Honokiol Alleviates Dry Eye-Related Corneal Epithelial Damage by Activating SIRT3/PINK1-Mediated Mitophagy.

BACKGROUND: Dry eye is a common ocular surface disease characterized by corneal epithelial damage, yet effective therapies targeting mitochondrial dysfunction remain limited. PURPOSE: To investigate the protective effects of honokiol (HKL) against dry eye-related corneal epithelial injury and elucidate the underlying mechanisms focusing on SIRT3 and PINK1-mediated mitophagy. METHODS: A benzalkonium chloride (BAC)-induced dry eye mouse model (n=6) was established and treated with HKL eye drops. Corneal damage was assessed by slit-lamp photography, Schirmer I test, and H&E staining. SIRT3, PIN

Acquired Distichiasis in Dermatomyositis: A Possible Delayed Sequela of Chronic Periocular Inflammation.

Acquired distichiasis is an uncommon eyelid disorder characterized by an accessory row of eyelashes arising from the meibomian gland orifices, most often in the setting of chronic inflammatory or cicatricial disease. We report a case of a 65-year-old woman with longstanding dermatomyositis, recurrent periocular inflammation, and episodes of blepharitis who developed bilateral upper-eyelid distichiasis. Review of clinical photographs from 2020 and from a severe periocular flare in 2023 showed no clearly visible accessory row. At follow-up in January 2026, clinical and dermoscopic examination de

Sjögren Disease-B Cells at the Brink: From Autoimmunity to Lymphomagenesis and the Rise of Novel B Cell-Targeted Therapies.

Sjögren disease (SjD) is a common systemic autoimmune disorder characterized by inflammation of the exocrine glands, resulting in dryness. Patients frequently exhibit extraglandular manifestations affecting various organ systems. To date, there are no US Food and Drug Administration (FDA)-approved disease-modifying therapies for SjD. In this review, we explore the expanding field of SjD endotyping as a tool to enhance patient stratification, prognostication, and clinical decision-making. SjD endotypes driven by heightened B cell activity are linked to increased lymphoma risk. B cells play a ce

Exercise, Medicine and Icing (EMI) for the Knee: Small Instalments, Big Gains in Early Osteoarthritis.

Adenine di-Phosphate (ADP) is an important physiological agonist that plays a vital role in normal hemostasis and thrombosis. The importance of ADP in normal hemostasis is clearly demonstrated in patients suffering from storage pool disease who show excessive bleeding tendencies. It is well established that ADP activates platelets through 3 purinergic receptors, namely P2Y1, P2Y12 and P2X1. The P2Y1 receptor is a Galpha q coupled G-protein receptor that is important for platelet shape change, aggregation, thromboxane A2 generation, procoagulant activity, adhesion to immobilized fibrinogen and

TLR7 responses to nucleosides drive sialadenitis in Slc29a3-deficient mice.

Autoimmune sialadenitis is a hallmark of IgG4-related disease (IgG4-RD) and Sjögren syndrome. The single-stranded RNA sensor TLR7 has been shown as a driver of sialadenitis. Although TLR7 is activated by ssRNA degradation products such as nucleosides and oligoribonucleotides, the role of these ligands in sialadenitis development remains unclear. Here, we demonstrate that lysosomal accumulation of endogenous nucleosides is sufficient to drive autoimmune sialadenitis. Loss-of-function genetic variations in the nucleoside transporter SLC29A3 cause lysosomal nucleoside accumulation, leading to con

IL-36 mediates immune activation in Sjögren's disease and may represent a novel biomarker of disease.

Sjögren's disease (SjD) is a systemic autoimmune disease. The underlying disease mechanisms remain poorly understood, and there are no curative therapies. MyD88-mediated signaling is essential for SjD, although the pathways that rely on MyD88 are not well characterized. Our objective was to determine if MyD88-dependent IL-1 cytokines mediate inflammation in SjD. Using a SjD mouse model and patient samples, RNA sequencing was performed on salivary tissue and peripheral B cells. Splenocytes from SjD mice were stimulated with IL-36 cytokines and B cell activation was assessed. Finally, ELISAs wer

Ocular Matrix Metalloproteinases in Spaceflight.

INTRODUCTION: Spaceflight presents unique challenges to ocular health, as visual disturbances such as dry eye symptoms have been frequently reported by astronauts. Matrix metalloproteinases (MMPs), a family of extracellular matrix-degrading enzymes, are known mediators of ocular surface inflammation and tissue remodeling on Earth, particularly in dry eye disease. This review explores the potential role of MMPs in spaceflight-associated ocular conditions, including spaceflight-associated dry eye syndrome and spaceflight-associated neuro-ocular syndrome. METHODS: We searched literature on topic

Longitudinal Immune Profiling of T Cell Exhaustion During IL-17A Blockade in a Patient With HLA-B27-negative Spondyloarthritis and Sjögren's Syndrome: A Case Report.

BACKGROUND/AIM: Coexisting Sjögren's syndrome (SS) and human leukocyte antigen-B27 (HLA-B27)-negative ankylosing spondylitis (AS) is rare and therapeutically challenging. In patients with prior Stevens-Johnson syndrome (SJS), non-steroidal anti-inflammatory drugs (NSAIDs) are contraindicated and tumor necrosis factor (TNF) inhibitors may be insufficient. Interleukin-17A (IL-17A) blockade with secukinumab offers an alternative, though its long-term effects on T cell exhaustion and regulation remain unclear. This report examines immune exhaustion and regulatory dynamics during IL-17A inhibition

Neonatal Thymic Dynamics Influence Autoimmune Pathology by Shaping the Suppressive Potential of Regulatory T Cells.

Neonatal thymectomy (TX) has been known to induce experimental autoimmune disease models in mice for over half a century. The thymic microenvironment, including thymic epithelial cells (TECs), plays a crucial role in establishing self-tolerance in T cells. However, the extent to which the dynamic changes in the neonatal thymic environment contribute to the onset of autoimmunity remains incompletely understood. In this study, the detailed alterations in the neonatal thymus and peripheral lymphoid organs were analyzed using a mouse model of primary Sjögren disease. Mice treated with TX at 3 days
