A longitudinal investigation of locus coeruleus norepinephrine system integrity and cognitive performance across the Alzheimer's spectrum.

Loss of locus coeruleus (LC) integrity, a debilitating feature of Alzheimer's disease (AD), can be visualized in vivo with MRI. Longitudinal investigation across the AD spectrum can elucidate the timing of LC damage and its behavioral correlates. Cognitively normal adults (n = 153), individuals with mild cognitive impairment (n = 73), and with AD (n = 43) underwent MR imaging to assess LC integrity, PET imaging to assess tau and Aβ ([18F]MK6240 and [18F]AZD4694, respectively) and completed a battery of cognitive tests. For a subset of participants, these assessments were repeated annually for

Sequential associations of myelin dysfunction with Aβ and Tau pathology in Alzheimer's disease: a dynamic myelin perturbation.

BACKGROUND: Myelin dysfunction has emerged as a potentially pivotal factor in Alzheimer's disease (AD); however, its temporal dynamics and relationship with hallmark pathological processes remain incompletely understood. This study aimed to investigate dynamic demyelination and its association with the progression of Aβ and Tau pathology in AD. METHODS: A total of 1,237 participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) with 18F-florbetapir (FBP) PET imaging were included. White matter SUVR (WM SUVR), defined as the adjusted FBP signal in white matter, was used as a prox

Andrographolide-loaded PLGA nanoparticles protect retinal ganglion cells against experimental acute glaucomatous injury and attenuate mitochondrial dysfunction and retinal PANoptosis-related changes.

Acute glaucomatous injury is characterized by abrupt intraocular pressure elevation, retinal ischemia/reperfusion-like stress, and rapid retinal ganglion cell (RGC) degeneration, in which oxidative stress, mitochondrial dysfunction, and regulated cell death play important roles. Emerging evidence further implicates PANoptosis in glaucomatous retinal injury. Andrographolide (ADR) possesses antioxidant and neuroprotective activities, but its poor aqueous solubility and low bioavailability limit its therapeutic application. Here, we developed ADR-loaded poly(lactic-co-glycolic acid) nanoparticles

Role of Polyethylene Glycol-Mediated Axonal Fusion in Early Recovery of Median and Ulnar Nerve Injury: A Pilot Double-Blind Randomized Clinical Trial.

BACKGROUND: Up to 40% of peripheral nerve repairs fail to achieve functional recovery. Polyethylene glycol (PEG) attenuates Wallerian degeneration (WD) by facilitating early axonal fusion, promoting improved functional recovery. This study is the first randomized clinical trial assessing PEG-mediated fusion of mixed motor-sensory nerves. STUDY DESIGN: Patients 18 to 75 years of age with Sunderland V upper extremity nerve injuries were randomized into PEG-mediated and standard neurorrhaphy groups. Sphingosine-1-phosphate attenuates seizures and cognitive deficits, associated with reduced hippocampal IL-1β in a PTZ-induced epilepsy model.

BACKGROUND AND AIM: Epilepsy is a prevalent neurological disorder frequently accompanied by cognitive impairments and neuronal damage. Neuroinflammation-particularly mediated by interleukin-1β (IL-1β)-plays a critical role in epileptogenesis. Sphingosine-1-phosphate (S1P), a bioactive lipid mediator, has been implicated in neuroprotection and anti-inflammatory signaling; however, its role in epilepsy remains unclear. This study aimed to investigate the effects and underlying mechanisms of S1P in a pentylenetetrazol (PTZ)-induced mouse model of epilepsy. METHODS: Male C57BL/6 mice were adminis

TEMPOL Enhances Polyethylene Glycol Axon Fusion Following Sciatic Nerve Transection in Adult Rats.

Neurotmesis, the most severe form of peripheral nerve injury, involves complete transection and loss of motor and sensory function. Surgical repair, typically via end-to-end neurorrhaphy (NRR), is often required. A key pathological feature is Wallerian degeneration (WD) in the distal stump, which, along with slow axonal regeneration, leads to muscle atrophy and poor functional recovery. Polyethylene glycol (PEG)-mediated axonal fusion has emerged as a promising strategy to bypass WD by rapidly reconnecting severed axons and restoring conduction. Methylene blue (MB) has previously enhanced PEG-

WTAP mediated m6A methylation modulates retinal photoreceptor function via facilitating of REEP6, PDE6B and RDH12 translation.

