Mass Spectrometry-Based Structural Proteomics and Analytical Ultracentrifugation Characterize the Structures of Oligomeric Apolipoprotein E4 and Its Variants.

Apolipoprotein E4 (ApoE4), one of the three common isoforms of ApoE, differs from ApoE3 and ApoE2 by just one amino acid substitution each. Yet these changes in ApoE4 profoundly impact protein structure and function to make it the strongest genetic risk factor for late-onset Alzheimer's disease (AD). ApoE4 exhibits a tendency to form oligomers (e.g., tetramers) that complicate high-resolution structural characterization in aqueous environments. Very little is known about the self-associated oligomeric structures. In this study, we employed ultracentrifugation, native mass spectrometry (MS), an

Suppression of pain transmission and behavior by inhibition of peripheral diacylglycerol metabolism.

Diacylglycerol lipase (DAGL) produces 2-arachidonoylglycerol (2-AG) and other proinflammatory lipids. Inactivation of DAGLs reduces the production of 2-AG, arachidonic acid (AA) and eicosanoids and elicits antinociceptive and anti-(neuro)inflammatory effects in rodents. However, inhibitors that enter the brain can cause significant central nervous system (CNS) side effects. Using activity-based protein profiling (ABPP), we report the discovery of A1480LS, a potent, in vivo active, small molecule dual inhibitor of DAGLα/β that is functionally biased to the periphery. We demonstrate that A1480LS

The role of inflammatory cells in the association between helicobacter pylori and metabolic syndrome: a cross-sectional study.

BACKGROUND: Helicobacter pylori (H. pylori) infection may be related to metabolic disorders, with inflammation activation considered as a potential link. The study investigated the association between H. pylori infection and metabolic syndrome (MetS), and explored the potential role of inflammatory cells in this association. METHODS: This study was conducted in a health check-up group from a tertiary hospital in northwest China. Logistic regression models were used to examine the association between H. pylori and MetS. Restricted cubic splines (RCS) were employed to examine both linear and no

APOE ε4 accelerates plasma p-tau217 trajectory: a Bayesian joint longitudinal analysis of ADNI.

BACKGROUND AND OBJECTIVE: Plasma GFAP and p-tau217 are leading blood biomarkers for Alzheimer's disease (AD), but whether APOE ε4 modulates their longitudinal rates of change - directly or via amyloid burden - is unresolved. We tested APOE ε4 modulation of plasma GFAP and p-tau217 trajectories and their coupling with cognitive/structural decline in ADNI participants. METHODS: A total of 325 ADNI participants aged ≥ 65 (CN 181/MCI 106/AD 38) with ≥ 2 plasma GFAP and ≥ 2 p-tau217 (UPENN-Fujirebio) measurements over ≤ 10 years were analyzed. Linear mixed-effects models tested APOE ε4 × time inte

Overinvestment in ultra‑rare APOE variants and highly speculative apoE-inhibitor docking risks undermining translational progress in sporadic Alzheimer's disease.

Misclassification of ultra-rare and isoform-defining APOE variants, together with unsupported attempts to dock small-molecule 'inhibitors' to apolipoprotein E (apoE), continues to appear in the structural bioinformatics literature. Such inconsistencies stem from a broader misunderstanding of apoE as a highly flexible, two-domain protein with extensive intrinsically disordered regions (IDRs), a context essential for interpreting sequence variation. A recurring issue is the treatment of the pathogenic ε4 and protective ε2 substitutions as independent rare variants rather than as the canonical AP

Cardiovascular risk factors in autoimmune thyroiditis: role of adiponectin, resistin, inflammation and auto-immunity - a cross-sectional study.

OBJECTIVE: Autoimmune thyroiditis (AIT) has been associated with an increased cardiovascular risk. Whether adipokines, inflammation and auto-immunity contribute to this increased risk is uncertain. Our aim was to evaluate the interrelationships of adipokines, inflammation, auto-immunity, and thyroid function with cardiovascular risk in autoimmune thyroiditis. METHODS: Sixty-two patients with AIT, with normal free T4 (FT4) and free T3 (FT3), without other comorbidities were studied. Patients were divided into two groups: AIT with thyroid-stimulating hormone (TSH) ≤2.5 mU/L and AIT with TSH >2.

