DRP1 lysine 616 lactylation exacerbates cerebral ischemia-reperfusion injury by activating the STING inflammatory pathway.

Ischemia-hypoxia-induced inflammation and glycolysis are linked to the severity of cerebral ischemia-reperfusion injury (CIRI), but the mechanisms are unclear. Current research suggests that the inflammatory response of immune cells activated by STING is a key regulatory molecule in cellular inflammatory damage. However, the specific mechanisms underlying STING-mediated CIRI inflammatory responses remain unclear. This study found that STING expression was specifically elevated in microglia in the damaged side of the hippocampus in CIRI model mice, and this elevation was positively correlated w

EXPRESS: Chronic Intermittent Hypoxia Induces Persistent Venous Narrowing of Downstream Parenchymal Microvessels and Pial Veins in the Male Mouse Cortex.

Chronic intermittent hypoxia (IH), a hallmark of sleep apnea, can lead to cerebral hypoperfusion and cognitive decline, though its precise impact on cerebral microcirculation remains unclear. This study investigated the longitudinal effects of 2-week chronic IH (8% O2, 6.5-min cycle for 6 hours/day) on brain microcirculation in the male mouse cortex. We observed a significant reduction in the diameters of pial veins (from 55 to 46 μm) and parenchymal venules (from 9.9 to 8.2 μm) after chronic IH relative to pre-IH conditions, while upstream arteries (47 to 44 μm) and arterioles (12.4 to 12.0 μ

Brain-derived neurotrophic factor in solid tumor angiogenesis and neurovascular-immune interactions (Review).

Brain-derived neurotrophic factor (BDNF), a prototypical neurotrophin, together with its high-affinity receptor tropomyosin receptor kinase B (TrkB), regulates neuronal survival, synaptic plasticity and neurovascular unit homeostasis. Beyond the nervous system, BDNF and TrkB are aberrantly expressed in multiple solid tumors and have been implicated in proliferation, invasion, epithelial-mesenchymal transition, perineural invasion and resistance to therapy. Experimental evidence demonstrates direct vascular activity in selected settings: BDNF/TrkB promotes endothelial survival, migration and tu

Precision Oxygen Modulation in the Reperfusion Era: Conditioning and Therapeutic Strategies in Ischemic Stroke.

Endovascular thrombectomy (EVT) and advanced perfusion imaging have transformed acute ischemic stroke (AIS) treatment, yet tissue hypoxia may persist because of delayed presentation, incomplete reperfusion, microvascular no-reflow, and secondary injury. Oxygen is an essential substrate for cerebral metabolism and a modifiable therapeutic exposure; however, clinical studies of oxygen-based interventions have yielded inconsistent results because of heterogeneity in timing, dose, delivery, and patient selection. Here we propose precision oxygen modulation (POM) as a stage-, phenotype-, and tissue

Prematurity insults remodel cerebellar development and behaviour.

Preterm survivors often develop motor and socio-cognitive impairments that implicate altered cerebellar development, yet the underlying mechanisms remain poorly understood. A key challenge is that prematurity involves overlapping perinatal insults that converge on the developing brain, making their individual effects difficult to disentangle. Here, we model two major prematurity-associated insults, maternal immune activation (MIA) and neonatal hypoxia (Hx), in mice. By controlling timing and sequence, we define how these insults shape cerebellar assembly during rapid maturation. Comparative an

Itaconate protects the blood-brain barrier by preserving microvascular bioenergetics after traumatic brain injury.

Traumatic brain injury (TBI) is a leading cause of mortality and long-term disability, yet effective pharmacological therapies remain unavailable. Early blood-brain barrier (BBB) disruption accelerates neuroinflammation and secondary brain injury. It has been well recognized that brain microvascular endothelial cells (BMECs) depend on mitochondrial oxidative metabolism to maintain BBB integrity. Here, we investigated the role of the mitochondria-derived immunometabolite itaconate in regulating cerebrovascular bioenergetics and BBB integrity after TBI. Using a controlled cortical impact model,

Modulation of Postictal Hypoxia: Sex- and Route-Specific Effects of Acute and Chronic Cannabidiol and Delta9-tetrahydrocannabinol.

