INTRODUCTION: Hermansky-Pudlak syndrome (HPS) is a rare autosomal recessive disorder characterized by oculocutaneous albinism (OCA), ocular nystagmus, bleeding diathesis and, in certain forms, pulmonary fibrosis (PF). We report a case of genetically confirmed HPS type 1 (HPS-1) with pulmonary involvement in a patient with rheumatoid arthritis (RA).
PATIENT CONCERNS AND CLINICAL FINDINGS: A 62-year-old Caucasian woman, former smoker, was referred for progressive pulmonary fibrosis (PPF) documented since 2019. History included chronic kidney disease and RA diagnosed in 2022, treated with hydrox

Ocular manifestations in oculocutaneous albinism (OCA), a hereditary condition, are well documented. Retinal detachment in individuals with OCA is uncommon and poses challenges for retina specialists in terms of diagnosis and management. We reviewed the literature on the presentation and management of retinal detachment (RD) in oculocutaneous albinism (OCA). Identifying retinal breaks in the absence of retinal and choroidal pigments can be challenging. Chorioretinal sealing around retinal holes and breaks achieved through photocoagulation and/or diathermy is less effective compared to that ach

Griscelli syndrome type 2 (GS2), caused by biallelic RAB27A variants, is classically associated with hypopigmentation and life-threatening hemophagocytic lymphohistiocytosis (HLH). However, a few patients exhibit normal pigmentation, termed GS2 sine albinism. We identified two unrelated Japanese patients with this presentation caused by compound heterozygous RAB27A variants, representing the first functionally characterized cases among Japanese patients. Functional studies were performed using RAB27A-null cell systems to evaluate the two allele pairs: p.Trp73Arg/p.Val143Ala (patient 1) and p.S

We report the first pediatric case with molecular confirmation of TYR-related oculocutaneous albinism type 1 and FBN1-associated Marfan syndrome. The patient exhibited high myopia, diffuse myopic chorioretinopathy, bilateral foveal hypoplasia, and electrophysiologic evidence of optic pathway misrouting. Systemic findings included tall stature, mild scoliosis, and marfanoid features. Pathogenic variants were confirmed in both TYR and FBN1.The coexistence of these traditionally unrelated disorders broadens the phenotypic spectrum of both OCA1 and Marfan syndrome and suggests the potential involv

BACKGROUND: Although drugs are a common cause of hyperpigmentation, the pathogenesis is unclear and varies based on the offending agent. Classic medications associated with hyperpigmentation secondary to increased melanin deposition include tetracyclines, prostaglandins, nicotine, and antimalarial medications. We report the case of a 70-year-old female who developed photodistributed hyperpigmentation due to increased melanin deposition following COVID-19 vaccination.
CASE REPORT: A 70-year-old female presented with a one-year history of diffuse blue-grey hyperpigmentation in a photodistribute

STUDY OBJECTIVES: Sleep disorders are common non-motor symptoms of Parkinson's disease (PD) that significantly impact patients' quality of life. Specifically, alterations in sleep microstructure - such as reduced slow-wave activity and sleep spindles - are prevalent in PD. The locus coeruleus (LC), the brain's primary source of noradrenaline, plays a pivotal role in regulating sleep and wakefulness and is highly vulnerable to neurodegeneration in PD. This study explores whether disruptions in sleep microarchitecture in PD are linked to LC degeneration.
METHODS: We assessed polysomnography for

Tryptophan (Trp) metabolism follows three main branches: the kynurenine (KP), serotonin, and indole (IP) pathways. These pathways generate bioactive metabolites that regulate immune responses, redox balance, neurotransmission, metabolic homeostasis, inflammation, and circadian rhythms. A common theme across these pathways is the activation of the aryl hydrocarbon receptor (AhR). Several metabolites from KP, IP, and serotonin act as endogenous AhR ligands or interact indirectly with AhR, but the downstream consequences of this interaction depend on the cellular environment and inflammatory cont

Akkermansia muciniphila (A. muciniphila), a beneficial gut bacterium, has increasingly attracted interests in Alzheimer's disease (AD) research, its specific role in the microbiota-gut-brain axis still remains unclear. In this study, we demonstrated that A. muciniphila administration improve cognitive deficits and reduce amyloid-beta (Aβ) deposition in APP/PS1 mice, a transgenic model of AD. Subsequently, it is revealed that A. muciniphila administration significantly alters gut microbiota diversity and composition. Mechanically, our metabolomics analysis of cecal contents indicates A. mucinip

BACKGROUND: Obesity prevalence is increasing globally, accompanied by increases in obesity-related diseases. While obesity is usually defined by BMI, the development of obesity-related disease might be better characterized by specific adipose tissue (AT) distributions or body composition subphenotypes. Serum metabolite patterns reflecting AT distribution could provide insights into potential underlying pathophysiological pathways and the interplay between AT depots. We therefore aim to identify metabolite signatures associated with specific AT depots and body composition subphenotypes.

