Albinism is characterized by a reduced or absent production of melanin in the skin, hair and eyes to a variable extent. Various inheritance patterns and genes can lead to a typical clinical phenotype. A distinction is made between three manifestations, oculocutaneous, ocular and syndromic forms of albinism. In an ophthalmological context, albinism is usually manifested in early childhood through nystagmus and visual impairment. Typical findings include iris transillumination, foveal hypoplasia and retinal hypopigmentation, each varying in severity. Children with albinism also present with stra

Media and humanitarian discourses surrounding violence against people with albinism in Tanzania have fostered forms of biosocial relatedness and public recognition. Yet albinism does not consistently consolidate into a stable biosocial identity. Drawing on ethnographic research with Farida, Baraka and other interlocutors with albinism, this article ethnographically develops the concept of "modalities of enfleshment" to attend to how albinism is lived through shifting interpretations and embodied experiences. In this paper, I argue that the condition offers a case study for understanding how bo

N6-methyladenine (6mA) DNA accumulates during chloroplast maturation in Agave angustifolia Haw. and covaries with METTL4 protein abundance, identifying 6mA as a candidate epigenetic mark associated with plastid biogenesis. Chloroplast development requires precise coordination of nuclear and organellar genomes, yet the epigenetic mechanisms regulating this process remain poorly understood. Here, we provide the first description of N6-methyladenine (6mA), a dynamic DNA modification, during Agave angustifolia Haw. chloroplast biogenesis. Using naturally occurring somaclonal variants-an albino phe

Canonical base editors (BEs) enable programmable nucleotide conversions without inducing DNA double-strand breaks (DSBs), yet the significant off-target (OT) effects restrict their applications. The transformer BE (tBE), a BE derivative, minimizes DNA and RNA OT mutations, but its dual single-guide RNA (sgRNAs) requirement limits its targeting scope. To address this, we integrated SpRY, a protospacer-adjacent motif (PAM)-relaxed Streptococcus pyogenes Cas9 variant, into tBE. The resulting tBE-SpRY achieved robust C-to-T editing across various cell lines in a PAM-unlimited manner, with minimal

Oculocutaneous albinism (OCA) is a genetic condition associated with impaired visual acuity and increased skin cancer risk. When OCA is due to defects in melanosome ion transport, abnormally acidic conditions in the melanosome lumen inhibit tyrosinase, the critical pigment synthetic enzyme. Hence, a therapeutic approach that optimizes melanosome pH to increase pigment production presents a potential treatment for OCA and a method for decreasing skin cancer risk. Here, we report that reduction in sAC ( ADCY10 ) activity via naturally occurring human variants in ADCY10 restores OCA pigmentation,

BACKGROUND: Albinism, a group of rare genetic conditions characterized by visual impairment and variable hypopigmentation, is extensively studied in biomedical research. However, the psychosocial aspects, particularly in France, remain under-investigated, contributing to the invisibility of this sensory disability.
OBJECTIVES: The ALBIPSY project aimed to explore the lived experience of persons with albinism (PWAs) and their significant others (SOs), and to examine the role of dyadic adjustment in maintaining their quality of life.
METHODS AND SETTINGS: This study employed a sequential mixed

BACKGROUND: Persons with albinism (PWA) in Ghana are disproportionately affected by climate change due to a lack of melanin in their skin, which increases their exposure and vulnerability to the effects of climate change. However, research on this group in Ghana remains limited.
PURPOSE: To explore the lived experiences of PWA regarding climate change knowledge, impact and adaptation strategies in the Sekondi-Takoradi Metropolis (STM).
METHODS: Data were collected from 17 PWA in the STM, using a five-item interview guide and analysed through constitutive phenomenological analysis. Recruitmen

Tyrosinase-related protein 1 (Tyrp1) is a melanosomal glycoprotein required for eumelanin biosynthesis through the oxidation of 5,6-dihydroxyindole-2-carboxylic acid (DHICA). Pathogenic variants in Tyrp1 cause oculocutaneous albinism type 3 (OCA3), but the molecular basis by which individual substitutions impair Tyrp1 stability and activity remains incompletely understood. Here, we examined wild-type Tyrp1 and three missense variants associated with OCA3: R356Q and R326H as OCA3-related variants, and D308N as a benign control; these were under conditions relevant to melanosome maturation. To a

