Left ventricular remodeling in atrial fibrillation is associated with elevated NAP-2 and 3-nitrotyrosine.

BACKGROUND: Atrial fibrillation (AF) coexists with left ventricular (LV) dysfunction and remodeling. Neutrophil infiltration of cardiac muscle tissue and oxidative stress are associated with cardiac remodeling. MATERIALS AND METHODS: In 221 AF patients aged 70 (62-76) years, LV remodeling was assessed using echocardiography. Plasma concentrations of neutrophil-activating peptide 2 (NAP-2) and 3-nitrotyrosine were measured. RESULTS: AF patients with concentric and eccentric hypertrophy showed higher NAP-2 levels than those with normal LV geometry (+41% and + 29%, p = 0.001 and p = 0.025, resp

Levothyroxine supplementation trials in preconception and pregnant women.

Thyroid disorders are common in women of childbearing age, representing the second most prevalent endocrine disorder in this population. Low thyroid function preconception and during pregnancy has been associated with a range of adverse outcomes including subfertility, miscarriage, preterm birth, and ongoing health problems in the neonate. Levothyroxine (LT4) treatment is routinely used in women with overt hypothyroidism (OH) to achieve a euthyroid status and consequently reduce the risk of these adverse outcomes. However, in other groups, such as those with subclinical hypothyroidism (SCH) or

Microbes, macrophages, and melanin: a unifying theory of disease as exemplified by cancer.

It is widely accepted that cancer mostly arises from random spontaneous mutations triggered by environmental factors. Our theory challenges the idea of the random somatic mutation theory (SMT). The SMT does not fit well with Charles Darwin's theory of evolution in that the same relatively few mutations would occur so frequently and that these mutations would lead to death rather than survival of the fittest. However, it would fit well under the theory of evolution, if we were to look at it from the vantage point of pathogens and their supporting microbial communities colonizing humans and muta

Familial Non-Autoimmune Hyperthyroidism Caused by an Extracellular Domain Variant (p.Leu267Phe) of the TSH Receptor.

INTRODUCTION: Non-autoimmune hyperthyroidism (NAH) is a rare genetic disorder caused by germline-activating variants in the TSH receptor (TSHR) gene. While most NAH-related TSHR variants are located in the seven-transmembrane domain (7TMD), three variants (p.Leu267Phe, p.Ser281Asn, and p.Asn406Ser) have been identified in the extracellular domain (ECD), with p.Leu267Phe previously not showing constitutively active in vitro. METHODS: We searched for TSHR variants in a Japanese family with NAH using PCR-based direct sequencing. We created three ECD variants (p.Leu267Phe, p.Ser281Asn, and p.Asn4

Molecules in the Serotonin-Melatonin Synthesis Pathway Have Distinct Interactions with Lipid Membranes.

The neurotransmitter serotonin is involved in physiological processes such as appetite, sleep, and mood and diseases such as anxiety and depression. Traditionally, the effects of serotonin were thought to be initiated by binding to its target transmembrane receptors. It is also known that serotonin can bind directly to the membrane with high affinity and modulate lipid dynamics, lateral segregation of lipids, vesicular association, and membrane protein activity. We investigated if other small molecules in the serotonin metabolic pathway, some of which are known to be signaling molecules while

Investigating dopaminergic abnormalities in schizophrenia and first-episode psychosis with normative modelling and multisite molecular neuroimaging.

Molecular neuroimaging techniques, like PET and SPECT, offer invaluable insights into the brain's in-vivo biology and its dysfunction in neuropsychiatric patients. However, the transition of molecular neuroimaging into diagnostics and precision medicine has been limited to a few clinical applications, hindered by issues like practical feasibility, high costs, and high between-subject heterogeneity of neuroimaging measures. In this study, we explore the use of normative modelling (NM) to identify individual patient alterations by describing the physiological variability of molecular functions.

The Roles of Neuroinflammation in l-DOPA-Induced Dyskinesia: Dissecting the Roles of NF-κB and TNF-α for Novel Pharmacological Therapeutic Approaches.

Levodopa-induced dyskinesia (LID) is a common and debilitating complication of long-term Parkinson's disease treatment. This review explores the roles of NF-κB and TNF-α signalling pathways in LID pathophysiology and potential therapeutic approaches targeting these mechanisms. Chronic levodopa treatment leads to aberrant neuroplasticity and neuroinflammation, involving activation of NF-κB and increased production of pro-inflammatory cytokines like TNF-α. NF-κB activation in glial cells contributes to sustained neuroinflammation and exacerbates dopaminergic neuron loss. TNF-α levels are elevate

Acridine Benzimidazolium Derivatives Induced Protective Microglia Polarization and In Silico TDP-43 Interaction─Potential Implications for Amyotrophic Lateral Sclerosis.