Objectives: Oculocutaneous albinism (OCA), a group of inherited disorders characterized by deficient melanin synthesis, leads to hypopigmentation of the skin, hair, and eyes. OCA type 2 (OCA2) is caused by mutations in OCA2. This study aimed to characterize the comprehensive clinical and genetic spectrum of OCA2, and to identify the key ocular determinants of visual acuity. Methods: We enrolled 90 probands clinically diagnosed with albinism. Whole-exome sequencing and comprehensive ophthalmic examinations were performed. Results: OCA2 was confirmed in 29 probands (32.2%). Visual impairment was

Albinism is characterized by a reduced or absent production of melanin in the skin, hair and eyes to a variable extent. Various inheritance patterns and genes can lead to a typical clinical phenotype. A distinction is made between three manifestations, oculocutaneous, ocular and syndromic forms of albinism. In an ophthalmological context, albinism is usually manifested in early childhood through nystagmus and visual impairment. Typical findings include iris transillumination, foveal hypoplasia and retinal hypopigmentation, each varying in severity. Children with albinism also present with stra

Media and humanitarian discourses surrounding violence against people with albinism in Tanzania have fostered forms of biosocial relatedness and public recognition. Yet albinism does not consistently consolidate into a stable biosocial identity. Drawing on ethnographic research with Farida, Baraka and other interlocutors with albinism, this article ethnographically develops the concept of "modalities of enfleshment" to attend to how albinism is lived through shifting interpretations and embodied experiences. In this paper, I argue that the condition offers a case study for understanding how bo

N6-methyladenine (6mA) DNA accumulates during chloroplast maturation in Agave angustifolia Haw. and covaries with METTL4 protein abundance, identifying 6mA as a candidate epigenetic mark associated with plastid biogenesis. Chloroplast development requires precise coordination of nuclear and organellar genomes, yet the epigenetic mechanisms regulating this process remain poorly understood. Here, we provide the first description of N6-methyladenine (6mA), a dynamic DNA modification, during Agave angustifolia Haw. chloroplast biogenesis. Canonical base editors (BEs) enable programmable nucleotide conversions without inducing DNA double-strand breaks (DSBs), yet the significant off-target (OT) effects restrict their applications. The transformer BE (tBE), a BE derivative, minimizes DNA and RNA OT mutations, but its dual single-guide RNA (sgRNAs) requirement limits its targeting scope. To address this, we integrated SpRY, a protospacer-adjacent motif (PAM)-relaxed Streptococcus pyogenes Cas9 variant, into tBE. The resulting tBE-SpRY achieved robust C-to-T editing across various cell lines in a PAM-unlimited manner, with minimal

Oculocutaneous albinism (OCA) is a genetic condition associated with impaired visual acuity and increased skin cancer risk. When OCA is due to defects in melanosome ion transport, abnormally acidic conditions in the melanosome lumen inhibit tyrosinase, the critical pigment synthetic enzyme. Hence, a therapeutic approach that optimizes melanosome pH to increase pigment production presents a potential treatment for OCA and a method for decreasing skin cancer risk. Here, we report that reduction in sAC ( ADCY10 ) activity via naturally occurring human variants in ADCY10 restores OCA pigmentation,

BACKGROUND: Albinism, a group of rare genetic conditions characterized by visual impairment and variable hypopigmentation, is extensively studied in biomedical research. However, the psychosocial aspects, particularly in France, remain under-investigated, contributing to the invisibility of this sensory disability.
OBJECTIVES: The ALBIPSY project aimed to explore the lived experience of persons with albinism (PWAs) and their significant others (SOs), and to examine the role of dyadic adjustment in maintaining their quality of life.
METHODS AND SETTINGS: This study employed a sequential mixed

BACKGROUND: Persons with albinism (PWA) in Ghana are disproportionately affected by climate change due to a lack of melanin in their skin, which increases their exposure and vulnerability to the effects of climate change. However, research on this group in Ghana remains limited.
PURPOSE: To explore the lived experiences of PWA regarding climate change knowledge, impact and adaptation strategies in the Sekondi-Takoradi Metropolis (STM).
METHODS: Data were collected from 17 PWA in the STM, using a five-item interview guide and analysed through constitutive phenomenological analysis. Recruitmen

Tyrosinase-related protein 1 (Tyrp1) is a melanosomal glycoprotein required for eumelanin biosynthesis through the oxidation of 5,6-dihydroxyindole-2-carboxylic acid (DHICA). Pathogenic variants in Tyrp1 cause oculocutaneous albinism type 3 (OCA3), but the molecular basis by which individual substitutions impair Tyrp1 stability and activity remains incompletely understood. Here, we examined wild-type Tyrp1 and three missense variants associated with OCA3: R356Q and R326H as OCA3-related variants, and D308N as a benign control; these were under conditions relevant to melanosome maturation. To a
