Effect of Vitamin B12 Supplementation on Hematological and Athropometric Parameters in Children With Severe Acute Malnutrition, Aged 1 to 60 Months in Nutritional Rehabilitation Center (RCT).

Microcytic hypochromic anemia is common in severe acute malnutrition (SAM) children, but macrocytic anemia (vitamin B12/folate deficiency) is not uncommon. Vitamin B12 deficiency leads to anemia and developmental slowing. We looked for the response of vitamin B12 supplementation on hematological and anthropometric parameters in SAM children. All children aged 1 to 60 months with SAM were included and divided in 2 groups. Group A children received vitamin B12 for 6 weeks additionally as compared with Group B. Both groups were followed up at discharge and at 6 weeks for hematological and anthrop

Association between homocysteine level and unexplained recurrent pregnancy loss: a meta-analysis.

BACKGROUND: Unexplained recurrent pregnancy loss (uRPL) remains a major clinical challenge, with increasing evidence suggesting a potential role of hyperhomocysteinemia (HHcy) in its pathogenesis. However, findings from individual studies are inconsistent. A meta-analysis was therefore performed to summarize the findings. METHODS: A systematic search of PubMed, Embase, and Web of Science was conducted to identify observational studies evaluating the association between elevated homocysteine (Hcy) levels and uRPL. Pooled odds ratios (ORs) with 95% confidence intervals (CIs) were synthesized us

Chronic choline restriction remodels hepatic lipid metabolism and drives insulin resistance through a CD36-ETNPPL regulatory axis.

Chronic choline insufficiency reprograms hepatic metabolism and drives insulin resistance independent of obesity. While complete choline deficiency causes liver injury, the metabolic consequences of sustained, suboptimal intake, observed in ∼90% of US adults, remain poorly defined. Here, we used integrated lipidomic, metabolomic, and transcriptomic profiling to determine how graded choline intake (0.5, 1.4, or 6.3 g/kg) regulates hepatic metabolism during a control (Con) or high-fat (HF) diet-induced obesity regimen. Under Con diets, low choline intake induced a distinct metabolic state charac

Thiamine deficiency in patients with chronic migraine: A case-control study.

OBJECTIVES/BACKGROUND: Thiamine deficiency is well recognized in several neurological disorders, and subclinical deficiency has been associated with nonspecific symptoms, including headache. However, thiamine deficiency is not currently recognized as a cause of headache in standard headache classifications. Chronic migraine (CM) is often accompanied by symptoms such as nausea, vomiting, and reduced appetite, which may influence nutritional intake and micronutrient status, including thiamine depletion. These factors raise the possibility of an interaction between migraine and thiamine status. O

High Burden of Iron and Vitamin B12 Deficiency Among Apparently Healthy Indian Infants Aged 9-12 Months: A Secondary Analysis of Cross-Sectional Data.

Micronutrient deficiencies during infancy adversely affect neurodevelopment, growth and long-term health, yet often remain clinically silent and under-recognized. This retrospective cross-sectional secondary analysis estimates proportion of iron and vitamin B12 deficiency among apparently healthy Indian infants aged 9-12 mo enrolled in a vaccine trial and examines associations with anthropometric indicators. Among 137 infants without overt illness (mean age 9.2 mo, 60% males), ferritin, vitamin B12 and concurrent deficiencies were noted in 89.1%, 72.3%, and 65.7% respectively. Deficiencies wer

Severe Perioperative Lactic Acidosis and Hyperglycemia Responsive to Thiamine in a Patient With Prior Semaglutide Use and Restrictive Dieting: A Case Report.

Thiamine deficiency is a rare but potentially fatal cause of type B lactic acidosis, classically associated with alcoholism or severe malnutrition. Restrictive dieting and weight-loss regimens, including glucagon-like peptide-1 (GLP-1) receptor agonist use, may unmask thiamine deficiency under perioperative stress. We report a young, nondiabetic woman on a restrictive diet who developed severe lactic acidosis and hyperglycemia after total abdominal hysterectomy, with worsening lactate despite hemodynamic stability and adequate hydration. Empiric intravenous thiamine produced rapid normalizatio

Transcriptomic Profiling Reveals Inflammatory, Fibrotic, and Apoptotic Signatures in a Methionine-Choline-Deficient Diet-Induced Murine Model of Metabolism-Dysfunction-Associated Steatohepatitis.

Metabolic dysfunction-associated steatohepatitis (MASH; formerly non-alcoholic steatohepatitis, NASH) is characterized by oxidative stress, inflammatory activation, hepatocellular injury, and progressive liver dysfunction. However, the global transcriptomic landscape underlying stress-induced hepatic injury remains incompletely understood. In this study, we employed a methionine-choline-deficient (MCD) diet-induced murine model to characterize the phenotypic and transcriptomic alterations associated with liver injury. Male C57BL/6J mice were fed either a control or MCD diet, and hepatotoxicity

Disruption of branched-chain amino acids homeostasis exacerbates MCD-induced steatohepatitis by amplifying a pro-inflammatory positive feedback loop between hepatocytes and macrophages.

