The virulence regulator ToxT influences Vibrio cholerae motility through the expression of the methyl-accepting chemoreceptors AcfB and TcpI.

Vibrio cholerae is the causative agent of the human diarrheal disease cholera. Previous studies identified "hyperswarming" (HS) mutant V. cholerae with enhanced motility and decreased virulence factor expression, but the genetic basis of HS was unknown. Through whole-genome sequencing, we have identified point mutations within toxT, which encodes the master virulence regulator, as being responsible for the HS phenotype. These mutations are loss of function, and they eliminate transcription of cholera toxin and toxin-coregulated pilus genes. Using single-cell video tracking, these HS strains we

Antibody-recruiting molecules based on mimotope fusion peptides and a dimeric antibiotic.

New therapeutic approaches are needed to treat infections due to multidrug resistant (MDR) Gram-negative bacteria. The use of antibody-recruiting molecules (ARMs) is a promising approach to address the MDR problem given their potential to engage both the adaptive and innate immune systems to treat bacterial infections. Here we present two new strategies to create ARMs. First, we show that a 9-mer antimicrobial peptide (K6) can be fused to a 11-mer mimotope peptide recognized by antibodies targeting the Vibrio cholerae O1 O-antigen to produce an ARM peptide (FZ5) that can render E. coli more se

Avian cholera causes mass mortality at penguin mega-colonies amid H5N1 HPAI outbreaks in Antarctica.

The characterization of pathogens affecting Antarctic wildlife has been historically limited and sporadic. With the introduction of H5N1 high pathogenicity avian influenza virus (HPAIV) in Antarctica in the austral summer 2023/2024, concerns about the health of Antarctic seabirds and mammals prompted immediate surveillance efforts. In March 2024, we visited the Danger Islands archipelago, that hosts two mega-colonies of Adélie penguins, and observed an unusual mortality estimated in thousands of Adélie penguins and other species. Swabs and tissues were collected for molecular detection of infe

The Estuary Effect: Variations in Temperature and Salinity Alter msh Promoter Activity in Vibrio cholerae.

UNLABELLED: Vibrio cholerae , the facultative pathogen underlying cholera, naturally inhabits warm aquatic estuaries. Environmental persistence is enhanced by the ability of V. cholerae to colonize host reservoirs and form multicellular biofilms, causing seasonally endemic outbreaks in many tropical regions. Most toxigenic strains utilize the type IVa mannose-sensitive hemagglutinin (MSHA) pilus for host reservoir colonization and biofilm formation. Temperature and salinity can alter V. cholerae biofilm formation, yet their impact on MSHA production specifically remains largely unknown. Here,

The future of cholera genomics through the lens of clinical, public health, and research practitioners in Africa.

Cholera remains a persistent public health threat, complicating efforts to track transmission across borders and undermining control measures. Surveillance systems often remain fragmented and reactive, and many aspects of Vibrio cholerae evolution continue to be challenging, restricting the development of effective, data-driven interventions. Although genomics is increasingly transforming our understanding of cholera by revealing new insights into pathogen biology, transmission dynamics, and evolution, its routine integration into public health practice remains low. In this Personal View, we p

Knowledge and preventive practices associated with anticipatory action for climate-related cholera outbreaks in Mozambique: a gender perspective.

Background: Cholera remains a major public health concern in sub-Saharan Africa, exacerbated by extreme climatic events and inadequate WASH-related practices. As both climate change impacts and WASH practices are gender-sensitive, this study adopts a gendered perspective to examine the relationship between anticipatory action (AA) for climate-induced cholera outbreaks and knowledge, attitudes and practices (KAP) in Zambézia Province, Mozambique. Methods: A cross-sectional survey was conducted in Quelimane (urban) and Mopeia (rural) using a multistage sampling design. A total of 448 respondents

Cholera Epidemics in Nineteenth-Century Poznań (1831-1873).

The article presents the history of cholera epidemics in Poznań to explain how, and with what results, the Prussian authorities used medical knowledge and practices to control the non-German population of the annexed territories of Poland in 1831-1866. The study proves the problematic nature of attributing susceptibility to cholera to Poles as a backward and inferior race, which the Prussian authorities did to justify their racist policy towards Poles. The Berlin government's instrumental treatment of the Jewish population in the conquered Polish lands is also explored. The study emphasizes th

Vibrio repeats (VXRs) play a role in horizontal dissemination of sedentary chromosomal integron-associated antibiotic resistance genes across Vibrio species.

Sedentary chromosomal integrons (SCIs) are large integrons that harbor extensive arrays of gene cassettes and constitute major reservoirs of bacterial genetic diversity. In Vibrio, SCI cassettes are flanked by species-associated attC repeats, here referred to as Vibrio species repeats (VXRs). Although SCIs have been proposed as ancestral sources of antibiotic resistance genes (ARGs), the extent to which SCI-associated ARGs are exchanged among Vibrio species remains poorly understood. To investigate the dynamics of SCI-associated ARGs, we analyzed 37,812 genomes representing 181 Vibrio species.

A Single Amino Acid in the A2 Linker of Cholera Toxin Affects the Efficiency of Toxin Disassembly and Resulting Toxicity.

Cholera toxin (CT) from Vibrio cholerae and heat-labile enterotoxin (LT) from enterotoxigenic Escherichia coli are highly related AB5-type protein toxins that contain a catalytic A1 subunit, an A2 linker, and a cell-binding homopentamer of B subunits. The two proteins bind the same GM1 receptor and have similar levels of enzymatic activity toward their shared Gsα target, yet CT is more potent than LT. This difference has been mapped to a sequence of 11 amino acids (residues 226-236) near the C-terminus of the A2 linker. We now identify a single amino acid, aspartic acid at position 229, as a k

Conflict-Related WASH System Failure and Public Health Preparedness in the Middle East: A Policy-Oriented Narrative Review.

