MeCP2 functions as a DNA methylation reader in immune homeostasis and cancer.

DNA methylation is a key epigenetic mechanism that mediates gene expression and informs cellular identity. Methyl-CpG binding protein 2 (MeCP2) is a protein capable of recognizing and interpreting the methylated DNA code to bring about phenotypic outcomes through co-factor interactions. Altered DNA methylation patterns and MeCP2 function, therefore, disrupt cellular epigenetic homeostasis and contribute to human diseases. MeCP2 loss-of-function or amplification events are responsible for Rett syndrome and MECP2 duplication syndrome, respectively, where patients present with multisystem pathoph

Designing and Delivering a Dissemination Programme With Co-Researchers With Intellectual Disabilities.

BACKGROUND: While dissemination is core to the research cycle, producing accessible materials has received scant attention. Having finished a co-designed project we wanted to share our findings with a range of audiences. METHOD: Using the community engaged research dissemination (CERD) framework helped identify a range of dissemination audiences and appropriate materials. Various dissemination materials were co-created with facilitators. Focus groups and interviews explored people's experiences of taking part in this process and analysed using Template Analysis. RESULTS: A dissemination matr

Development and external validation of a machine learning model for three-tier disability risk stratification in older adults.

BACKGROUND: Global aging has made disability among older adults a major public health challenge. Existing prediction models often lack external validation and actionable risk stratification. This study aims to develop and externally validate an explainable, three-tier machine learning framework for disability risk prediction and stratification among older adults. METHODS: The training set and internal validation set were derived from the longitudinal development cohort of the China Health and Retirement Longitudinal Study (CHARLS, n = 3,599). An independent cross-sectional survey (n = 2,066)

Design and validation of a multisensory game-based tooth brushing education tool for children with visual and hearing impairment.

BACKGROUND: Children with sensory disabilities exhibit a poor oral hygiene and a high prevalence of gingivitis and dental caries. Tailored multisensory oral health education interventions may improve tooth brushing skills and oral health. To develop and validate a multisensory game-based educational intervention to teach tooth brushing to children with visual and hearing impairments. MATERIALS AND METHODS: A developmental research design was used. A prototype was developed after literature review and expert interviews. This was validated by dentists and special educators and field tested amon

Reconnecting the disconnected forebrain: multimodal resting-state bold dynamics underlying zolpidem-induced arousal in incomplete Locked-in syndrome.

PRIMARY OBJECTIVE: Zolpidem paradoxically restores behavioral responsiveness and communication in select patients with severe disorders of consciousness and profound motor deafferentation. While the anterior forebrain mesocircuit hypothesis attributes this awakening to central thalamic disinhibition, macroscopic network dynamics and lower brainstem gating remain under-characterized in locked-in syndrome (LIS). METHODS AND PROCEDURES: A 45-year-old male with ventral pontine necrosis initially misdiagnosed as vegetative state/unresponsive wakefulness syndrome (VS/UWS) was formally identified wi

Assessing formal service providers' readiness to address IPV among women with disabilities in Sweden: A mixed-methods analysis.

Women with disabilities face a heightened risk of intimate partner violence (IPV), yet formal support systems often remain ill-equipped to address their specific needs. This study assesses indicators of readiness among formal service providers in Sweden, including healthcare, social services, shelters, the police, and the Centre against Violence, to support women with disabilities who experience IPV. Employing a sequential exploratory mixed-methods design, the study combined qualitative interviews (N = 18) with a national survey (N = 1,151). Data analysis was guided by an adapted version of Co

Coexisting Hearing Impairment in Children Presenting With Speech and Language Delay: A Case Series Highlighting the Importance of Routine Audiological Assessment.

BACKGROUND: Speech and language delay is a common reason for referral to developmental pediatric services. Hearing impairment is an important and potentially manageable cause but may be overlooked because its clinical presentation often overlaps with autism spectrum disorder (ASD), attention-deficit/hyperactivity disorder (ADHD), and other neurodevelopmental disorders. In Myanmar, where universal newborn hearing screening has not yet been routinely implemented, delayed recognition of hearing impairment remains an important clinical challenge. CASE PRESENTATION: We describe four children aged

Reliability of Childhood Autism Rating Scale, Second Edition, Assessments Using Telepsychiatry for Children With Neurodevelopmental Disorders and Their Caregivers: Randomized Feasibility Study.

