Characterization of the rock pigeon type IV interferon system reveals conserved tyrosine-based signaling via the IFN-υR1/IL10RB complex.

Type IV interferon (IFN), a recently identified class of IFN, exhibits broad-spectrum antiviral and antibacterial functions across species. In this study, the type IV IFN gene, IFN-υ, and its cognate receptor subunits (IFN-υR1 and IL10RB) were functionally characterized in an avian species, the rock pigeon (Columba livia). Phylogenetic analysis showed that rock pigeon IFN-υ, IFN-υR1, and IL10RB were clustered closely with their respective orthologs in other vertebrates. Expression profiling revealed the constitutive transcription of these genes across multiple tissues. Furthermore, the express

Dietary Tryptophan Allocation in Depression: Serotonin-Kynurenine Balance, Microbial Indole Pathways, and Inflammatory Phenotypes.

Depression is defined by clinical symptoms, but its biology varies considerably among patients. In this review, dietary tryptophan allocation describes the routes taken by tryptophan after intestinal absorption. Some is incorporated into proteins. Some enters serotonin and melatonin synthesis or the kynurenine pathway, while gut bacteria convert another fraction into indole compounds. Brain availability also depends on the circulating free pool and competition with other large neutral amino acids. This term does not imply a new biochemical pathway. It allows these known processes to be conside

Natural polysaccharide-based molecularly imprinted polymers as sorbents for selective removal of amino acids as a potential approach for management of aminoacidopathies: Template and monomer selection study.

Phenylketonuria and histidinemia are metabolic conditions characterized by the buildup of certain amino acids resulting from enzymatic deficiencies. Approaches that diminish the intestinal uptake of these amino acids may assist in regulating their systemic concentrations. Molecularly imprinted polymers provide specialized recognition sites that facilitate the precise binding of particular molecules. Natural polymers like chitosan and alginate offer biocompatible frameworks that could improve the efficacy of these systems. Four molecularly imprinted polymers were created utilizing l-phenylalani

Thyroid hormone replacement is associated with improvement in renal dysfunction in patients with hypothyroidism: a retrospective observational study.

BACKGROUND: Hypothyroidism is a common endocrine disorder with multisystem involvement and is increasingly recognized as a potentially reversible contributor to renal dysfunction. However, renal impairment related to thyroid hormone deficiency is frequently overlooked or misclassified as chronic kidney disease in clinical practice. METHODS: In this single-center retrospective observational study, 40 hospitalized adults with newly diagnosed hypothyroidism were included. Renal dysfunction was defined by elevated serum creatinine and reduced estimated glomerular filtration rate at admission. All

Pharmacological Role of 3-O-Methyldopa in the Central Nervous System-Mediated Behavioral Effects of L-DOPA in Rats.

Long-term l-3,4-dihydroxyphenylalanine (l-DOPA) therapy for Parkinson's disease increases plasma concentrations of 3-O-methyldopa (3-OMD), a major metabolite of l-DOPA. This study aimed to investigate the role of 3-OMD in the central nervous system (CNS)-mediated behavioral effects associated with chronic l-DOPA treatment in rats. The effects of 3-OMD on CNS function were examined using two rat models: repeated oral co-administration of l-DOPA/benserazide and single or repeated intraperitoneal (i.p.) administration of 3-OMD. Behavioral assessments included spontaneous locomotor activity, spont

Real-Life Application of a Point-of-Care Biosensor for Phenylalanine in Patients With Phenylketonuria.

Phenylketonuria (PKU) is a rare metabolic disorder causing elevated phenylalanine (PHE) levels requiring lifelong dietary or pharmacological management and regular monitoring. Current PHE monitoring methods, such as tandem mass spectrometry (FIA-MS/MS), are laboratory and sample transport-dependent, leading to delays in obtaining results. This highlights the need for accessible point-of-care (PoC) solutions. Building on a previously developed bioluminescence-based PHE quantification method, we created a PoC PHE sensor integrated with a smartphone app (phenyx app) for real-time monitoring and d

Dietary conflicts with standard clinical procedures for colonoscopy in Phenylketonuria: a case report.

BACKGROUND: Colonoscopy is a diagnostic technique used for gastrointestinal tract pathologies that presents additional challenges when used in individuals with phenylketonuria (PKU). The dietary requirements for colonoscopy preparation conflict with a phenylalanine (Phe) restricted diet essential in classical PKU, and aspartame containing laxatives must also be avoided. This case study highlights the complexities of applying conventional colonoscopy protocols to patients with PKU and emphasis the importance of personalized preparation strategies that consider strict dietary requirements. Behavioral Phenotyping of the Pahenu2 Mouse Model for Phenylketonuria-A Scoping Review and Future Perspectives.

Phenylketonuria (PKU) is a rare metabolic disorder resulting from a mutation in the gene encoding the enzyme phenylalanine hydroxylase (PAH), resulting in very high phenylalanine (Phe) levels in blood and brain. A PKU mutant mouse model was developed via N-ethyl-N-nitrosourea (ENU) mutagenesis, mimicking the high brain Phe content seen in classic PKU patients. A large variety of behavioral tasks have been employed to explore the behavioral phenotype of the Pahenu2 mouse. The purpose of this scoping review is to provide an overview of the behavioral performance of PKU mutant mice. We addressed

[Hormones and skin pigmentation: fundamentals and clinical relevance].

Skin pigmentation by the endogenous pigment melanin is a highly coordinated process in which hormones play a crucial role. They are synthesized not only in classical endocrine organs but also in the skin itself, which acts as an independent endocrine organ. Among the endocrine target structures of the skin, the melanocortin 1 receptor (MC1R) is of particular importance. Via its high expression and tonic activity in melanocytes, as well as by binding to natural melanocortins such as α‑melanocyte-stimulating hormone (α-MSH), being generated in the skin following ultraviolet (UV) light irradiatio

The AP-1 adaptor complex is required for cell surface modifications and the survival of Cryptococcus neoformans in phagocytic cells.

The pathogenic yeast Cryptococcus neoformans causes life-threatening meningoencephalitis in individuals with compromised immune systems. The ability of the fungus to cause disease depends on key cell-surface features, such as a polysaccharide capsule that protects it from the mammalian immune system. However, the mechanisms by which C. neoformans traffics polysaccharide capsule, melanin, and other materials to the cell surface are poorly understood. In this study, we employed mutants lacking specific subunits of the adaptor protein complex 1 (AP-1) to investigate its role in the elaboration of

Pyridoxine/pyridoxal-5'-phosphate repletion to improve outcomes of alcohol withdrawal syndrome and promote long-term abstinence: a hypothesis.

INTRODUCTION: Deficiency of pyridoxal-5'-phosphate, the clinically measured and active form of pyridoxine (vitamin B6), is prevalent among those with alcohol use disorder. However, unlike thiamine, pyridoxal-5'-phosphate is not routinely repleted. Evidence is presented that its repletion would improve outcomes of alcohol withdrawal syndrome and promote long-term abstinence. PYRIDOXAL-5′-PHOSPHATE DEFICIENCY CAN CAUSE A CLINICALLY CONSEQUENTIAL GLUTAMATERGIC/GABAERGIC SIGNALING IMBALANCE: Excessive excitatory glutamatergic signaling and inadequate inhibitory GABAergic signaling are central to
