Brain functional mediation of vitamin D and development in children with autism spectrum disorder.

BACKGROUND: Children with autism spectrum disorder (ASD) often present with developmental delays and brain functional abnormalities. Vitamin D is thought to play an important role in neurodevelopment and brain function, but its association with brain functional metrics in ASD remains unclear. This study aimed to examine the relationship between serum 25-hydroxyvitamin D [25(OH)D] levels, brain function, and developmental outcomes in children with ASD. METHODS: We included 207 children with ASD and 209 typically developing (TD) children. Serum 25(OH)D levels were measured in all participants.

Neurogenic colonic motility patterns and gut transit disrupted in a mouse model of Rett Syndrome.

Gastrointestinal dysfunction is a prominent feature of neurodevelopmental disorders and contributes significantly to visceral discomfort, but the specific etiology is unclear. In particular, it is unknown which cell types within central, autonomic or enteric nervous systems, gut muscle, epithelium or immune system are the principal drivers of gut dysfunction. We report that the Mecp2 mouse model of Rett syndrome, a neurodevelopmental disorder with profound gut dysmotility, exhibits impaired whole gut transit times and constipation in vivo as well as striking alterations to propagating neurogen

From self-narration to a worldview: a phenomenological, narratological, and linguistic case study of a patient with a complex clinical picture of bipolar disorder.

BACKGROUND: Comorbidity of bipolar disorder and aspects of neurodevelopmental disorders present unique challenges and opportunities in understanding the formation and maintenance of selfhood and worldview in psychopathology. Traditional cognitive models often overlook the narrative and phenomenological dimensions of patient experience, particularly how autobiographical narration and emotionally charged worldviews mediate lived experience. OBJECTIVES: This study aims to explore (i) how worldviews are shaped by emotion and memory in psychiatric illness, and (ii) how narrative forms provide exis

Serving Differently: Pioneering the Inclusion of Autism in the Greek Military.

Autism spectrum disorders are typically considered a disqualifying factor for military service due to their high demands on the environment. This case study describes the implementation of a social skills training program for a military candidate with Asperger's syndrome. The intervention employed four strategies: Social Stories, which used structured scenarios to teach appropriate social responses; Social Autopsy, which analyzed real-life interactions to identify strengths and areas for improvement; Role-Playing, which provided guided practice of social interactions; and a Cardboard Game, des

The Gut Microbiota and Autism Spectrum Disorder: Current Research and Therapeutic Insights.

Autism Spectrum Disorder (ASD) is a collective term for neurodevelopmental disorders with core features of social communication impairment, restricted and repetitive behaviors, and narrow interests. These include classic autism, Asperger's syndrome, and pervasive developmental disorder not otherwise specified. ASD is currently managed with behavioral interventions, rehabilitation training, and family support, but there is no curative medication. Recent studies suggest that some patients with ASD may experience gastrointestinal symptoms. Perhaps this is associated with the disturbances of gut m

Essential element and toxic metal dyshomeostasis in childhood autism spectrum disorder: a comprehensive narrative review of correlations, tissue-specific distribution, and multifactorial regulation.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by impaired social communication and repetitive stereotyped behaviors, with a rising global prevalence. Its etiology involves genetic, environmental, and epigenetic interactions, among which essential elements [including trace elements such as zinc (Zn), selenium (Se), copper (Cu), iron (Fe), manganese (Mn) and macrominerals like magnesium (Mg)] and toxic metals (lead (Pb), mercury (Hg), cadmium (Cd)) are key environmental regulators of neurodevelopment. This narrative review systematically summarizes the association

A massively parallel reporter assay of MECP2cis-regulatory elements reveals genetic candidates for male-biased autism.

Autism affects male individuals four times more often than female individuals, yet the basis of this sex bias remains unclear. One hypothesis is that this sex bias is due to genetic variants in X chromosome genes where haploinsufficiency causes neurodevelopmental disorders (NDDs). In heterozygous XX female probands, a complete loss-of-function variant in one of these genes often causes syndromic NDD, but the same variant is too deleterious for survival in hemizygous XY males. A mild hypomorphic variant, however, may be tolerated in a female carrier but may produce milder neurological phenotype

Evolutionary conservation of MECP2 and its relevance to neuronal stability and behavioral adaptation.