BACKGROUND: The COVID-19 pandemic has increased the demand for telepsychiatry in neurodevelopmental assessment; however, few studies have examined the interrater reliability of clinician-rated autism scales, such as the Childhood Autism Rating Scale, Second Edition (CARS2), when used remotely. Although the feasibility and validity of telehealth assessments have been explored previously, the consistency between remote and in person CARS2 ratings by different clinicians remains underinvestigated. OBJECTIVE: The objective of this study was to evaluate agreement between CARS2 total scores obtaine

The ADHD advantage: Comorbid attention-deficit/hyperactivity disorder is associated with earlier diagnosis and better academic outcomes in children with specific language disorder.

Specific learning disorder (SLD) is a neurodevelopmental disorder characterized by difficulties in reading, writing, and mathematics despite normal intelligence. One of the most common comorbidity of SLD is attention-deficit hyperactivity disorder (ADHD). However, differences between patients with SLD alone and SLD with ADHD remain underinvestigated. This study aims to compare clinical and psychosocial characteristics between these two groups. This retrospective study analyzes pediatric psychiatric patient data from Research Electronic Data Capture (REDCap) at the Learning Disorder Outpatient

Etiological Diagnoses Versus Descriptive Developmental Diagnoses.

Jane is a 7-year-old girl who presented for a developmental-behavioral pediatrics (DBP) evaluation because of parental concern about developmental delays and atypical behavior. Jane is a former term infant who was small for gestational age. She experienced some initial feeding difficulties and was discharged home from the nursery at 5 days of age. As a toddler, Jane was mildly delayed in acquiring gross motor developmental milestones, but she was more significantly delayed in acquiring fine motor and adaptive developmental milestones. She began receiving private occupational therapy (OT) servi

Neural Cell Models of Specific Reading Disabilities: Identification of Cellular and Transcriptome Alterations.

Based on post-mortem studies, a long-standing hypothesis on the biological bases of specific reading disabilities (RD) has been that alterations in neuronal migration alter language neural circuits. However, there are questions concerning the original studies (small sample size, comorbidities). Further, this idea has not been strongly supported by current genetic studies. In this study, we test altered neural cell phenotypes using cells derived from two pairs of siblings strongly discordant for reading-probands with severe RD and their strong-reader siblings. We observed significant difference

Artificial intelligence and twice-exceptional (2e) learners in educational settings: a scoping review of an emerging evidence base (2020-2026).

Twice-exceptional (2e) students are intellectually or creatively gifted and also have one or more disabilities, and they require provision that develops talent and addresses disability at the same time. Artificial intelligence (AI) is increasingly promoted as a route to personalized instruction, and its adaptive character appears conceptually well suited to the heterogeneous profiles of 2e learners. Whether that promise has been examined empirically with this population has not been established. This scoping review asked whether and how AI technologies have been reported in educational researc

Supporting Neurodivergent Learners in Pharmacy Education: a Commentary.

This commentary discusses the importance of embracing neurodivergent (ND) learners in pharmacy education, identifies barriers and gaps within current educational systems, and offers practical strategies to foster all-embracing and fair learning environments across didactic and experiential settings. Literature about ND learners in pharmacy education is limited. Despite legal requirements under the Americans with Disabilities Act and related legislation, institutions remain unprepared to support ND learners, with persistent gaps between policy and practice. Common reported barriers include defi

An automated machine vision-based index for Rett Syndrome provides a generalizable framework for modeling rare disease therapeutics.

Rare diseases collectively affect millions of people, yet therapeutic development remains limited by preclinical phenotyping which is often dependent on subjective, low-resolution manual behavioral assessment. Rett syndrome (RTT), a rare disease affecting 1 in 10,000 female births, is caused by a MeCP2 deficiency. In mouse models for this disease, symptom severity is measured primarily with the observational Bird score. This subjective six-item ordinal scale was used to demonstrate that MeCP2 deficiency is reversible, but offers limited resolution for ranking candidate therapies or detecting p
