BACKGROUND: Cystinosis is a multisystemic lysosomal storage disorder caused by pathogenic variants in CTNS, the gene encoding cystinosin, a lysosomal transmembrane cystine transporter. In patients with cystinosis, cystine accumulates within lysosomes in all organs. The cystine-depleting agent cysteamine delays but does not prevent disease progression.
METHODS: In this phase 1-2, open-label, ongoing clinical study, we performed a preliminary assessment of CTNS-RD-04, which consists of autologous CD34+ cells transduced with lentiviral vectors carrying CTNS complementary DNA, in patients with cy

Altered systemic metabolism contributes to Alzheimer’s disease (AD) pathology, yet the links between specific metabolites, cortical amyloid deposition, and cognition remain unclear. This study conducted targeted profiling of over 600 serum metabolites across 26 biochemical classes in 746 ADNI participants (CN = 248, MCI = 403, AD = 95), identifying propionylcarnitine and methionine as significantly associated metabolites. Relationships among these metabolites, cortical Aβ accumulation, and cognitive performance were examined using regression and mediation analyses, adjusting for age, sex, educ

BACKGROUND: Albuminuria is associated with the progression of renal disease. Epithelial-mesenchymal transition (EMT) plays a significant role in cell migration and fibrosis. Podocytes may undergo EMT after injury, leading to podocyte migration and detachment, which results ultimately in defective glomerular filtration. Endoplasmic reticulum (ER) stress and reactive oxygen species (ROS) play major roles in EMT. The pathophysiology linking albuminuria to the progression of renal disease is complex and not yet fully understood. In this study, we evaluate whether podocytes can undergo EMT through

Airway mucus presents a significant barrier to inhaled drug delivery, particularly for nanoparticle-based interventions, with this barrier exacerbated in chronic respiratory diseases (CRDs) due to hyperviscous secretions and persistent inflammation. In this study, a dual-functional lipid-polymer hybrid nanoparticle was developed to combine rapid mucolysis with sustained anti-inflammatory activity, and its performance was evaluated using both conventional in vitro assays and a physiologically relevant lung-on-a-chip model. Dipalmitoylphosphatidylcholine (DPPC)-coated PLGA nanoparticles (hydrody

AIM: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major cause of chronic liver disease, yet its pathogenesis remains incompletely understood. Oxidative stress is thought to play a key role in MASLD progression. This study aimed to investigate the role of cystathionine γ-lyase (CSE), an enzyme essential for cysteine and glutathione (GSH) biosynthesis, in MASLD development.
RESULTS: Choline-deficient high-fat diet (CDHFD) feeding led to elevated aspartate aminotransferase, alanine aminotransferase, hepatic triglyceride accumulation, vacuolization, macrophage infiltratio

Hepcidin is a cysteine-rich peptide that functions both as an antimicrobial peptide and a central regulator of iron homeostasis in vertebrates. Structurally, it consists of an N-terminal signal peptide, a propeptide region and a C-terminal bioactive mature peptide. The canonical mature hepcidin contains eight conserved cysteine residues that form four disulfide bonds, stabilising a characteristic β-sheet hairpin-like structure. In the Antarctic toothfish (Dissostichus mawsoni), which inhabits extremely cold marine environments, a unique hepcidin variant was identified possessing only four cyst

α,β-Unsaturated carbonyl compounds in cigarette smoke, such as acrolein and acrylamide, are considered as possible causative factors for chronic obstructive pulmonary disease (COPD) in people who smoke cigarettes. A major route of metabolism of these compounds is reaction with glutathione to form adducts that are further metabolized and excreted in the urine as mercapturic acids, which serve as useful dosimeters of exposure. In this study, we analyzed urine samples from the Singapore Chinese Health Study, a prospective epidemiology study of 63,257 men and women 45-74 years old with Chinese ori

Reactive sulfur species (RSS) are increasingly recognized as important bioactive agents across phyla. Application of newly developed chemical tools, detection methods and multi-omics techniques has uncovered new and specific roles of endogenous RSS and revealed that a number of biological actions previously attributed to reactive oxygen species or reactive nitrogen species can also be mediated by RSS. This Review describes the versatile chemical biology of RSS with focus on persulfide formation on cysteine residues. We examine their pro-oxidant and antioxidant capacities, involvement in redox

Hydrogen sulfide (H2S) has emerged as a key neuromodulator in the central nervous system, yet its accurate quantification in the brain system remains challenging due to strong interference from endogenous thiols and other sulfur-containing species. Here, we report an excited-state intramolecular proton transfer (ESIPT)-based fluorescent probe designed for selective and sensitive detection of H2S. The probe incorporates a cyano-substituted carbamate unit that suppresses ESIPT, producing a distinct turn-on fluorescence response at 594 nm upon H2S-induced cleavage. The probe exhibits a strong lin

