[Evaluation of a multidisciplinary care programme for people with obesity using patient reported outcome and experience measures (PROMs and PREMs)].

BACKGROUND: PROMs and PREMs complement clinical indicators by incorporating patient-reported health outcomes and experiences, respectively. However, their combined integration into the multidimensional assessment of care for individuals with obesity remains limited. The aim of this study is to evaluate, using PREMs and PROMs, patient-reported perceptions following a multidisciplinary intervention in individuals with obesity. METHODOLOGY: An observational, descriptive, cross-sectional study evaluating PREMs and PROMs using ad hoc, non-validated questionnaires answered by patients who completed

The Impact of Diabetes on Risk of Revision Following Anterior Cruciate Ligament Reconstruction: A Real-World Data Study.

INTRODUCTION: Given the increasing prevalence of diabetes mellitus (DM), understanding its impact on anterior cruciate ligament reconstruction (ACLR) revision risk is important for clinical decision-making and patient counselling. This study evaluated whether DM is associated with an increased risk of revision ACLR within 5 years of primary reconstruction. METHODS: We conducted a retrospective observational cohort study using a global federated health research network (TriNetX, Cambridge, USA), including 73,951 patients who underwent primary ACLR across 53 healthcare organisations, of whom 99

Protocol for the isolation, enrichment, and culture of murine adipose progenitor cells.

We present a protocol for isolating and culturing murine adipose progenitor cells (APCs) from adipose tissue. We describe steps for tissue dissection, unit-normalized enzymatic digestion, stromal vascular fraction (SVF) isolation, APC enrichment, and cell culture. We then detail quality control procedures that maximize cell yield, viability, and purity. This protocol combines enzymatic SVF isolation with magnetic-activated cell sorting enrichment to obtain a highly enriched APC population. It emphasizes reproducibility through unit-normalized digestion, controlled processing conditions, and st

Addressing the weight of the issue: gaps in obesity education among US osteopathic medical students.

CONTEXT: Obesity is a significant public health concern, with the United States experiencing some of the highest obesity rates worldwide. Despite the widespread prevalence and negative impacts on patient health, obesity education in medical school curricula remains insufficient. This training deficit contributes to a lack of preparedness among medical students in the management of obesity, leading to healthcare provider bias and inadequate patient care. OBJECTIVES: This study aims to identify potential gaps in obesity education within US osteopathic medical schools by assessing current osteop

GLP-1 receptor agonist treatment and cardiovascular outcomes following aortic valve replacement: a nationwide observational study.

BACKGROUND: GLP-1 receptor agonists reduce major adverse cardiovascular events among patients with diabetes, overweight, and obesity. Whether these benefits extend to patients undergoing aortic valve replacement (AVR) for severe aortic stenosis (AS) remains unknown. Thus, we investigated the association between GLP-1 receptor agonist treatment and cardiovascular outcomes after AVR. METHODS: Nationwide population-based cohort study including consecutive patients with severe AS undergoing TAVI or SAVR in Western Denmark between January, 2016 and April, 2022. Patients were followed until death,

Small area estimates of co-occurring health risk factors: A test of two synthetic population methodologies to estimate small-area prevalence of smoking, harmful alcohol consumption and obesity in England.

INTRODUCTION: Small area estimates in health research currently capture single indicators but fail to reflect concurrent overlaps in health risk factors like smoking, increased alcohol consumption, and obesity. Local governments need detailed estimates of these risk factors to plan public health intervention strategies and tackle health inequalities. However, local data are often unavailable or highly restricted. This paper addresses this critical data gap by providing novel small area estimations of co-occurring health risk factors in England. METHODS: To generate the local estimates, we emp

Feasibility and Effects of Exercise Snacks on Fitness and Body Composition in Women With Overweight or Obesity: A Randomized Controlled Trial.

BACKGROUND: We investigated the effects of exercise snacks (ES) on cardiorespiratory fitness, muscular strength, and body fat in women with overweight or obesity. METHODS: Twenty-seven women (age: 31.5 [4.6] y, body mass index: 25.5 [2.5] kg/m2) were randomized to an ES group (n = 13) or a nonexercising control group (n = 14). Participants in the ES group completed 3 supervised 1-minute bouts per day of stair climbing, jump squats, and high knees at 77% to 95% of maximum heart rate, with bouts separated by 1 to 4 hours, and performed on 3 days per week for 6 weeks. Real-world, digital-first delivery of GLP-1 receptor agonist therapy in women.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have demonstrated substantial weight reduction and cardiometabolic benefit in randomized trials; however, prospective implementation evidence from digital care models and female sex community populations remains limited. Here we report outcomes from an observational study of 8,127 women initiating GLP-1RA therapy (99.8% tirzepatide) through a structured digital program incorporating mandatory biochemical monitoring, weekly video-supported follow-up and clinician-guided dose titration in the UK. Mean weight reduction was 16.1 kg (95% confiden

CRISPR screens identify ACSL4 as a key regulator in macrophage activation and metabolic inflammation.

BACKGROUND AND PURPOSE: Pro-inflammatory macrophages drive obesity-associated metabolic inflammation. Identifying key regulators of their activation is crucial for new therapies. This study aimed to identify novel regulators of pro-inflammatory adipose tissue macrophage (ATM) activation and assess their therapeutic potential. EXPERIMENTAL APPROACH: We combined genome-wide CRISPR-Cas9 screening with scRNA-seq data from human adipose tissue and identified long-chain acyl-CoA synthetase 4 (ACSL4) as critical for pro-inflammatory ATM activation. Validation used myeloid-specific Acsl4 knockout mic

Divergent associations between adiposity and hernia subtypes in European populations: A Mendelian randomization study.

Obesity is a suspected risk factor for hernia, but observational studies are susceptible to confounding and reverse causation. We aimed to investigate the causal effects of general and central adiposity on 5 hernia subtypes using Mendelian randomization (MR). Two-sample MR analyses were conducted using genetic instruments for 7 adiposity traits from the UK Biobank and hernia outcome data (diaphragmatic, femoral, inguinal, umbilical, and ventral hernias) from the Finn Gen consortium. The inverse-variance weighted method was the primary analysis, complemented by MR Egger and weighted median meth

Policy themes and instruments in WHO and UNICEF child food-environment guidance, 2015-2025: a computational content analysis.

In children's food environments, global guidance can shape policy discourse and instrument choice, but its content does not establish implementation or outcomes. In 2015-2025, the World Health Organization (WHO) and the United Nations Children's Fund (UNICEF) published English-language guidance at the global level. A documented search, 2-author screening, and author-decision-masked expert review produced a corpus of 22 expert-refrozen documents (1,099 pages; 1,259 primary chunks). The BERTopic with document-capped discovery yielded six provisional clusters: food-marketing restrictions and chil

Oleanolic acid inhibits diet-induced adiposity through blocking neovascularization and macrophage accumulation involving Delta-like 4-Notch 1-nuclear factor-κB signaling.

BACKGROUND/OBJECTIVES: Obesity is a major risk factor in the pathogenesis of cardiometabolic disorders. Most of these obesity-related disorders are closely associated with vascular dysfunctions. Due to the increase of fat mass in obesity, oxygen deficiency is induced, thus developing adipose tissue vascularization. Additionally, local hypoxia generates adipokine expression and proinflammatory macrophage recruitment. This study was conducted to test efficiency of oleanolic acid on angiogenesis and proinflammation in diet-induced obese mice model. MATERIALS/METHODS: C57BL/6 mice were fed 10% fa