MECP2 encodes a methyl-DNA-binding chromatin protein whose dosage is critical for nervous system function: loss-of-function variants cause Rett syndrome, whereas increased dosage causes MECP2 duplication syndrome. Despite extensive research on MECP2 in neurodevelopmental diseases, its unexplored evolutionary history, which we derived from comparative genomic analysis of hundreds of species, provides a novel evolutionary framework for understanding the profound consequences of MECP2 perturbation on brain and behavior. In this review, we analyze the phylogenetic profile of MECP2 and discuss how

Altered toll-like receptor expression in autism spectrum disorder: Evidence from mRNA and serum protein analyses.

Autism Spectrum Disorder (ASD) is a neurodevelopmental condition in which immune dysregulation and neuroinflammatory processes are increasingly implicated. Toll-like receptors (TLRs), as key components of innate immunity, may represent a biological interface between peripheral immune activity and neurobehavioral manifestations; however, comprehensive investigations of the full TLR family in ASD remain scarce. This study aimed to evaluate peripheral blood mRNA expression of all human TLRs (TLR1-TLR10) and corresponding serum protein levels in children and adolescents with ASD. Thirty-one drug-n

Serum levels of FGF-2, VEGF, and EGF and repetitive behaviors in pediatric ASD, OCD, and comorbid groups.

Autism spectrum disorder (ASD) and obsessive-compulsive disorder (OCD) exhibit notable phenomenological overlaps. However, relatively few studies have simultaneously examined both the biological mechanisms and phenomenological overlaps of these two conditions. The aim of this study is to examine serum fibroblast growth factor 2 (FGF-2), vascular endothelial growth factor (VEGF), and epidermal growth factor (EGF) levels and their phenomenological characteristics in patients diagnosed with ASD, OCD, and ASD + OCD. The study sample consisted of 84 children categorized into ASD, OCD, and comorbid

Effectiveness of Parent-mediated Intervention Versus Standard Therapist-based Intervention in Indian Children With Autism Spectrum Disorder: A Randomized Controlled Trial.

OBJECTIVE: The global prevalence of autism spectrum disorder (ASD) has increased substantially in recent decades. In India, prevalence estimates range from 0.74% to 1.68% among children aged 2 to 6 years. Therapist-based interventions are resource-intensive and impractical in low- and middle-income countries. Parent-mediated interventions (PMIs) offer an alternative; however, Indian comparative data are limited. This RCT compared the effectiveness of the Value-based, Quality interaction, Unique for the child, Responsive and Adapted for family (VQURA), a PMI, with standard Therapist-Based Inter

Structures of MeCP2 bound to nucleosomes reveal distinct mechanisms of Rett syndrome mutations.

Methyl-CpG binding protein 2 (MeCP2) is a chromatin-associated regulator essential for neuronal gene regulation, and pathogenic mutations in MeCP2 cause Rett syndrome (RTT). However, the mechanisms governing MeCP2 engagement on chromatin and the effects of RTT mutations on nucleosome interactions remain poorly understood. We determined cryo-EM structures of MeCP2 bound to mono-nucleosomes with or without linker DNA methylation. In the absence of linker DNA methylation, the methyl-CpG binding domain (MBD) engages nucleosomal DNA near superhelical location ±7, whereas a methylated linker CpG red

MeCP2 functions as a DNA methylation reader in immune homeostasis and cancer.

DNA methylation is a key epigenetic mechanism that mediates gene expression and informs cellular identity. Methyl-CpG binding protein 2 (MeCP2) is a protein capable of recognizing and interpreting the methylated DNA code to bring about phenotypic outcomes through co-factor interactions. Altered DNA methylation patterns and MeCP2 function, therefore, disrupt cellular epigenetic homeostasis and contribute to human diseases. MeCP2 loss-of-function or amplification events are responsible for Rett syndrome and MECP2 duplication syndrome, respectively, where patients present with multisystem pathoph