Anti-inflammatory versus metabolic therapy in MASLD: a randomized clinical trial of aspirin and dapagliflozin using fibroscan outcomes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major hepatic consequence of metabolic dysfunction, yet evidence-based pharmacologic options remain limited. Repurposing widely available agents with metabolic and anti-inflammatory actions may offer a clinically meaningful benefit. To compare the effects of aspirin and dapagliflozin on hepatic steatosis, fibrosis, liver enzymes, and lipid profile in patients with MASLD. In this prospective randomized interventional study, 80 adults with confirmed MASLD were allocated (1:1) to aspirin 100 mg once daily or dapagliflozin 10 mg

Familial hypercholesterolaemia and thymic ageing in a Chinese cohort.

BACKGROUND AND AIMS: Lifelong exposure to elevated low-density lipoprotein cholesterol in familial hypercholesterolaemia (FH) patients not only increases the risk of atherosclerotic cardiovascular disease, but also affects other organs beyond the cardiovascular system. In a large FH cohort, this study investigated the effects of hypercholesterolaemia on the thymic output and explored whether lipid-lowering therapy can reverse these effects. METHODS: According to the Dutch Lipid Clinic Network criteria, 562 heterozygous FH patients (236 treatment-naïve, 326 on lipid-lowering therapy) and 295 a

Forecasting risk-factor distributions by sex and age, 1990-2050: a parametric estimation framework using GBD summary exposure values, demonstrated for Australia.

BACKGROUND: This paper presents a method to convert existing Global Burden of Disease (GBD) forecasts of summary exposure values (SEVs) into business-as-usual forecasts of risk-factor distributions. METHOD: SEVs from the GBD are a relative-risk-weighted summary of risk-factor exposure, ranging from 0 (everyone below the theoretical minimum risk exposure level) to 1 (everyone above the 99th percentile of global exposure). Given the known inputs to calculate SEVs, with one additional assumption of the coefficient of variation of the risk factor, it is possible to generate parametric distributio

Canonical and Non-Canonical Regulators of Lipid Metabolism in the Liver and the "Liver-Other Organs" Axes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common chronic liver disease and is closely associated with obesity, insulin resistance, and the recently conceptualized cardiovascular-kidney-metabolic syndrome. For decades, the pathogenesis of steatosis has been viewed primarily through the lens of insulin and the canonical transcriptional programs it regulates-sterol regulatory element-binding protein 1c, carbohydrate response element-binding protein, peroxisome proliferator-activated receptors, and AMP-activated protein kinase. At the same time, recent data point

Lipoprotein(a) and long-term revascularization burden: a recurrent-event analysis of the UK Biobank.

AIMS: To investigate whether elevated Lp(a) is independently associated with cumulative coronary, peripheral, and carotid revascularization burden in a primary prevention cohort. METHODS AND RESULTS: We analysed 237 729 UK Biobank participants of European ancestry free of ASCVD at baseline, followed for a median of 14.1 years. Lp(a) was analysed using the ESC/EAS 2025 threshold of 105 nmol/L and continuously (per 50 nmol/L increment). The primary outcome was the total number of coronary, peripheral, and carotid revascularization procedures per person. Given the genetically determined, lifelon

A Chemokine Profile Linked to Both Triglyceride-Rich Lipoproteins and Lipoprotein(a), But Not LDL, Is Associated With Cardiovascular Disease.

Triglyceride-rich lipoproteins (TRL) and lipoprotein(a) [Lp(a)] demonstrate heightened per-particle atherogenicity compared with low-density lipoproteins (LDL), yet mechanistic underpinnings remain unclear. Using Mendelian randomization in >46,000 UK Biobank participants, we linked genetically determined variation in TRL/remnants, LDL, and Lp(a) to 2,920 plasma proteins and found causal associations with 432, 58, and 111 proteins, respectively. Notably, TRL/remnants and Lp(a) both impacted chemokine-signaling and leukocyte-chemotaxis pathways, distinct from LDL's effect on lipid metabolism. A

Lowering of Lipoprotein(a) with Olpasiran.