Significant loss of pigmentation can increase visual disability, skin cancer risk, and psychosocial stress. Tyrosinase (TYR) catalyzes the first and rate-limiting step of melanin synthesis. Inhibitors of TYR are well established and are currently used in clinical settings; however, there is a dearth of direct activators of TYR. Here, using a human TYR construct, we developed high-throughput screening methods, in cell confirmatory assays employing 13C-tyrosine tracing, and computational analysis techniques, and identified ampyrone (4-aminoantipyrine) as a TYR activator. Ampyrone increased the i

Hermansky-Pudlak Syndrome (HPS) is a rare autosomal recessive disorder characterized by oculocutaneous albinism, platelet dysfunction, and bleeding diathesis. This case discusses the anaesthetic and hemostatic management of a paediatric patient with HPS undergoing scoliosis surgery, which has not been previously documented. A 15-year-old female patient with HPS presented with scoliosis requiring surgical correction. Key clinical findings included albinism, neutropenia, cardiovascular abnormalities, and prolonged platelet function test results. Preoperative assessments identified significant ri

The hepatotherapeutic agents from plants have become more predominant around the world. Evening primrose oil (EPO) is a popular alternative treatment that contains several phytoconstituents with strong anti-inflammatory and anti-oxidant properties. The current work inspected the potential protective effect of EPO against carbon tetrachloride (CCl4)-induced hepatotoxicity in rats. Twenty-four adult male albino Wistar rats were divided into 4 equal groups: group I received the vehicle only (control rats), group II was treated with EPO at 300 mg/kg/day, group III was treated with CCl4 at 1 mL/kg,

The Yangtze sturgeon (Acipenser dabryanus), a critically endangered living fossil whose wild populations are now extinct, faces new challenges to survival in captive breeding. Among these, the emergence of albino and gray color morphs raise fundamental questions about the molecular basis and physiological consequences of pigmentation loss. Here, we integrated histological, transcriptomic, and quantitative PCR to investigate pigmentation variation and associated immune alterations in this species. Histology revealed a complete absence of melanin in albino individuals and marked reduction in gra

BACKGROUND: People with albinism, a genetic condition characterized by reduced melanin production, have an increased risk of developing skin cancer due to their diminished photoprotection. The global prevalence of albinism is estimated at 1 in 17,000 individuals, but it varies significantly by region, being more common in certain parts of Africa. Despite this heightened vulnerability, data on the prevalence of skin cancer and its subtypes in this population remain limited.
OBJECTIVE: This study aimed to determine the prevalence of Skin Cancer (SC) and Actinic Keratosis (AK) among patients wit

Albinism is characterized by generalized hypopigmentation and ocular features resulting from impaired melanin biosynthesis. Most known pathogenic variants are rare (MAF < 0.001) and found in coding regions. The role of non-coding variants, especially those with higher allele frequencies, is generally not investigated. Our next generation sequencing panel that includes the entire sequence of five major albinism genes (TYR, OCA2, SLC45A2, GPR143 and HPS1) identified compound OCA2 heterozygosity in a patient with a rare (MAF:0,0003) coding variant, NM_000275.3:c.1025 A > G;p.(Tyr342Cys) and a mor

The molecular diagnosis of albinism is hampered by a significant number of genetic variants of unknown significance (VUS) including a majority of missense and in-frame insertion deletion variants. This contributes to the high rate of unresolved genetic diagnosis for this disease. We designed a straightforward test of missense VUS in albinism genes based on functional rescue. As a proof of concept, the assay was set up for testing variants in the TYR gene associated with oculocutaneous albinism type 1. The TYR gene was knocked-out in the human melanogenic MNT1 cell line and the resulting unpigm

PURPOSE: To examine the relationship between multifocal ERG (mfERG) topography and peak cone density measured with adaptive optics scanning light ophthalmoscopy (AOSLO) in persons with albinism (PWA).
METHODS: We obtained best corrected visual acuity (BCVA), mfERG, retinal imaging with AOSLO, and optical coherence tomography (OCT) data in seven PWA and one control. The relationship between central peak cone density, mfERG amplitude topography, and foveal hypoplasia (FH) grade was calculated using Spearman rank correlation. Parafoveal cone densities among a subset of PWA were compared to a pre