Postictal hypoxia causes several consequences that are detrimental to the quality of life of people with epilepsy. Cannabidiol (CBD) and Δ9-tetrahydrocannabinol (THC), the primary phytocannabinoids from cannabis, have been shown to exhibit vasoactive and antioxidant properties that may mitigate the local oxygen deficiency observed after seizures. Here, we focus on the effects of CBD and THC on seizure-induced hypoxia. Adult Long-Evans rats of both sexes were assigned to drug-specific cohorts and the effects on local hippocampal oxygen were measured before and after seizure elicitation followin

Sulforaphane alleviates DSS-induced colitis and gut-brain axis dysfunction potentially via microbiota-metabolism-HIF-1α crosstalk.

Ulcerative colitis (UC) is a chronic inflammatory disorder of the gastrointestinal tract, often accompanied by anxiety- and depression-like symptoms, which are closely associated with gut microbiota dysbiosis, metabolic disturbances, immune activation, and dysfunction of the gut-brain axis. Sulforaphane (SFN), a natural isothiocyanate, has been demonstrated in our previous studies to alleviate colitis in mice; however, its effects on UC-associated neurobehavioral abnormalities remain unclear. In this study, a dextran sulfate sodium (DSS)-induced colitis model was employed to investigate the ef

Antagonizing central GABAA and adenosine receptors reverses the autoresuscitation defects caused by reduced brainstem serotonin.

Sudden Infant Death Syndrome (SIDS) remains a leading cause of infant death, and its incidence is increasing. SIDS infants harbor numerous defects within the brainstem serotonin (5-hydroxytryptamine, 5-HT) system. There is also evidence that in hypoxic conditions SIDS infants do not successfully autoresuscitate, the integrated response to severe hypoxia necessary to reperfuse the brain, restore eupnea, and promote arousal from hypoxic coma. In animals, a loss of 5-HT or 5-HT2A receptor activity compromises autoresuscitation. Caffeine, via undefined mechanisms, rescues the autoresuscitation of

Combined PET imaging of tumor hypoxia and proliferation in Glioblastoma using a single injection of 18F-FMISO and 18F-FLT: Association with sites of subsequent recurrence.

BACKGROUND: Glioblastoma (GBM) is characterized by intra-tumoral heterogeneity, rapid proliferation, hypoxia-driven radio-resistance, and early recurrence. Gadolinium-Enhanced MRI, the standard of care imaging modality for surgical and radiotherapy planning, lacks specificity for viable tumor burden, particularly in non-enhancing regions. PURPOSE: This pilot study aimed to evaluate whether preoperative dual-tracer PET using a combined dose of 18FMISO (hypoxia) and 18FLT (proliferation) improves tumor delineation and predicts subsequent local recurrence compared with MRI. METHODS: Seven patie

Mitochondrial electron transport chain activity is not essential for the acute hypoxic response of spinal oxygen sensors.

Spinal sympathetic preganglionic neurons (SSPNs) are oxygen sensitive. These spinal oxygen sensors (SOS) increase sympathetic output when subjected to acute hypoxia, orchestrating cardiorespiratory responses to protect against the deleterious effects of oxygen deprivation. Most other oxygen-sensing systems rely on mitochondrial mechanisms to detect and signal decreases in oxygen. Here, we apply pharmacological blockers of the mitochondrial respiratory chain and a mitochondrial-targeted antioxidant to a rat perfused in situ spinal cord preparation to test the role of mitochondria in the SOS. We

Renin-Aldosterone Activation and Enhanced Urinary Potassium Excretion in Asphyxiated Newborns Treated with Therapeutic Hypothermia: A Prospective Study.

Hypoxic-ischemic encephalopathy (HIE) is a critical neonatal condition for which therapeutic hypothermia (TH) remains the cornerstone of neuroprotective intervention. Clinical observations and prior studies note that electrolyte imbalances-particularly hyponatremia and hypokalemia-frequently emerge during TH. Therefore, this study explores how activation of the renin-angiotensin-aldosterone system (RAAS) may contribute to sodium (Na) and potassium (K) disturbances in asphyxiated neonates receiving TH. In this prospective study, 38 newborns with HIE treated with TH and 21 healthy term newborns

Capillary stalling after traumatic brain injury: an emerging microcirculatory contributor to secondary brain injury.

Secondary brain injury after traumatic brain injury (TBI) has traditionally been understood and managed through the lens of intracranial pressure, cerebral perfusion pressure, and global cerebral blood flow. Yet a substantial proportion of patients continue to exhibit persistent tissue hypoxia and metabolic crisis even when these macrocirculatory parameters appear adequately controlled. Capillary stalling-the transient or sustained interruption of red blood cell transit within individual or focal capillary networks-may represent an under-recognized microcirculatory disorder that conventional m

Altitude-induced plasticity of respiratory central-peripheral chemoreflex interaction.

