Roles of systemic CD4+ T cells and local mast cells in a porcine model of eosinophilic esophagitis.

Large-animal models are valuable for investigating the pathogenesis, natural history and therapeutic responses of eosinophilic esophagitis (EoE). We previously developed a porcine EoE model using hen egg white protein (HEWP) sensitization and oral challenge, but additional immunophenotyping is needed - particularly of esophageal mast cells (MC) and systemic CD4 T-cell responses. This study evaluated whether the model induces MC infiltration into the esophagus and whether systemic allergen-specific CD4 T cells correlate with esophageal eosinophilia. Pigs underwent three weekly intraperitoneal s

Real-time analytical insights for disease surveillance and response during the severe drought and food security crisis, Somalia 2022-2023.

BACKGROUND: Decades of conflict, epidemics, and climatic shocks have severely weakened Somalia's health system. The failure of five consecutive rainy seasons in 2022-2023 led to the longest and most severe drought in recent history, resulting in an unprecedented nutrition and food security crisis. These conditions have heightened the risk of disease epidemics, particularly cholera and measles. To assess the effect of interventions on disease transmission in real time, make short-term projections of disease incidence, and inform response efforts, the World Health Organization Somalia Country Of

Modelling the impact of climate on cholera: a case study of Kolkata.

Cholera is highly climate sensitive, however previous attempts to model its future under climate change have been limited to statistical analyses. Mechanistic models are an essential addition because they permit a deeper understanding of the complex feedback loops involved in infectious disease transmission, allowing for better modelling of potential scenarios such as interventions or changes in pathogen dynamics. We compare four mathematical models with differing assumptions of climate sensitivity and fit them to a cholera dataset from Kolkata, India using MCMC. We then use bias-corrected cli

Antimicrobial susceptibility and virulence profile of Vibrio cholerae from Delhi, India: effect of floods on epidemiology.

BACKGROUND: Cholera remains endemic in India, with outbreaks frequently intensifying during the monsoon due to flooding and disruption of water and sanitation systems. Recent floods in Delhi have raised concerns regarding changing epidemiology, antimicrobial resistance (AMR) and circulation of virulent Vibrio cholerae lineages. METHODS: This was a retrospective analysis of V. cholerae isolates obtained from stool samples at a tertiary care hospital in Delhi from May 2018 to December 2024. Isolates were identified by conventional methods and serotyping. Antimicrobial susceptibility testing was

Nighttime lights as a proxy for conflict intensity and infrastructure recovery in Yemen and Ukraine.

INTRODUCTION: Quantifying the impacts of armed conflict on civilians and infrastructure remains a major challenge, particularly where reporting is limited. Most conflict measurement tools require affected populations to report events and are limited by short time series, under-reporting, and varying methods. These tools do not capture infrastructural rebuilding, which has important health implications. Given this, we demonstrate the utility of nighttime lights (NTL) as a complementary tool for measuring conflict dynamics and infrastructure recovery with an epidemiological application. METHODS

Relative contributions of public and domestic transmission domains in cholera outbreaks in displacement camps: an exploratory agent-based modeling study.

Cholera is associated with devastating outbreaks among forcibly displaced people. Insights into the relative contributions of the public (extra-household) and domestic (intra-household) domains to cholera spread in camps, as well as the circumstances under which each may drive transmission, can support the design of response strategies. However, these have yet to be systematically investigated. We developed an agent-based model of cholera transmission in camps informed by a rapid appraisal conducted in Northeast Nigeria, an expert consultation, and humanitarian minimum standards. We simulated

Ordinary Differential Equation Modeling and a Novel Zubair-Fréchet Distribution for Cholera in Ghana With Control Strategies.

BACKGROUND AND AIMS: In this paper, we present a valuable and novel interdisciplinary study that applies a classic epidemiological model, Susceptible-Infected-Recovered (SIR) to capture the mean disease dynamics, with a novel Zubair Fréchet (ZF) distribution to model the extreme, heavy-tailed nature of cholera outbreak sizes. Furthermore, the proposed model is linked to both public health and actuarial risk. We demonstrate that the ZF provides a superior fit to cholera case data compared to traditional distributions. METHODS: Data was collected from both primary sources (such as the Ghana Hea

Exploring the effective strategies for allocating limited resources to minimize cholera outbreaks.

Despite the near-complete elimination of cholera in many developing countries, numerous lower-income countries continue to face recurring epidemics. Although extensive research has been conducted on cholera transmission dynamics and control, a comprehensive approach to managing outbreaks in resource-limited settings remains elusive. This study introduces a cholera epidemic model incorporating a resource allocation strategy to balance efforts between reducing transmission and enhancing recovery rates. The model is validated using weekly cholera data from the resurgence in Haiti (October 2022-Ma

Engineered Coiled-Coils Convert Cholera Toxin B-Pentamers into Programmable Membrane Fusogens.

Membrane fusion is central to biological function and bioengineering, yet few design rules exist that enable proteins to be programmed to drive fusion at defined membrane interfaces. Here, we show that cholera toxin B-subunit (CTB), a naturally occurring glycolipid-binding pentamer, can be re-engineered into a programmable membrane fusogen by assembling two CTB units through rationally designed coiled-coil linkers attached to the CTA2 peptide that threads through the CTB pentamer. Using discrete parallel and antiparallel coiled-coil architectures, we generated CTB dimers with defined orientati

Quantifying relative health impact across Gavi, the Vaccine Alliance's portfolio in 117 countries at the subregional level: a modelling study.

BACKGROUND: Estimates of vaccine impact have typically been used to quantify the effects of, and inform, immunisation strategies. Given the growing resource constraints on health systems worldwide, robust estimates of vaccine impact that allow comparison across different vaccines are now more crucial for decision making than ever. Building on previous modelling studies, we aimed to estimate vaccine impact ratios for an expanded portfolio of Gavi, the Vaccine Alliance-supported vaccination programmes against 14 vaccine-preventable diseases across 117 low-income and middle-income countries using

Epidemiological characteristics and outcomes of the 2023-2024 cholera outbreak in South Yemen: a retrospective descriptive study from MSF-supported centres.

BACKGROUND: The cholera burden in Yemen is the highest in the world, with the country facing recurrent outbreaks. It is worsened by the ongoing conflict, limited access to clean water, and the deteriorated sanitation and health systems. In November 2023, a cholera outbreak started in Shabwah governorate and seemed to be controlled in January 2024. However, in March 2024, cholera spread westward, causing 40,000 suspected cases in South Yemen. METHODS: Routinely collected data were used to provide descriptive statistics of the 2023-2024 cholera outbreak of patients managed in the Médecins Sans

Pasteurella multocida causes liver pyroptosis in broilers through the MAPK-NLRP3-GSDMD signaling pathway.

Avian cholera is a bacterial disease caused by infection with Pasteurella multocida (P. multocida), which seriously threatens the poultry industry. Liver injury is a key factor in the acute death of infected chickens. This study sought to clarify how P. multocida causes liver injury in broilers, specifically examining liver pyroptosis and the involvement of oxidative stress and inflammation. Experiments with 28-day-old broilers infected with P. multocida showed that post-infection serum indicators were significantly higher for ALT, AST, and T-Bil, but lower for ALB. Histopathological evaluatio
