknown. Herein, we report a case of a 56-year-old woman with stroke whose computed tomography

A Pheochromocytoma With Adrenocorticotropic Hormone Secretion and Subsequent Fatal Outcome: A Case Report.

Hypercortisolemia and hypercatecholaminemia in ectopic adrenocorticotropic hormone (ACTH)-secreting pheochromocytoma represent life-threatening conditions, particularly when diagnosis is delayed. In this case, a 66-year-old male patient with poorly controlled diabetes presented with severe constipation in the absence of the classic Cushingoid phenotype. Computed tomography revealed bilateral adrenal masses, and 123I-metaiodobenzylguanidine scintigraphy revealed radiotracer uptake in the left adrenal gland. Endocrine assessments confirmed elevated catecholamine, ACTH, and cortisol levels, indic

Excess fibroblast growth factor 23 in alcoholic osteomalacia is derived from the bone.

Excess fibroblast growth factor 23 (FGF23), a mature osteocyte-derived phosphaturic hormone, causes chronic hypophosphatemic osteomalacia in adults. This rare condition was recently reported in 2 alcoholic patients, with marked improvement upon cessation of alcohol consumption, suggesting a link between alcohol and FGF23-related hypophosphatemia within the highly limited cases. This study aimed to investigate whether the source of excess FGF23 in alcohol-induced FGF23-related hypophosphatemic osteomalacia is the bone or the other organs. To achieve this goal, an immunohistochemical approach fo

Single-cell transcriptomic atlas of different endometriosis indicating that an interaction between endometriosis-associated mesothelial cells (EAMCs) and ectopic stromal cells may influence progesterone resistance.

BACKGROUND: Endometriosis is a hormone-dependent disease, which can usually be divided into peritoneal endometriosis (PEM), deep-infiltrating endometriosis (DIE) and ovarian endometriosis (OEM). Although the three pathologic types are essentially the same disease, they differ in pathological manifestations, molecular features, pain symptoms and hormonal responsiveness. However, there is limited literature focusing on the differences among these types. In this study, we employed single-cell RNA sequencing (scRNA-seq) to profile the transcriptome of each type using surgical biopsy samples obtain

Neoadjuvant Combined Preoperative Embolization of a Highly Vascularized Paraganglioma Associated With SIADH.

BACKGROUND: Device-based embolization is a well-established medical procedure for treating several pathological conditions, including cerebral and peripheral pseudoaneurysms, congenital defects, and active bleeding. Nonetheless, an intriguing and relatively underexplored application of coils and microparticles involves the preoperative reduction of blood supply to ectopic masses. CASE SUMMARY: A 58-year-old woman was admitted to our institution with nausea and confusion, with a suspected diagnosis of syndrome of inappropriate antidiuretic hormone secretion. Systematic review on the DNA methylation role in endometriosis: current evidence and perspectives.

BACKGROUND: Endometriosis appears to have a multilayered etiology, with genetic and epigenetic factors each contributing half of the pathogenesis. The molecular processes that underlie the onset of endometriosis are yet unclear, but it is assumed that an important contributor in the etiopathology of the disease is DNA methylation. METHODS: We conducted a systematic review of the literature regarding DNA methylation in endometriosis following PRISMA guidelines. Records were obtained from PubMed and Web of Science on May 31, 2024. Original research articles analyzing regional or genome-wide DNA

Increased seizure susceptibility in thyroid hormone transporter Mct8/Oatp1c1 knockout mice is associated with altered neurotransmitter systems development.

Thyroid hormone (TH) transporters such as the monocarboxylate transporter Mct8 and the organic anion transporting protein Oatp1c1 facilitate TH transport into target cells. In humans, inactivating mutations in MCT8 result in Allan-Herndon-Dudley syndrome (AHDS), a severe psychomotor retardation with hallmarks of a central TH deficit and frequently observed seizures of unknown etiology. Here, we aimed to investigate seizure susceptibility in AHDS by using Mct8/Oatp1c1 double-knockout (Dko) mice, a well-established AHDS model. We tested seizure susceptibility using the pilocarpine model and obse

Unraveling hypertension through mineralocorticoid receptor activation in Cushing's syndrome: A case report.

