AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

The cognitive effects of creatine supplementation in older adults and preclinical models of dementia: A scoping review.

BACKGROUND & OBJECTIVES: Dementia represents a growing global health crisis, yet disease-modifying therapies remain limited. Impaired brain energy metabolism, neuroinflammation, and synaptic dysfunction are some shared pathological features across dementia subtypes that may be amenable to metabolic intervention. Creatine supplementation, through its role in the phosphocreatine/ATP buffering system and proposed neuroprotective properties, has attracted growing scientific and public interest as a candidate intervention for dementia. This scoping review maps the existing evidence on creatine supp

Developmental variation in second-to-fourth digit ratio (2D:4D) in children with cerebral palsy: an exploratory morphometric analysis.

OBJECTIVE: Cerebral palsy (CP) is an etiologically heterogeneous neurodevelopmental condition arising from disturbances of the developing brain during prenatal, perinatal, or early postnatal life. The second-to-fourth digit ratio (2D:4D) has been proposed as an imperfect indirect correlate of first-trimester prenatal sex-steroid exposure. This study aimed to compare 2D:4D ratios between children with CP and healthy controls and to examine whether digit ratio was associated with functional and clinical severity in children with CP. METHODS: This case-control, cross-sectional study included 75

Beyond attendance: participation and relational work in planned group recreational activities in a long-term care village setting.

BACKGROUND AND OBJECTIVES: Person-centered care (PCC) has shaped dementia care reforms and informed innovative long-term care models such as care villages. Recreational activities are often seen as a site where PCC is enacted, yet little empirical work has examined how participation in structured activities is enabled, mediated, and constrained through everyday social, relational, and organizational dynamics. This study examines how participation in structured group activities was produced in a care village. RESEARCH DESIGN AND METHODS: An ethnographic study was conducted in Canada's first pu

Physiological classification of Parkinson's disease severity using multimodal speech biomarkers with a hybrid CNN-Mamba framework.

INTRODUCTION: Hypokinetic dysarthria in Parkinson's disease provides an accessible non-invasive biomarker, but multi-class severity grading remains difficult because of overlapping acoustic patterns and limited long-range temporal modeling in existing approaches. METHODS: We developed a hybrid CNN-Mamba framework using multimodal speech features transformed into 2D representations. The model was trained and validated on speaker-disjoint PC-GITA Spanish data and tested on an independent Mandarin clinical cohort, with additional external evaluation on a public Parkinsonian speech corpus. Improving Parkinson's disease diagnosis by non-invasive detection of retinal biomarkers in MPTP monkeys using ERG and pupillometry.

Parkinson's disease (PD) diagnosis is currently made by clinical observation of motor symptoms when around 50% of dopamine neurons in the substantia nigra pars compacta are already lost. Non-motor symptoms, such as vision problems, occur earlier during disease progression and could be caused by altered retinal functioning. Various techniques exist to detect retinal alterations and, therefore, could provide biomarkers for earlier diagnosis of PD. The aim of this project is to determine potential retinal biomarkers for PD by using electroretinography (ERG) and pupillometry. In vivo measurements

Lifting the Veil on Neuroinflammation in Parkinson's Disease: A Bibliometric Analysis.

Parkinson's disease (PD) is a chronic neurodegenerative disorder characterized by gradual progression. A significant pathophysiology of its development and evolution is neuroinflammation. A previous review assessed the neuroinflammation research in PD, but limitations in its search period and analytical methods restricted its ability to reflect the full landscape of the field and recent research trends. To solve this problem, we performed a bibliometric and visual review in order to describe the current state of research and outline the recent progress in PD-related neuroinflammation. The Web

Nanotechnology for efficient delivery of TCM active ingredients in treating brain diseases.

Brain diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), brain tumors, and stroke, remain difficult to treat due to the restrictive nature of the blood-brain barrier (BBB). Traditional Chinese medicine (TCM) bioactive compounds offer multitarget therapeutic potential, but their clinical translation is limited by poor solubility, instability, rapid clearance, and insufficient brain exposure. Nanodelivery systems can enhance pharmacokinetics and enable brain-targeted delivery via engineered nanocarriers that exploit multiple BBB-crossing routes. This review summarizes nanote

Microstimulation of the human dopaminergic midbrain enhances outcome-related happiness and reduces loss seeking.

Midbrain circuits are thought to govern reward-guided behavior and momentary happiness, yet causal evidence in humans remains scarce. Here, we show that focal microstimulation of the substantia nigra pars compacta alters both decision making and momentary happiness in patients undergoing awake deep-brain stimulation surgery for Parkinson's disease. During a gambling task with interleaved momentary happiness ratings, stimulation delivered at outcome reduced risk-seeking in the loss domain without increasing overall gambling probability. Stimulation improved choice efficiency, guiding behavior t

Astrocytic ACSBG1 depletion improves lipid-cytokine signaling and attenuates α-Synuclein pathology in a Parkinson's disease mouse model.

Astrocytes are key regulators of lipid metabolism, and dysregulated astrocytic lipid processing is implicated in Parkinson's disease (PD) pathogenesis. Our prior genome-wide screens identified ACSBG1, an astrocyte-enriched acyl-CoA synthetase, as a candidate regulator of α-synuclein (α-Syn) levels. However, how ACSBG1 links lipid reprogramming to inflammatory astrocyte activation and α-Syn pathology remains unknown. We compared the transcriptomic, cytokine, and lipid secretomes of TNF-α and IL-1α stimulated primary astrocytes from wild-type (WT) and Acsbg1 knockout (KO) mice. In vivo, we cross

An initial machine learning model applied to local field potential data from the subthalamic nucleus to detect freezing of gait in Parkinson's disease.

INTRODUCTION: Freezing of gait (FOG) is an unpredictable and debilitating symptom of Parkinson's disease (PD). Current approaches to treating FOG are lacking; adaptive deep brain stimulation (aDBS) holds promise in treating FOG. However, a necessary precursor to using aDBS is understanding changes in the subthalamic nucleus (STN) prior to and during FOG. The aim of this project was to develop a machine learning model to detect FOG episodes using local field potential (LFP) data from the STN in individuals with PD. METHODS: The LFP data were collected from five individuals with PD using the Me

Toward future diagnostics of Parkinson's disease: a perspective on multimodal motor assessment and personalized digital twins.

The accurate diagnosis of Parkinson's disease (PD) remains challenging due to the heterogeneity of motor symptoms and the reliance on subjective clinical assessments. Although biomarkers such as dopaminergic imaging, fluid assays, and genetic testing can support diagnosis, their clinical utility is limited by cost, accessibility, and low sensitivity to early motor changes. Consequently, diagnosis depends largely on clinical observation, highlighting the need for more accurate, objective, and integrable tools to detect early motor symptoms. Advances in wearable sensing and computational modelin

ALDH1A1-dopaminergic gene co-expression in human substantia nigra: meta-analysis of disease-associated correlation changes across seven independent Parkinson's disease datasets.

BACKGROUND: Parkinson's disease (PD) involves progressive dopaminergic neuron loss in the substantia nigra (SN). Aldehyde dehydrogenase 1A1 (ALDH1A1), the rate-limiting enzyme in retinoic acid biosynthesis, is enriched in vulnerable dopaminergic neuron subpopulations and is consistently downregulated in PD. However, the relationship between ALDH1A1 expression and broader dopaminergic pathway gene co-expression has not been systematically characterized across multiple independent datasets. METHODS: Gene expression correlations were analyzed across seven independent human SN microarray datasets
