BACKGROUND: Aberrant DNA methylation patterns are increasingly recognized as contributors to a wide range of conditions, including metabolic, oncogenic, and neurodevelopmental disorders. Nutritional factors, such as choline, can shape methylation potential via methyl group donation. The purpose of this narrative review is to synthesize current evidence on the DNA methylation landscapes underlying health and disease paradigms, with a focus on the role of choline as a compelling target for modulating epigenetic states. A comprehensive literature review search was conducted in PubMed to identify

Plants do not produce Vitamin B12, creating a nutrient insufficiency risk for those who do not consume animal-derived foods without supplementation. Furthermore, various diseases cause Vitamin B12 deficiency. Here, we establish an approach for B12 dietary supplementation that harnesses a horticultural technology to deliver the recommended daily allowance (RDA) of B12 within a single portion of a salad crop (pea shoots). We demonstrate the effectiveness of the approach in a commercial and scalable growing environment, conducted an economic evaluation, find that it has versatility for growers, d

BACKGROUND Nitrous oxide (N₂O) deactivates vitamin B12 and, when inhaled regularly, can lead to vitamin B12 deficiency, resulting in temporary or permanent peripheral neuropathy. This case report describes a young adult with N₂O-induced B12 deficiency, whose neurological symptoms were initially misattributed to a distracting traumatic injury from a motor vehicle collision (MVC). CASE REPORT We present a case of a previously healthy 24-year-old man who presented with low back pain and bilateral leg paresthesia that began 2 days after being a passenger in an MVC. There was no head trauma, loss o

Folate is an essential nutrient that supports the formation of S-adenosyl methionine (SAM) in the pathway of one carbon metabolism. Dietary folate intake has been shown to affect the SAM-dependent methylation of diverse substrates, including DNA and inorganic arsenic (iAs). The methylation of iAs by arsenic methyltransferase (AS3MT) plays a key role iAs detoxification in both humans and mice. Our recent studies using wild-type C57BL/6N mice showed that preconception exposure to iAs resulted in heritable changes in DNA methylation in paternal sperm and differential expression of genes in tissue

Nutrient deficiency can cause increased susceptibility to infectious diseases in fish, thus leading to high rates of morbidity and mortality. Thiamine deficiency complex (TDC) in fish can lead to low reproductive success and high mortality rates. Columnaris disease in salmonids, caused by Flavobacterium columnare, has resulted in devastating losses in aquaculture production and wild populations of Pacific salmon particularly associated with climate change and high water temperatures. There is growing awareness that both TDC and columnaris are emerging diseases of salmonids on the west coast of

BACKGROUND: Diabetics often develop vitamin B12 deficiencies, which are crucial for blood, nerve, cognitive, and cardiovascular functions. The impact of metformin on vitamin B12 levels, leading to complications such as peripheral neuropathy and anemia, is well-known; yet no studies focus on deficiency status at diabetes diagnosis or the start of treatment.
METHODS: A cross-sectional study was conducted at 2 tertiary care institutions in India, Command Hospital (Western Command), Haryana, and Civil Hospital in Sirsi, Karnataka, from July 2022 to November 2023. The study included 326 newly diag

Folate is an indispensable nutrient involved in key biological processes, including enzymatic reactions, DNA replication, metabolic regulation, and methylation. Studies indicate that folate promotes neuronal regeneration and repair in patients with spinal cord injury (SCI); however, its precise mechanisms remain unclear. We employed network pharmacology to identify potential targets of folate for SCI treatment; we conducted a clinical retrospective study, selecting 50 SCI patients and 50 non-SCI control subjects from Tianjin Hospital between 2022 and 2025 to validate predictions by assessing s

BACKGROUND: Thiamine-responsive megaloblastic anemia syndrome represents a rare autosomal recessive condition originating from mutations in the SLC19A2 gene. It is characterized by a classical triad of megaloblastic anemia, insulin-dependent diabetes mellitus, and sensorineural hearing loss. We present the case of a woman diagnosed with thiamine-responsive megaloblastic anemia, with no history of consanguinity, in whom genetic testing revealed novel SLC19A2 mutations and an unreported clinical manifestation.
CASE PRESENTATION: This case describes an 18-year-old mestizo female patient who pres

BACKGROUND: Hyperhomocysteinemia (HHcy) is an important risk factor for Alzheimer's disease (AD), but its differential effects on tau pathology and beta-amyloid (Aβ) deposition, as well as the key mediating molecules involved, remain unclear. This study investigates how HHcy influences AD pathology and examines whether interleukin-1β (IL-1β) neutralization can mitigate HHcy-accelerated neurodegeneration.
METHODS: Female 12-month-old 3 × Tg-AD mice were supplemented with methionine water for 7 weeks to induce HHcy. Brain tissues were analyzed for Aβ deposition, tau phosphorylation, oligomeriza

Many coregonine species have declined drastically across the Northern Hemisphere, including populations of Coregonus maraena (whitefish) in the Baltic Sea, and the mechanisms leading to these declines are not well investigated. An abrupt population crash occurred in the 1990s, coinciding with heavy declines in salmonid recruitment, also known as thiamine deficiency syndrome. Thiamine, i.e., vitamin B1, is an essential micronutrient needed for a functional metabolism. Offspring with thiamine deficiency have a high mortality posing significant negative impact on populations. Here, we aim to dete

BACKGROUND: Vitamin B12 deficiency is common in populations with limited animal-source foods and has been linked to delayed infant neurodevelopment and adverse pregnancy outcomes. Evidence on the benefits of maternal B12 supplementation for improving infant neurodevelopment remains mixed, particularly in low-income and middle-income countries where deficiency is prevalent.
METHODS: This double-blind, randomised controlled trial was conducted in two tertiary maternity care centres in India and Nepal. Pregnant women in their first trimester, following a vegetarian diet, were enrolled and random

Metabolic dysfunction-associated steatohepatitis (MASH) is a leading cause of cirrhosis and liver-related mortality, but it remains unclear how nutrient stresses drive coordinated transcriptional remodeling in the pathogenesis of MASH. Clinical studies reported that methionine and choline deficiency (MCD) promotes chronic liver diseases. Multiple types of MCD diets have been adopted to establish MASH mouse models. However, how MCD modulates cell-intrinsic transcriptional responses across parenchymal and nonparenchymal liver cell types, and whether these effects recapitulate human MASH, remains

RATIONALE: Although vitamin B12 deficiency is a well-known complication following total gastrectomy, subacute combined degeneration (SCD) rarely develops beyond 5 years postoperatively. Eight-year latency cases are rarely reported, especially when combined with chronic alcohol use, which accelerates neurologic decline and worsens prognosis. This report aims to present a rare case of delayed SCD after total gastrectomy in a patient with chronic alcoholism, emphasizing the importance of lifelong vitamin B12 monitoring in high-risk individuals.
PATIENT CONCERNS: A 49-year-old man presented with

Liver fibrosis, characterized by excessive extracellular matrix deposition, is a critical stage in chronic liver disease progression. Angiopoietin-like protein 6 (ANGPTL6), a hepatocyte-secreted protein involved in metabolism, has an unclear role in fibrosis. This study investigated ANGPTL6's function in hepatic fibrosis, focusing on its suppression of hepatic stellate cells (HSCs) activation via the transforming growth factor-beta 1/Smad3 (TGF-β1/Smad3) pathway. Mouse models of metabolic dysfunction-associated steatohepatitis (MASH) and fibrosis were established using a methionine-choline def