Elevated lipoprotein(a) [Lp(a)] is associated with an increased risk of aortic stenosis and Atherosclerotic Cardiovascular Disease (ASCVD), including myocardial infarction. Therefore, Lp(a) has emerged as a potential target for reducing residual cardiovascular risk. Conventional lipid-lowering interventions, including lifestyle modifications such as diet and exercise and statin therapies, have not been shown to effectively reduce Lp(a) levels. Ezetimibe, which inhibits cholesterol absorption and can reduce LDL-C, does not significantly affect Lp(a) levels, even when used in combination with st

Efficacy and Safety of Enlicitide in Hypercholesterolemia: A Systematic Review and Meta-Analysis of Randomized Controlled Trials.

BACKGROUND: Atherosclerotic cardiovascular disease remains a leading cause of global mortality, with low-density lipoprotein cholesterol (LDL-C) reduction being the cornerstone of prevention. Despite effective therapies, many high-risk patients do not achieve recommended LDL-C targets. Enlicitide (MK-0616), an oral proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor, offers a potential alternative to injectable PCSK9-directed therapies. OBJECTIVE: To evaluate the efficacy and safety of enlicitide in adults with hypercholesterolemia and assess its dose-dependent effects on lipid pa

A rare case of familial chylomicronemia in infancy due to homozygous LMF1 variant.

Familial chylomicronemia syndrome (FCS) is a rare autosomal recessive disorder of triglyceride (TG) metabolism, with an estimated prevalence of 1 in 100 000 to 1 in 1 000 000. It typically presents in childhood or adolescence, while presentation in infancy is exceedingly rare. Loss-of-function variants in lipoprotein lipase gene account for 60-80% of FCS cases. Other causes of FCS include loss-of-function variants in glycosylphosphatidylinositol-anchored high-density lipoprotein-binding protein 1, apolipoprotein A-V, apolipoprotein C-II, and lipase maturation factor 1 (LMF1) genes. LMF1 varian

Biallelic ABCG8 Variants or Biochemical Evidence of Sitosterolemia: A Case Report.

Sitosterolemia is a rare lipid disorder characterized by xanthomas, premature atherosclerotic cardiovascular disease (ASCVD), and abnormally elevated circulating low-density lipoprotein cholesterol (LDL-c) measurements. Pathogenic variants in adenosine triphosphate (ATP)-binding cassette transporters lead to the accumulation of phytosterols. A child presented to the pediatric cardiology clinic with an incidentally noted presumed pathogenic variant and a variant of unknown significance in a sitosterolemia gene but had normal fasting lipids and no accumulation of circulating phytosterols. Elevated Lipoprotein(a) in Asian Populations and its Implications for Cardiovascular Disease.

Elevated lipoprotein (a) (Lp[a]) is becoming increasingly recognized as a cardiovascular disease risk factor. The relationship of Lp(a) to cardiovascular disease is well characterized in European populations; however, data are lacking in the Asian region. In this scoping review, we aimed to characterize the distribution and prevalence of elevated Lp(a) in the Asian region and its relationship to coronary artery disease, cerebrovascular disease, and peripheral vascular disease. Thirty-one Asian studies met the inclusion criteria and were compared with 6 representative European studies. The grea

Genome-wide synthetic lethality screen of Bam complex-associated genes in Escherichia coli.

Biogenesis of the bacterial outer membrane is key to bacterial survival and antibiotic resistance. Central to this is the β-barrel assembly machine (Bam) complex and its associated chaperones, which are responsible for transport, folding, and insertion of outer membrane proteins (OMPs). The Escherichia coli Bam complex is composed of two essential subunits, BamA and BamD, and three non-essential accessory lipoproteins, BamB, BamC, and BamE. Optimal Bam function is further dependent on the non-essential periplasmic chaperones DegP, Skp, and SurA. Despite intensive study, the specific function o