Abnormal protein aggregation and associated neuronal-glial cell cytotoxicity lead to a plethora of neurodegenerative disorders. Most of the earlier investigations on understanding neurodegenerative disease progression and cure focused on neuronal damage and restoration potential. With increased evidence on the role of glial cells like microglia and astrocytes in mediating these disorders, more studies are dedicated to understanding the role of inflammatory responses mediated by glial cells and how they lead to neuroinflammation. Amyotrophic lateral sclerosis (ALS) is a late-onset neurodegenera

Plasma tryptophan levels are linked to hippocampal integrity and cognitive function in individuals with mild cognitive impairment.

Tryptophan has been shown to improve cognitive functions, but whether these benefits emanate from changes in hippocampal structure or other mechanisms like enhanced serotonin pathways remains unclear. This study aimed to examine the relationship between tryptophan levels and hippocampal volumes in individuals with mild cognitive impairment (MCI) and to determine if changes in hippocampal volume correlate with cognitive function. A total of 499 individuals with MCI were recruited based on ADNI's clinical criteria. Cognitive function was assessed using the ADAS-Cog scale, and hippocampal volumes

Lactoferrin-modified organic-inorganic hybrid mesoporous silica for co-delivery of levodopa and curcumin in the synergistic treatment of Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) is a chronic neurodegenerative disorder primarily characterized by oxidative stress and dopaminergic neuron damage. While levodopa remains the cornerstone of PD treatment, its efficacy is limited by poor bioavailability and neuroprotective effects. Curcumin, a potent antioxidant derived from turmeric, demonstrates neuroprotective promise but also suffers from low bioavailability, hindering its therapeutic application. The combined therapeutic use of levodopa and curcumin offers a potential synergistic approach, though its neuroprotection potential through b

Characteristics of patients with Graves' disease who developed drug resistance and required surgery.

Although antithyroid drugs (ATDs) are the first-line treatment for patients with Graves' disease (GD) in Japan and other countries, some patients do not achieve remission due to drug resistance, leading to surgery. Even if ATD doses are increased, they often have uncontrolled thyroid function and enlarged goiters, necessitating high-risk emergency surgical treatment. In this study, we aimed to identify the characteristics of patients resistant to ATDs who underwent thyroidectomy and those who achieved remission. We retrospectively analyzed 45 patients with GD who underwent thyroidectomy and 73

The prognosis of severe subclinical hyperthyroidism with TSH below 0.1 μU/mL due to Graves' disease in the Japanese population.

To determine the prognosis of Graves' disease initially presenting with severe subclinical hyperthyroidism, we investigated 110 patients with Graves' disease with normal FT3 and FT4 levels and TSH below 0.1 μU/mL. Graves' disease was diagnosed based on the diffuse accumulation of radioiodine in the thyroid in 83 patients, while the other 27 patients were diagnosed based on positive anti-TSH receptor antibodies. Seventy patients did not receive immediate medical treatment for the hyperthyroidism. Forty-four patients developed overt hyperthyroidism after 1-131 (median 3) months. In 19 patients,

Progress in AAV-Mediated In Vivo Gene Therapy and Its Applications in Central Nervous System Diseases.

As the blood-brain barrier (BBB) prevents molecules from accessing the central nervous system (CNS), the traditional systemic delivery of chemical drugs limits the development of neurological drugs. However, in recent years, innovative therapeutic strategies have tried to bypass the restriction of traditional drug delivery methods. In vivo gene therapy refers to emerging biopharma vectors that carry the specific genes and target and infect specific tissues; these infected cells and tissues then undergo fundamental changes at the genetic level and produce therapeutic proteins or substances, thu

[Determination of four oxidative stress biomarkers in human urine using solid-phase extraction coupled with ultra performance liquid chromatography-tandem mass spectrometry].

Oxidative stress biomarkers are measurable biological indicators that reflect the balance between the production of reactive oxygen species (ROS) and the body's ability to neutralize them using antioxidants. Elevated oxidative stress is associated with a number of health effects. Herein, we report the development of a comprehensive and sensitive method for quantifying four typical oxidative stress biomarkers in human urine using solid-phase extraction (SPE) in conjunction with ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The quantified biomarkers include L,L-d

Shared early molecular mechanisms revealed in P301S and 5xFAD Alzheimer's disease mouse models.

Alzheimer's disease (AD) is the most common neurodegenerative disorder characterized by early molecular events that influence disease progression. Still, the molecular mechanisms caused by different mutations of AD are not understood. We have performed a multidisciplinary study to investigate and compare the early stages of the pathology in two transgenic AD mouse models: P301S and 5xFAD. Using SNOTRAP-based mass spectrometry, we assessed changes in S-nitrosylation, a nitric oxide-mediated post-translational modification, of proteins in both models during their juvenile age. The increased leve