Parkinson's disease (PD) entails dopaminergic neuronal loss in a gradual manner, which is typically linked to oxidative stress, neuroinflammation, and the neurotransmitter homeostasis disruption. This research compares the neurochemical and neuroprotective impacts of lauric acid (LA), ribose-cysteine (RC), and levodopa (LD) in a transgenic Drosophila melanogaster model where human α-synuclein (α-syn) was expressed in dopaminergic neurons. The transgenic flies received treatment with LA, RC, or LD (250 mg/kg diet) for three weeks and were then evaluated by behavioral assays in conjunction with

Fetal Alcohol Spectrum Disorder (FASD) represents a major global public health concern, affecting approximately 7.7 per 1000 births worldwide and remains as the most common preventable cause of lifelong neurodevelopmental impairment. Despite its prevalence, current clinical interventions are largely symptom-supportive and fail to address the underlying developmental pathology, underscoring the need for targeted, mechanism-based therapeutic strategies. Given the central involvement of oxidative stress and inflammation in FASD pathogenesis, this study evaluated the protective efficacy of S-adeno

Anxiety disorders are highly prevalent mental illnesses characterized by pathological worry and anxiety, often comorbid with depression and insomnia, which complicate treatment. This study aimed to investigate the anxiolytic potential and underlying neural mechanisms of L-Cysteine in a mouse model of chronic restraint stress (CRS). Using behavioral tests across multiple dose groups, administration of medium-dose L-Cysteine (50 mg/kg) alleviated anxiety-like behaviors induced by CRS. Immunofluorescence staining showed that L-Cysteine reversed CRS-induced neuronal hyperactivation in the paravent

Colorectal cancer (CRC) incidence is increasing in many low- and middle-income countries, including Egypt, partly due to urbanization and lifestyle changes. Metaproteomic approaches remain underutilized in these settings. In this study, we applied fecal metaproteomics in Egyptian CRC patients to characterize disease-associated protein expression patterns, explore host-microbiota functional interactions, and identify metabolic pathways that are altered in the CRC gut environment. Stool samples from 10 CRC patients and 10 healthy controls were analyzed. A total of 441 differentially expressed pr

Cystinosis is a rare autosomal recessive lysosomal storage disorder that is caused by mutations in the CTNS gene. The hallmark of this disease is the accumulation of cystine within lysosomes, which functions as a pivotal diagnostic and monitoring biomarker. Cysteamine therapy has been demonstrated to reduce lysosomal cystine and improve outcomes; however, it does not fully halt progression, particularly renal decline. Consequently, preclinical research relies on diverse in vitro and in vivo models to explore mechanisms and test new treatments. Accurate intracellular cystine quantification is v

Diabetes Mellitus (DM) is a metabolic disease characterized by hyperglycemia resulting from impaired insulin secretion and resistance. Berberine (BBR) and N-acetylcysteine are anti-inflammatory and antiapoptotic compounds that have beneficial effects on diabetic complications. This study aimed to investigate the effects of BBR and NAC on the Hippocampus in an experimental diabetes model in rats. Rats were divided into 5 groups: control, diabetes (D), D + NAC, D + BBR, and D + BBR + NAC. STZ (45 mg/kg, i.p.) was administered to the other four groups except the control group. Homocystinuria (HCU) is an inborn error of metabolism and a conformational disorder chiefly caused by missense mutations in the cystathionine beta-synthase (CBS) gene. These mutations often cause CBS destabilization, misfolding and dysfunction resulting in CBS deficiency and pathological accumulation of homocysteine. Morphological changes in mitochondria were described in HCU patients and mouse models; however, their functional significance has remained unknown. Here, we characterized the impact of CBS deficiency due to expression of the most common HCU-causing variant CBS I278T on mitochondri

Influenza pneumonia is characterized by excessive inflammatory responses that contribute to severe lung injury and mortality. Supersulfides, endogenously produced cysteine-derived persulfides and polysulfides, exert potent antioxidant, anti-ferroptotic, and anti-inflammatory activities; however, their therapeutic potential after disease onset remains unclear. Here, we investigated the efficacy of N-acetylcysteine tetrasulfide (NAC-S2), a highly water-soluble and cell-permeable supersulfide donor, in a mouse model of influenza A virus (IAV)-induced pneumonia. Subcutaneous administration of NAC-