Conflicts in the Middle East have intensified public health emergencies through progressive failure of water, sanitation, and hygiene (WASH) systems. This policy-oriented narrative review synthesizes peer-reviewed and humanitarian evidence published between January 2020 and March 2026 on WASH system failure and related public health consequences in Gaza and the occupied Palestinian territory, Yemen, Syria, Lebanon, and Iraq. Across these settings, four recurring mechanisms emerged: physical damage; loss of electricity, fuel, chemicals, and spare parts; restrictions on access and workforce safe

Subtypes of Bladder-Innervating Sensory Neurons revealed by Immuno-Fluorescence In Situ Hybridization.

Sensory neurons that innervate the urinary bladder are critical for triggering voiding and mediating pain perception under pathological conditions. Despite functional and immunohistochemical evidence indicating the existence of several bladder afferent populations, the subtypes of dorsal root ganglion (DRG) neurons that innervate this organ are poorly defined and specific markers to study them have not been described. Here, we combined retrograde tracing with the cholera toxin β subunit and immunofluorescence in situ hybridization with markers defined by transcriptomic analysis of C and Aδ aff

Genomic characterization of CTX-M-15-producing Klebsiella pneumoniae ST709-KL9 from neonatal bacteraemia in eastern India.

BACKGROUND: Klebsiella pneumoniae, a WHO-designated critical-priority pathogen, is a predominant cause of neonatal bacteraemia in India, associated with high morbidity and mortality. The global emergence and spread of high-risk clones, combined with convergence of antimicrobial resistance (AMR) and virulence traits, underscores the need for close genomic surveillance. This study presents AMR and virulence traits of K. pneumoniae ST709-KL9 strain isolated from neonatal blood in West Bengal, India. METHODS AND RESULTS: The blood isolate from a 10-day-old neonate (November 2021) underwent identi

Somalia is battling concurrent outbreaks of cholera, measles, and diphtheria driven by severe drought and displacement.

Somalia is experiencing concurrent cholera, measles, and diphtheria outbreaks driven by climate change, conflict, displacement, malnutrition, inadequate water and sanitation, and fragile health systems. This letter argues that these overlapping epidemics should be recognized as a syndemic rather than independent outbreaks because they share common structural determinants that amplify transmission and mortality. We advocate for integrated surveillance, combined vaccination strategies, strengthened laboratory capacity, nutrition support, climate-informed early warning systems, resilient primary

Developing affordable monoclonal antibodies for LMICs through high performance manufacturing processes and LMIC based commercial manufacturing.

Monoclonal Antibodies (mAbs) are one of the most effective developments in disease prevention and treatment of the last century. Active treatment and passive immunization with mAbs targeting key antigens represents a promising approach to preventing and mitigating infectious pathogen-caused illness, particularly in Low and Middle-Income Countries (LMICs). While the development of mAbs against infectious disease has lagged behind that of immune disorders and oncology targets, recent efforts against infectious diseases have seen increasing focus with the development of mAbs against RSV, Ebola st

The impact of very early and early enteral nutrition on intestinal permeability in children with pancreatitis: A randomized controlled study.

INTRODUCTION AND OBJECTIVES: One of the pathophysiological mechanisms of acute pancreatitis (AP) is the reduced integrity of the intestinal mucosa. Enteral nutrition supports the gut barrier, but its effect on intestinal permeability in pediatric AP has not yet been evaluated. The aim of this study was to assess how very early enteral nutrition (within 24hours of admission) and early enteral nutrition (after the first 24hours) affect plasma zonulin levels in children with mild-to-moderate AP. METHODS: We conducted a prospective randomized study between January 2019 and September 2022 in three

Climate-informed surveillance for climate-sensitive infectious diseases in Somalia: early warning systems, policy gaps, and preparedness priorities.

Climate variability is reshaping infectious disease risk through ecological, environmental, social, and health-system pathways. However, its operational implications remain insufficiently defined in fragile and conflict-affected settings. Somalia is a critical case because recurrent droughts and floods intersect with displacement, food insecurity, weak water and sanitation infrastructure, disrupted immunisation, livestock dependence, and constrained health-system capacity. This narrative review synthesises evidence on climate-sensitive infectious disease risks in Somalia, examines existing sur

Unravelling Cholera's Footprints in India: A Data-Driven Investigation of its Burden, Risk, and Strain Variability.

INTRODUCTION: Binge Eating Disorder (BED) has high lifetime prevalence rates, low treatment success rates, and high rates of treatment dissatisfaction, early discontinuation of care, and recurrence. Complementary and integrative health (CIH) interventions (non-mainstream practices used with conventional approaches for whole-person treatment) hold potential to overcome many treatment barriers and improve BED treatment outcomes. Some CIH interventions have empirical support for use in eating disorders. However, little is known about the current state of CIH use in BED. METHODS: This mixed-metho

Renal manifestations of immune checkpoint inhibitors in the pediatric population.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have revolutionized cancer treatment and have become an essential part of therapy, but their use is associated with immune-related adverse events (irAE). Specifically, nephrotoxicity is well documented in adult populations but data regarding irAEs are limited in pediatric populations. This review examines the renal manifestations of ICIs and relevant clinical measures and treatments. MATERIALS AND METHODS: A comprehensive review of existing literature was conducted to assess the incidence, pathophysiology, and management of ICI-associated renal

Lysosomal metabolism in pancreatic cancer and its crosstalk with host physiology.

Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine
