Risk factors associated with acquiring gastrointestinal infections in UK international travellers: a case-control study.

International travel is thought to be a major risk factor for developing gastrointestinal illness in England. Transmission is thought to be more likely in countries which have lower food hygiene standards, poorer sanitation, and lack of access to clean water. However, many studies are conducted within travel clinic settings which may bias findings. Here, we present a case-control study undertaken in returning English travellers in the community conducted with cases of gastrointestinal illness notified to UKHSA.All Cryptosporidiosis, Giardiasis, non-typhoidal Salmonellosis, and Shigellosis case

Shigella flexneri evades septin-mediated cell-autonomous immunity via protein ADP-riboxanation.

Cell-autonomous immunity represents evolutionarily conserved defense mechanisms present in both immune and non-immune cells. One of such mechanisms is mediated by cytoskeletal septins that entrap cytosolic bacterial pathogens within cage-like structures. To promote infection, Shigella flexneri delivers effector proteins directly into host cells via a type III secretion system. Here we demonstrate that OspC effectors enable Shigella flexneri to evade septin cage entrapment. Mechanistically, OspC catalyzes ADP-riboxanation of SEPT9 at Arg561, a site essential for stabilizing septin hetero-oligom

Berberine Alleviates Shigella-Induced Dysentery by Regulating Intestinal Barrier and Inflammatory Responses.

Berberine (BBR), an isoquinoline alkaloid, has a long history of clinical use in treating dysentery. However, its precise mechanism has not been fully elucidated. This study aimed to investigate the intestinal protective mechanisms of BBR against Shigella flexneri (S. flexneri)-induced dysentery in mice. We found that BBR significantly upregulated the intestinal barrier proteins ZO-1, occludin, and E-cadherin, enhancing intestinal mucosal integrity to inhibit S. flexneri invasion. Moreover, BBR effectively attenuated M1 macrophage polarization and restored the Th1/Th17/Treg balance to reduce i

Immunogenicity evaluation of altSonflex1-2-3 Shigella vaccine across mice, rats, and rabbits to inform human translational insights.

Shigella, a leading cause of bacillary dysentery, represents a significant global health challenge, particularly in low- and middle-income countries. Shigellosis predominantly affects children under the age of five and is associated with high morbidity and mortality rates. To address this burden, a generalized modules for membrane antigens (GMMA)-based vaccine, altSonflex1-2-3, incorporating S. sonnei and S. flexneri 1b, 2a, and 3a O-antigens, has been developed. This study aimed to evaluate and compare the immunogenicity of the altSonflex1-2-3 vaccine in mice, rats, and rabbits. Significant i

Global distribution and genomic architectures of plasmid-mediated quinolone resistance genes in Shigella from 1998 to 2025.

OBJECTIVE: To perform genomic analysis on plasmid-mediated quinolone-resistance (PMQR) in Shigella. METHODS: We assembled a global collection of 8325 PMQR-positive Shigella isolates (1998-2025) and subjected them to comprehensive genomic analysis. RESULTS: Geographically, the isolates spanned 37 countries, with the majority sourced from the United States (47.02%) and the United Kingdom (32.28%). Eight distinct PMQR genes-aac(6')-Ib-cr, oqxAB, qepA, qnrA, qnrB, qnrD, qnrS, and qnrVC-were identified in the Shigella analysed. qnrS was the most predominant PMQR gene (62.05%), followed by qnrB (3

An In Silico-Designed Chimeric Antigen Confers Broad Protection Against Major Diarrheal Pathogens (Shigella, ETEC, and EHEC) in Preclinical Models.

Diarrheagenic E. coli (DEC) and Shigella species are leading causes of under-5 mortality globally, prompting the development of a broad-spectrum vaccine. Here, we rationally designed a novel chimeric antigen ("SEISL") incorporating five virulence determinants: StxB and EspA (EHEC), LTB and a detoxified STa (ETEC), and the N-terminal domain of IpaD (Shigella). Bioinformatically optimized for maximal antigenicity, SEISL was expressed in E. coli BL21(DE3), purified from inclusion bodies under denaturing conditions, and confirmed by western blot. Immunization with SEISL + Freund's adjuvants in pat

Optimizing Shigella isolation: a multi-site evaluation of laboratory culture methods for Shigella detection, speciation, and serotyping with different transport media and sample types in the Enterics for Global Health study.

Shigella is a leading cause of diarrhea and dysentery in children younger than 5 years of age in low-resource settings, and several vaccines are in development. Due to its fastidious nature, Shigella can be difficult to culture, and eventual vaccine trials will need to optimize the isolation of Shigella to ensure efficient sample sizes. In the recently concluded Enterics for Global Health (EFGH) Shigella Surveillance study, we compared Shigella culture isolation rates between rectal swab vs. whole stool, between two swabs vs. one, and between Cary Blair (CB) vs. modified-Buffered Glycerol Sali

Host response to bacteria induces a shift towards the enteroendocrine cell lineage in the murine enteroid model.

The host response to commensal and pathogenic bacteria has been extensively characterized using human cancer cell line models but remains less defined in primary intestinal cellular systems. Recent evidence has demonstrated that mice lacking the Nod-like receptor (NLR) protein NLRC4 are susceptible to Shigella flexneri infection and thus represent a new model to study the mechanistic aspects of S. flexneri-host interaction. Using ileal organoids from wild-type (WT) and Nlrc4-/- mice, we first confirmed that NLRC4 was required for the restriction of intracellular S. flexneri growth. Surprisingl

Integrated Morphological, Functional, and Genomic Analysis of a Novel Lytic Phage (VIT_AJPSC) Infecting Multidrug-Resistant Shigella dysenteriae.

Bacteria are historically known to cause a disproportionate number of deaths because of infectious diseases worldwide. Annually, 55 million deaths are accounted for because of bacterial infections, out of which approximately 3 million deaths are because of diarrhoea and dysentery. Shigellosis is an infectious disease caused by Shigella species, and it targets the intestinal tract. It is characterised by acute diarrhoea mixed with blood. The oral antibiotics recommended for shigellosis are ciprofloxacin and azithromycin, with ciprofloxacin as the first-line treatment. Recently, Shigella species