BACKGROUND: Cushing's syndrome is a rare cause of secondary hypertension. There are several mechanisms reported as contributing factors for blood pressure elevation in hypercortisolism patients. This case highlights the central role of mineralocorticoid receptors' activation by the excess cortisol in the development of hypertension. CASE PRESENTATION: A 45-year-old male presented with several months of cushingoid features and refractory hypertension on maximum doses of five antihypertensive drugs, not including a mineralocorticoid receptor blocker. Ethylene promotes SMAX1 accumulation to inhibit arbuscular mycorrhiza symbiosis.

Most land plants engage in arbuscular mycorrhiza (AM) symbiosis with Glomeromycotina fungi for better access to mineral nutrients. The plant hormone ethylene suppresses AM development, but a molecular explanation for this phenomenon is lacking. Here we show that ethylene inhibits the expression of many genes required for AM formation in Lotus japonicus. These genes include strigolactone biosynthesis genes, which are needed for fungal activation, and Common Symbiosis genes, which are required for fungal entry into the root. Application of strigolactone analogs and ectopic expression of the Comm

FOXA2 loss results in an increase of endometriosis development and LIF reveals a therapeutic effect for endometriosis.

Endometriosis, characterized by the growth of uterine-like tissue outside the uterus, causes chronic pain and infertility. Current diagnostic and therapeutic strategies have notable limitations, including delayed diagnosis and adverse effects. The transcription factor forkhead box A2 (FOXA2), which is exclusively expressed in the uterine glandular epithelium, regulates key genes involved in endometrial proliferation, differentiation, fertility, and hormone response. While FOXA2 expression is reduced in the endometrial tissue of women with endometriosis, its pathophysiological role in the disea

Altered development and lignin deposition in rice p-COUMAROYL ESTER 3-HYDROXYLASE loss-of-function mutants.

The aromatic composition of lignin greatly influences the potential utility of lignocellulosic biomass. Previously, we generated transgenic rice plants with altered lignin aromatic composition and enhanced biomass utilization properties by suppressing the expression of p-COUMAROYL ESTER 3-HYDROXYLASE (C3'H). While RNAi-derived C3'H-knockdown lines displayed relatively normal growth with substantially augmented levels of p-hydroxyphenyl-type lignin units, genome-edited C3'H-knockout lines exhibited severely impaired growth phenotype, leading to arrested seedling development. In this study, we f

Human placental mesenchymal stem cells ameliorates premature ovarian insufficiency via modulating gut microbiota and suppressing the inflammation in rats.

The core objective of this study was to explore the effects and potential mechanism of human placental mesenchymal stem cells (PMSCs) in improving early-onset ovarian dysfunction (POI). Mesenchymal stem cells with multidirectional differentiation ability were isolated from human placenta tissue and a culture system of human PMSCs was constructed for this study. Subsequently, we successfully constructed POI rat models using cisplatin induction. We randomly divided these models into four groups: CON group (blank control), MOD group (POI model), MED group (hormone therapy), and PMSC group (PMSCs

Control of alveolar bone development, homeostasis, and socket healing by salt-inducible kinases.

Alveolar bone supports and anchors teeth. The parathyroid hormone-related protein (PTHrP) pathway plays a key role in alveolar bone biology. Salt-inducible kinases (SIKs) are important downstream regulators of PTH/PTHrP signaling in the appendicular skeleton, where SIK inhibition increases bone formation and trabecular bone mass. However, the function of these kinases in alveolar bone remains unknown. Here, we report a critical role for SIK2/SIK3 in alveolar bone development, homeostasis, and socket healing after tooth extraction. Inducible SIK2/SIK3 (Ubq-creERt;Sik2f/f;Sik3f/f) deletion led t

The effects of SGLT2 inhibitors on metabolic phenotype and FGF-21 expression from the adipose tissue and the liver are less pronounced in ob/ob mice.

BACKGROUND: the metabolic effects of sodium-glucose cotransporter-2 inhibitors (SGLT2i), such as lipolysis and ectopic fat reduction, seem related to the synthesis of fibroblast growth factor-21 (FGF-21), and FGF-21 analogs are now under investigation for the treatment of obesity complications such as metabolic dysfunction-associated steatotic liver disease. However, FGF-21 levels are paradoxically higher in obesity, indicating a hormone-resistant state that may hinder the benefits of SGLT2i. METHODS: To define if a different energy status influences the response to SGLT2i, we evaluated the e