Inherited retinal dystrophies (IRDs) are a leading cause of visual impairment and irreversible blindness worldwide, yet their precise molecular and genetic mechanisms remain unclear. N6-1ent. In the mammalian retina, m6A methylation is widely distributed in various cellular layers and is essential for retinal homeostasis. In this study, we assessed the in vivo roles of WTAP, a crucial component of the m6A methyltransferase complex, in the mammalian retina and demonstrated that loss of Wtap led to diminished scotopic ERG responses, progressive retinal degeneration, and significant loss of rod c

Atg7-dependent autophagy is indispensable for mice spiral ganglion neurons.

Atg7-dependent autophagy is critical for the long-term maintenance of the post-mitotic cells, yet its role in spiral ganglion neurons (SGNs) remains incompletely defined. Here, we conditionally ablated Atg7 in SGNs by generating Bhlhe22Cre/+Atg7flox/flox mice and examined cellular p62 aggregates, ultrastructure, and auditory function at postnatal day (P)30 and P60. At P30, Atg7 was markedly reduced in SGNs. Immunofluorescence revealed progressive enlargement of p62-positive condensates from nano-scale at P30 to micron-scale by P60. Excess p62 activated the antioxidant transcription factor Nrf2

Chronic inhalation exposure to 0.5 ppm of 2-ethyl-1-hexanol induces histopathological changes in olfactory, respiratory, and skin tissue of mice.

OBJECTIVES: 2-Ethyl-1-hexanol (2EH) is a volatile organic compound that can cause sick building syndrome and can be released from flooring materials for more than 10 years through the hydrolysis of compounds with 2-ethylhexyl moieties. Previous studies using mice have raised concerns about tissue-damaging effects on the olfactory epithelium (OE), olfactory bulb (OB), brain, lungs, and skin. A subchronic mouse study observed degeneration of OE at concentrations as low as 20 ppm. However, the effects of long-term exposure to 2EH remain unclear. This study aimed to elucidate the histopathological

Decreased Length of Locus Coeruleus Norepinephrine Axons and Increased Amyloid Beta Pathology in Male APP/PS1 Mice During Protracted Abstinence From Alcohol.

Alzheimer’s disease (AD) is the leading cause of dementia and evidence suggests that alcohol, the most commonly used addictive substance, may increase AD risk. Locus coeruleus (LC) neurons are the primary source of norepinephrine in the brain and these neurons degenerate early in AD. In rodent models, lesioning the LC increases amyloid beta (Aβ) pathology suggesting that LC integrity and norepinephrine signaling obstruct Aβ pathogenesis. We recently reported a decrease in the number of LC norepinephrine neurons and increased Aβ pathology when measured after protracted abstinence from chronic i

Comparison of Optic Nerve Head Parameters in Alcoholic Liver Disease Patients With Age-Matched Controls.

Alcoholic liver disease (ALD) is associated with systemic neurodegeneration mediated by oxidative stress, mitochondrial dysfunction, and chronic inflammation. As the retina and optic nerve head represent extensions of the central nervous system, they may provide a non-invasive window for detecting alcohol-related neuroretinal damage. This retrospective case-control study compared optic nerve head and peripapillary retinal nerve fiber layer (pRNFL) parameters between 100 patients with ALD and 100 age- and sex-matched healthy controls who underwent comprehensive ophthalmic evaluation and spectra

Maternal Choline Supplementation in a Mouse Model of Down Syndrome and Alzheimer's Disease Generates Unique Expression Profile Mosaics Within Three Hippocampal Excitatory Neuronal Populations.

Individuals with Down syndrome (DS) are at risk for early-onset Alzheimer's disease (AD), marked by neurodegeneration in hippocampal and basal forebrain circuits. Early-life interventions offer therapeutic potential, including maternal choline supplementation (MCS). MCS improves cognitive outcomes and neuroplasticity in rodent models of neurodevelopmental and neurodegenerative disorders, yet cell-type specific molecular effects remain unknown. We investigated the effect of MCS upon the onset of septohippocampal degeneration at 6 months of age in the Ts65Dn mouse model of DS/AD. Using laser cap

Functional and Histological Outcomes After Polyethylene Glycol Fusion of Rat Sciatic Nerve Transection.

