Guidelines to Apply for Ethical Approval of Animal Experimentation and Use of Human Samples.

Animal experimentation is a fundamental tool in biomedical research for the study of diseases and the development of interventions such as drugs, vaccines, and probiotics for the treatment of human pathologies. Animal models enable analysis of pathophysiological processes in complex organisms with genetic and functional similarities to humans, which is essential for evaluating the safety and efficacy of new therapeutic interventions and surgical techniques before their clinical application. Among the most commonly used models are mice, rats, and fish, employed in the study of various diseases,

AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

Admission Neurological Status, Radiological Mass Effect, and In-Hospital Mortality Among Surgically Treated Patients with Traumatic Acute Subdural Hematoma: A Single-Center Experience from Türkiye.

BACKGROUND: Traumatic acute subdural hematoma (ASDH) carries high morbidity and mortality despite surgery. We evaluated associations of admission neurological status and radiological mass effect with in-hospital mortality. An exploratory analysis examined a composite unfavorable Glasgow Outcome Scale (GOS) outcome, with in-hospital mortality as its largest component. METHODS: This retrospective study included 82 adults treated surgically for traumatic ASDH between January 2020 and December 2025. The Zumkeller index (midline shift minus hematoma thickness) was categorized as <0, 0-3, or >3 mm.

A Second Pathogenic Protein, PolyGN2C-iso2, Reveals a Dual-Protein Pathology in Neuronal Intranuclear Inclusion Disease.

BACKGROUND: Neuronal intranuclear inclusion disease (NIID) pathogenesis has been strongly linked to uN2CpolyG translated from NOTCH2NLC transcript variant 1. However, emerging evidence suggests that NOTCH2NLC transcript variant 2 may also generate a disease-relevant protein, PolyGN2C-iso2, but isoform-discriminating and antibody-independent evidence remains incomplete. METHODS: We characterize the aggregation propensity of PolyGN2C-iso2 in vitro, develop isoform-discriminating monoclonal antibodies for its detection in patient tissues, and perform targeted proteomic analysis of laser-microdis

Attitudes toward sodium valproate use, withdrawal, and counseling in people of reproductive potential: A scoping review.

Regulatory restrictions and evolving clinical guidelines have aimed to reduce sodium valproate use in people of reproductive potential over the past decade due to its teratogenic risk. While clinical evidence and policy responses are well established, stakeholder perspectives remain less explored. This scoping review synthesizes the existing evidence on the attitudes of patients, caregivers and healthcare professionals regarding sodium valproate use, withdrawal and counseling in females of childbearing age. This scoping review, conducted according to JBI Methodology for a Scoping Review and re

Prototypical Representation Learning for Multi-Site Domain Generalization in Schizophrenia Diagnosis.

Brain functional networks (BFNs) derived from multi-site fMRI data have been widely used in the diagnosis of psychiatric disorders such as schizophrenia (SZ). However, site-induced distribution shifts pose a major challenge to the generalization of classification models based on BFNs. While domain generalization (DG) seeks to address this challenge without access to target domain data, most methods make the idealized assumption that each class exhibits consistent structures across sites. The inherent diversity within each class often violates this assumption, resulting in poor generalization t

Residence-Based Geospatial Analysis of Severe Traumatic Brain Injury in a Regional Neurotrauma System.

OBJECTIVE: Severe traumatic brain injury (sTBI) creates a substantial burden on neurotrauma systems. While geospatial analyses of TBI have focused on injury location, less is known about how patients' residential environments contribute to regional sTBI burden. This study aimed to identify residence-based spatial clustering of sTBI across Long Island and characterize community-level factors associated with excess incidence. METHODS: We conducted a retrospective cohort study of adults (≥18 years) with sTBI (Glasgow Coma Scale ≤8) admitted to a Level 1 trauma center (2010-2021). Patient-reporte

Accelerated Brain Atrophy in Epilepsy: A Prospective Population-Based Study.

OBJECTIVE: Previous longitudinal neuroimaging studies suggest that epilepsy is a progressive disorder. To date, these findings have relied largely on populations from tertiary centers, resulting in ascertainment bias, as severely affected individuals are more likely to be rescanned. We aimed to determine rates of progressive brain atrophy in a prospective, population-based cohort. METHODS: We analyzed longitudinal magnetic resonance imaging (MRI) data from 116 individuals with epilepsy and 89 age- and sex-matched healthy controls recruited from a prospective, longitudinal, community-based coh

Not Just for Kids: A Systematic Review of Outcomes of the Thenar Flap.

Fingertip injuries are common, and the thenar flap is a well-described technique used to maintain digital length. However, its use in patients aged 30 years and above is generally discouraged due to concerns regarding postoperative joint contracture. The purpose of this review was to evaluate whether these concerns are substantiated. A search of PubMed, Embase, and SCOPUS (1947-2025) identified 15 studies involving 519 flaps. Case reports and studies lacking functional outcome data were excluded. Bias was assessed using the Methodological Items for Non-Randomized Studies instrument, and result

"A rough and swirling sea": Voicing Amyotrophic Lateral Sclerosis through discourse analysis.

Amyotrophic Lateral Sclerosis (ALS) is a rare, incurable neurodegenerative disease that leads to death within 5 years of diagnosis and greatly impacts patients' and caregivers' quality of life (QoL). Typically leading to death within 5 years, ALS underscores the need for emotional and psychological support. This study analyzes 118 testimonies from patients (n = 67), informal caregivers (n = 41), and formal caregivers (n = 10), with a gender-balanced sample (n = 67 women), using discourse analysis. Narratives reveal a complex emotional landscape of suffering and resilience, showing ALS as a dis

Tau-induced mitochondrial reverse electron transport drives neurodegeneration.

Hyperphosphorylation and aggregation of tau are pathological hallmarks of tauopathies. Mitochondrial dysfunction is also a common feature of tauopathies. The mechanistic link between tau abnormalities and mitochondrial dysfunction and its relationship to the physiological function of tau, however, is unclear. Here, we demonstrate that tau regulates mitochondrial reverse electron transport (RET), which produces excess reactive oxygen species (ROS), reduces the NAD+/NADH ratio, and is activated by aging or stress. In flies, mice, and human induced pluripotent stem cell (hiPSC)-derived neurons, t

From theory to practice: physicians' knowledge, attitudes, and practices regarding amyotrophic lateral sclerosis in Egypt - a cross-sectional study.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder with heterogeneous presentations that often mimic other conditions, leading to frequent misdiagnosis and delayed recognition. Physician awareness and diagnostic confidence are critical for early detection, yet little is known about these factors in the Middle East and North Africa (MENA) region, where ALS services and specialized centers remain limited. METHODS: We conducted a nationwide cross-sectional survey of 1,320 physicians in Egypt. A validated online questionnaire assessed demographics, knowled