Stable breathing depends on the integrated action of central and peripheral chemoreceptors, but the nature of this interaction in humans and its physiological importance remain controversial. Previous studies reported additive, hypo-additive and hyper-additive responses across species, preparations and protocols. Here, we used a novel method to quantify peripheral chemoreflex sensitivity across graded levels of central chemoreceptor activation to test whether central-peripheral chemoreceptor interaction in awake humans is fixed or state dependent. At low altitude, the form of interaction varie

Cloning and Expression of CRH Genes in Epinephelus coioides and Their Application in Stress Resistance Evaluation.

Corticotropin-releasing hormone (CRH) is a hypothalamic regulatory peptide that modulates the hypothalamic-pituitary-interrenal (HPI) axis and mediates stress responses in fish. Fluctuations in environmental factors such as temperature, ammonia nitrogen, and dissolved oxygen are among the most common triggers of fish stress, which can disrupt homeostasis, increase disease susceptibility and mortality, and thereby constrain the sustainable development of grouper aquaculture. To identify more suitable stress tolerance indicators for timely assessment of fish stress status in grouper farming, thi

Comparison of Long-Term Outcomes of Combined Hypothermia and Whole Body Cooling Therapy in Newborns with Hypoxic-Ischemic Encephalopathy.

Therapeutic hypothermia is widely used as a neuroprotective intervention in infants with hypoxic-ischemic encephalopathy (HIE); however, limited data exist regarding the long-term neurodevelopmental outcomes of different cooling strategies. The aim of this study is to compare the long-term neurodevelopmental and cognitive outcomes of combined hypothermia (CH) and whole-body cooling (WBC) treatments in children diagnosed with HIE with each other and with a healthy control group without an HIE diagnosis. This retrospective study included a total of 50 children diagnosed with HIE and treated with

Convergent and Distinct Transcriptional Signatures in Suicide, Bipolar Disorder, and Major Depressive Disorder.

Globally, a person dies by suicide every 40 seconds, underscoring a major global public health concern. In the United States, suicide is the second leading cause of death among individuals aged 10-34 years. Suicide is a complex, multifactorial phenomenon influenced by genetic and epigenetic factors and is strongly associated with mood disorders, particularly major depressive disorder (MDD) and bipolar disorder (BD). However, the extent to which their molecular signatures converge or diverge across disorders and brain regions remains incompletely understood. To address this gap, we conducted a

Investigating the Role of Tetranectin in Ischaemic Heart Disease and Heart Failure.

BACKGROUND: Cardiac diseases represent 32% of deaths globally1. Ischaemic heart disease (IHD) is a leading cause of heart failure (HF). Progressive fibrosis in IHD contributes to the dysfunction in HF2. Investigation of novel proteins with prior association to extracellular remodelling provide invaluable information behind the underlying pathology of cardiac disease. Tetranectin is a plasma protein previously linked to cardiac remodelling. Recent literature highlighted its potential to compete against N-Terminal pro Brain Natriuretic Peptide as a diagnostic biomarker for HF3. This study aimed

[Highlands as a natural neuroprotection laboratory: hypoxia and neurodegenerative processes].

High altitude hypobaric hypoxia presents a unique stressor to the central nervous system, exerting a profound yet contradictory effect on the dopaminergic system. On one hand, uncontrolled hypoxia triggers pathological cascades characterized by a significant neurotoxic release of dopamine, increased oxidative stress, and the initiation of neurodegenerative processes akin to those observed in Parkinson's and Alzheimer's diseases. On the other hand, controlled moderate hypoxia, referred to as hypoxic preconditioning, along with the genetic adaptations found in highland natives, stimulates powerf

Beyond M1 M2: single-cell insights into how glioblastoma reshapes tumour-associated macrophages.

Glioblastoma (GBM) is an immunologically cold tumour in which glioma-associated microglia and macrophages constitute the predominant immune cell population, comprising brain-resident tumour-associated microglia (Mg) and infiltrating monocyte-derived (Mo) tumour-associated macrophages (TAMs). Recent single-cell and spatial transcriptomic studies have fundamentally revised the traditional M1/M2 activation paradigm, revealing a dynamic continuum of niche-dependent myeloid states rather than discrete polarisation phenotypes. In this review, we discuss how tumour-derived signals reshape Mg and Mo-T