PURPOSE: Polyethylene glycol (PEG) fusion has been reported to restore axonal continuity after nerve repair if initiated before Wallerian degeneration (WD). Sterile α and Toll/Interleukin-1 Receptor motif-containing 1 (SARM1) inhibition can potentially delay WD. We hypothesized that PEG fusion in SARM1 knockout (KO) rats would allow for improved strength and nerve regeneration following nerve transection. METHODS: A total of 32 wild-type (WT) and 32 SARM1-KO rats underwent sciatic nerve transection, followed by standard neurorrhaphy or PEG fusion. Baseline compound nerve action potentials (CN

Dual protective role of curcumin- encapsulated chitosan nanoparticles against gastric and neural injury in a rat model of gut-brain axis dysfunction: a histological and biochemical study.

Gastric ulcer (GU) and Alzheimer's disease (AD) are prevalent age-associated disorders frequently accompanied by systemic oxidative stress and inflammation. Emerging evidence suggests that gastrointestinal dysfunction and inflammatory signaling may aggravate neurodegenerative processes. Curcumin (Cur) exhibits well-established antioxidant, anti-inflammatory, neuroprotective, and gastroprotective properties; however, its therapeutic utility is limited by poor bioavailability. The present study aimed to formulate and characterize Curcumin- encapsulated chitosan nanoparticles (Cur-CSNPs) and eval

Impact of tension-free autograft repair on early sensory recovery in polyethylene glycol (PEG)-mediated axonal fusion following traumatic peripheral nerve injuries.

Autograft repairs remain inferior to primary neurorrhaphy, largely due to axonal regeneration across two coaptation sites. PEG-mediated neurorrhaphy has been shown to enhance recovery following peripheral nerve injury by mitigating Wallerian degeneration. Whether PEG can overcome these limitations in human autografts remains unclear. We therefore compared sensory recovery following PEG-mediated primary versus PEG-mediated autograft neurorrhaphy. In this study, prospectively collected clinical data were analyzed for patients undergoing PEG-mediated nerve repair with follow-up to 100 days. Senso

PEG-fusion of viable sciatic nerve isografts restores axonal structure and behavioral recovery after segmental-loss sciatic nerve injuries in Lewis rats.

Segmental-loss peripheral nerve injuries (SL-PNIs) often produce severe deficits in sensory/motor functions and voluntary behaviors. The current gold standard for repair is neurorrhaphy of a cable autograft that (1) produces donor site morbidity; (2) results in rapid Wallerian degeneration (WD) of severed distal nerve segments; (3) relies on slow (1-2 mm/day) axonal regeneration; (4) does not prevent atrophy of denervated muscles and sensory structures; and (5) results in poor to non-existent recovery of sensory/motor functions and voluntary behaviors, especially with longer segmental-loss gap

Hybrid dextran/fibronectin nanogels for brain-targeted mitophagy inducer delivery to alleviate neuroinflammation and neuronal loss of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuron loss, chronic neuroinflammation, and α-synuclein aggregation. Blood-brain barrier (BBB)-penetrable, dual-target nanomedicines for microglial inflammation and neuronal degeneration remain challenging. In this study, we fabricated a brain-targeted, pH- and reactive oxygen species (ROS)-responsive nanogel (NG) platform using dextran (Dex) as the main polysaccharide backbone, crosslinked with inflammation-targeting fibronectin (FN), and loaded with neuroprotective quercetin (Que). Protective effects of gastrodin against bisphenol A-induced dopaminergic dysregulation and cognitive impairment in rats.

Gastrodin (GAS) is a potent neuroprotective compound extracted from the traditional Chinese medicinal herb Gastrodia elata Blume. However, its role in mitigating bisphenol A (BPA)-induced dopaminergic dysfunction and cognitive impairment remains insufficiently explored. Many studies have shown that BPA exposure causes neurodegeneration via mechanisms involving dopaminergic system dysfunction, oxidative stress, and neuroinflammation. Therefore, the present study aimed to investigate whether GAS mitigates the effects of BPA-induced cognitive impairment through neuroinflammation in a rat model. W
