β-alanine alleviates gout by inhibiting NLRP3 inflammasome activation.

The NLRP3 inflammasome plays a pivotal role in mediating pro-inflammatory cytokine release and inducing pyroptosis. Its aberrant activation is implicated in various inflammatory diseases, including gout, a condition characterized by monosodium urate crystal deposition in the ankle joint. Here, we identify β-alanine, an endogenous amino acid, as a novel NLRP3 inflammasome inhibitor with promising therapeutic potential for gout. Mechanistic investigations reveal that β-alanine binds to NLRP3, sequestering it within the trans-Golgi network. This interaction disrupts NLRP3 inflammasome assembly, t

Associations between oxidative balance and arthritis: insights from the National health and Nutrition Examination Survey.

Oxidative stress is implicated in arthritis development, but its associations may differ by arthritis type. This study examined the relationship between oxidative balance score (OBS) and the prevalence of rheumatoid arthritis (RA), osteoarthritis (OA), and gout in a nationally representative population. Data were obtained from the National Health and Nutrition Examination Survey (NHANES). RA and OA were analyzed using data from 2001-2018, and gout from 2007-2018. OBS was constructed from multiple dietary and lifestyle pro-oxidant and antioxidant factors. Multivariable logistic regression and r

AI for Anterior Segment Disease Using Transfer Learning: Adapting Slit-Lamp-Trained Model for Analysis of Smartphone Corneal Images.

PURPOSE: The automated classification artificial intelligence (AI) for anterior segment corneal diseases that we developed is capable of classifying images into nine categories, including vision-threatening corneal diseases, such as infectious keratitis; however, it has been trained exclusively on slit-lamp images. We aimed to develop an AI model adaptable to smartphone images by applying transfer learning using smartphone images to the existing AI model. AI trained with transfer learning on smartphone images will be referred to as "Phone-tuned AI." METHODS: This study included 2530 images ca

Better performance when recalling speed from object motion compared to texture motion.

Our daily visual environment abounds with motion. Accurately perceiving object speed is essential when avoiding collisions or intercepting moving objects. Accurately remembering object speed is equally essential, as moving objects may become temporarily occluded. Most studies of speed use texture motion stimuli (e.g., dot motion), which minimize spatial cues but do not reflect the natural spatiotemporal dynamics of moving objects. In contrast, object motion contains rich spatial cues that better capture real-world motion. Neural mechanisms underlying texture and object motion processing differ

Lymphatic pumping technique in mice alters blood parameters and metastatic melanoma in an age-dependent manner.

Therapeutic touch applied to primary tumors can increase metastasis. The goal of this project was to determine whether touch applied to metastatic tumors also increases metastasis. We evaluated touch on a mouse model of experimental metastasis using a manual treatment called Lymphatic Pumping Technique (LPT), which increases lymphatic fluid flow and is contraindicated in patients with cancer. The LPT, or a sham treatment, was administered for 5 minutes while the mice were anesthetized with vaporized isoflurane. Young adult (3 months old) and aged (20-24 months old) mice received daily sham or

Using Multi-Omics Methods to Understand Gouty Arthritis.

Gouty arthritis is a common arthritic disease caused by the deposition of monosodium urate crystals in the joints and the tissues around it. The main pathogenesis of gout is the inflammation caused by the deposition of monosodium urate crystals. Omics studies help us evaluate global changes in gout during recent years, but most studies used only a single omics approach to illustrate the mechanisms of gout. In this review, we review the genomics, transcriptomics, epigenetics, proteomics, and metabolomics of gout, observing that different genes, DNA methylation, miRNAs, LncRNAs, circRNAs, protei

Colchicine-related effects on gene and protein expression in gout.

PURPOSE OF REVIEW: Gout is an inflammatory arthritis mediated by monosodium urate deposition in synovial tissue. Colchicine is a common treatment for acute gout flares and prophylaxis, with well established efficacy but an incompletely understood mechanism. Investigating colchicine's impact at a proteomic and transcriptomic level may highlight these mechanisms. Interest in colchicine's effect on atherosclerotic cardiovascular disease (ASCVD) has stimulated additional research in this area. RECENT FINDINGS: Proteomic studies have helped identify gout's inflammatory signature, supporting prior

Off-flavors in fish: origins, detection, and mitigation strategies.

The purpose of this study was to characterize the short-term effects of Achilles tendon ruptures on medial gastrocnemius. We hypothesized that the fascicles of the medial gastrocnemius muscle of the injured Achilles tendon would be shorter and more pennate immediately following the injury and would persist throughout 4 weeks post-injury. B-mode longitudinal ultrasound images of the medial gastrocnemius were acquired in 10 adults who suffered acute Achilles tendon ruptures and were treated non-operatively. Ultrasound images were acquired during the initial clinical visit following injury as wel

Uncommon Presentation of Giant Tophaceous Gout in the Hand: A Case Report.

BACKGROUND: Tophaceous gout masses are characterized by the accumulation of monosodium urate crystals in peripheral joints and soft tissues. The most commonly involved areas are the metatarsophalangeal and knee joints. Finger/hand localization is uncommon. If not correctly treated, a finger tophaceous mass can grow and, in rare cases, reach an abnormally large size, termed "giant." AIM: The aim of our study is to present a rare case of a large tophaceous mass of the hand, localized in the fourth finger, and to highlight the role of surgical excision combined with a multidisciplinary team appr

Do Monosodium Urate Crystals Reduce at Different Rates in Joints and Tendons During Urate-Lowering Therapy? A Dual-Energy Computed Tomography Study.

OBJECTIVE: Prior imaging studies have suggested that monosodium urate (MSU) crystal deposits in joints dissolve more rapidly than those in tendons during urate-lowering therapy (ULT) for gout. This study aimed to examine whether urate deposits visible on dual-energy computed tomography (DECT) reduce at different rates in joints and tendons during ULT. METHODS: Participants with gout from 2 clinical trials of oral ULT with the following criteria were included: paired DECT scans of the feet and ankles over 1-year of ULT, first DECT scan showing total urate volume ≥ 0.5 cm3, second DECT scan sho

Changes in rat intestinal motility and absorption following intestinal transplantation: an experimental study.

OBJECTIVE: Fibromyalgia is a chronic disorder with a very complex symptomatology. Although generalized severe pain is considered to be the cardinal symptom of the disease, many other associated symptoms, especially non-restorative sleep, chronic fatigue, anxiety, and depressive symptoms also play a relevant role in the degree of disability characteristic of the disease. Ozone therapy, which is used to treat a wide range of diseases and seems to be particularly useful in the treatment of many chronic diseases, is thought to act by exerting a mild, transient, and controlled oxidative stress that

Functional studies of human variants in C. elegans link iron metabolism to DPD deficiency and 5-FU sensitivity.

Dihydropyrimidine dehydrogenase (DPD), encoded by DPYD, is a key enzyme in pyrimidine catabolism, and its deficiency leads to severe toxicity in patients treated with 5-fluorouracil (5-FU). While pathogenic DPYD variants account for many cases of DPD deficiency, they do not fully explain all instances of 5-FU sensitivity, suggesting additional genetic factors are involved. Recent studies have implicated variants in CIAO1, a gene encoding a subunit of the cytosolic iron-sulfur (Fe-S) cluster assembly targeting complex, in reducing DPD stability and function. In this study, we established a C. e
