Vitamin D and gut microbiota in thyroid diseases: mechanisms and therapeutic potential.

Thyroid diseases have become the most common endocrine disorders globally, including hyperthyroidism, hypothyroidism, thyroid nodules (TN), and thyroid cancer (TC). Traditionally, the pathogenesis was attributed to autoimmune dysfunction, iodine metabolism imbalance, and genetic-environmental interactions. Recently, research suggests that vitamin D deficiency and gut microbiota dysbiosis are prevalent in patients with thyroid disorders, particularly in autoimmune thyroid disease (AITD). Vitamin D, as well as gut microbiota and its metabolites, such as short-chain fatty acids and bile acids, jo

Vitamin D status and supplementation as potential modifiers of allergen immunotherapy outcomes in respiratory allergy: A structured narrative review.

Allergen immunotherapy (AIT) represents a cornerstone in managing IgE-mediated allergic diseases, such as allergic rhinitis and asthma. By administering controlled doses of allergens over time, AIT aims to readjust immune tolerance, thereby modifying allergic responses. However, clinical responses to AIT remain heterogeneous, with a subset of patients achieving only partial benefit. This heterogeneity underlines the need for novel adjunctive approaches to potentiate the immunomodulatory effects of AIT. In this context, vitamin D has gained considerable interest, given its well-established role

Serum 25-hydroxyvitamin D levels are associated with thyroid diseases: A systematic review and meta-analysis.

BACKGROUND: Vitamin D, an important immunomodulator, has been implicated in the pathogenesis of thyroid disease. Although several studies have reported lower serum vitamin D levels in patients with autoimmune thyroid diseases (AITDS) and thyroid cancer, others have yielded shown inconsistent findings, leaving the clinical significance of vitamin D status in thyroid diseases unclear. This review aimed to clarify the association between serum vitamin D levels and thyroid diseases. METHODS: Observational studies, investigating the association between serum vitamin D levels and thyroid diseases,

Identification of risk factors for type 2 diabetes across age-based proxies of early and late postmenopausal stages in prediabetic women.

BACKGROUND: Prediabetes is increasingly prevalent, and women transitioning through menopause may face a particularly elevated risk of progressing to type 2 diabetes (T2DM). Estrogen deficiency promotes adiposity, insulin resistance, and inflammation, potentially amplifying the metabolic burden of aging. However, the determinants of T2DM in these women remain insufficiently characterized. METHODS: This retrospective study included 229 prediabetic women evaluated at a tertiary-care hospital and followed for up to 5.5 years. Prediabetes was defined according to ADA criteria and women were classi

Dose-dependent vitamin D3 supplementation attenuates endothelial dysfunction and renal injury in an obstructive sleep apnea rat model.

BACKGROUND: Endothelial dysfunction driven by intermittent hypoxia (IH)-induced oxidative stress, nitric oxide depletion, and adhesion molecule upregulation underlies obstructive sleep apnea (OSA)-related cardiovascular complications. Vitamin D deficiency, which frequently coexists with OSA, may further amplify this endothelial injury through impaired endothelial nitric oxide synthase (eNOS) regulation and heightened renin-angiotensin-aldosterone system (RAAS) activity; however, the dose-dependent therapeutic potential of vitamin D3 on combined endothelial and renal outcomes in an IH model rem

Vitamin D signaling in thymic development and longevity.

Vitamin D has pleiotropic physiological roles including immune system regulation. Notably, there are links between vitamin D deficiency and risk of autoimmunity. Hormonal vitamin D binds to the vitamin D receptor (VDR) to exert its genomic effects. In immune cells, these include regulation of activation, inflammatory responses, and cytokine production. However, vitamin D supplementation is not efficacious in treatment of existing autoimmune disease. Conversely, vitamin D intake during childhood is associated with reduced risk of developing autoimmune type 1 diabetes. This supports a prophylact

Familial occurrence of multiple sclerosis - current state of knowledge.

Multiple sclerosis (MS) is an immune-mediated, inflammatory, and neurodegenerative disease of the central nervous system with a complex, multifactorial etiology. Familial aggregation, along with a higher concordance rate in monozygotic than dizygotic twins, supports a significant, albeit non-Mendelian, genetic contribution to MS susceptibility. The strongest genetic association involves the human leukocyte antigen (HLA) region, particularly HLA-DRB1*15:01, while genome-wide association studies have identified over 200 additional susceptibility signals, most of which implicate immune regulation