Accumulating evidence links branched-chain amino acids (BCAAs, including leucine, isoleucine, and valine) to obesity, diabetes, and related metabolic disorders; however, their role in metabolic dysfunction-associated steatohepatitis (MASH) remains unclear. Interrogation of human and mouse transcriptomic datasets (GSE147304, GSE263770) revealed suppressed hepatic BCAAs catabolic gene expression in MASH. In a murine model of MASH induced by a methionine- and choline-deficient (MCD) diet, BCAAs catabolic gene expression was suppressed in the liver, accompanied with the accumulation of branched-ch

Maternal vitamin B12 deprivation exacerbates offspring obesity by reducing early-life colonization with Bifidobacterium pseudolongum.

Vitamin B12 deficiency during pregnancy and lactation is common, yet its mechanistic impact on reproductive outcomes and offspring health remains poorly understood. Here, we show that maternal dietary vitamin B12 deprivation not only impairs maternal glucose metabolism and reproductive outcomes but also exacerbates high-fat-diet-induced obesity in offspring. These effects are mediated by gut microbiota and associated with a marked reduction of Bifidobacterium pseudolongum (B. pseudolongum) in both dams and their offspring. Maternal vitamin B12 deprivation limits early-life acquisition of B. ps

Choline insufficiency in mothers during gestation abolishes the metabolically restrained hepatic phenotype induced by gestational protein restriction and promotes hepatic steatosis in offspring.

Maternal protein restriction during gestation in rodents is widely used as a model of developmental programming; however, its metabolic effects in offspring are often modest. In humans, protein deficiency is frequently accompanied by deficiencies in essential micronutrients. This study examined whether maternal choline insufficiency, an essential micronutrient whose deficiency in adult rodents induces metabolic dysfunction-associated steatotic liver disease, modifies the metabolically restrained hepatic phenotype conferred by maternal protein restriction in offspring. Pregnant ICR mice were fe

Vitamin B12-mediated microglial immunometabolic reprogramming: A novel mechanistic insight into diabetes-associated cognitive impairment.

Diabetes-associated cognitive impairment (DCI) is an increasingly recognized neurological complication of type 2 diabetes mellitus characterized by chronic neuroinflammation and microglial immunometabolic dysregulation. Vitamin B12 (VB12) deficiency, which is highly prevalent in patients with diabetes, has been strongly associated with cognitive decline, hippocampal atrophy, and white matter injury. Emerging evidence suggests that VB12 plays a critical role in maintaining one‑carbon metabolism, mitochondrial function, and redox homeostasis. Mechanistically, VB12 deficiency promotes homocystein

Vitamin B12 deficiency, restless legs syndrome and poor sleep quality in adolescents: A cross-sectional study.

Although the role of dopaminergic dysfunction and iron metabolism in the pathophysiology of restless legs syndrome (RLS) has been widely investigated, the potential association between vitamin B12 status and RLS remains insufficiently explored. In particular, data in pediatric and adolescent populations are limited and inconsistent. Moreover, while sleep disturbance is a well-recognized feature of RLS, the combined relationship between vitamin B12 levels, RLS, and sleep quality has not been adequately evaluated in adolescents. Therefore, the present study aimed to investigate the association b

The Dynamic Coenzyme Network of B Vitamins in Nutritional Neuropathy and Neuropsychiatric Vulnerability: A Mechanistic Narrative Review.

Nutritional peripheral neuropathy is a clinically important condition associated with inadequate intake, malabsorption, bariatric surgery, aging, metabolic disease, and chronic medication exposure. B vitamins serve as essential coenzymes in mitochondrial energy metabolism, one-carbon metabolism, neurotransmitter synthesis, redox regulation, and myelin maintenance. This narrative review synthesizes mechanistic and clinical evidence linking B vitamin-dependent coenzyme systems to nutritional neuropathy and selected neuropsychiatric manifestations. Particular emphasis is placed on the tricarboxyl

Tao-Hong-Si-Wu-Tang protects against methionine- and choline-deficient diet-induced hepatic steatosis in rats and is associated with inhibition of oxidative stress, inflammation, apoptosis, and pyroptosis.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD), which is the most prevalent chronic liver disease, affects more than 30% of the adult population worldwide. Lifestyle interventions and management of metabolic comorbidities are the current main treatments. Thus, it is urgent to find candidate drugs for NAFLD. Traditional Chinese medicine (TCM) formulas have been extensively applied to treat NAFLD in China. Tao-Hong-Si-Wu-Tang (THSWT), a classic TCM formula with anti-oxidation, anti-inflammation, and anti-apoptosis, has emerged as an important formula for hepatoprotection. Based on its bio
