Recent evidence has shown that the cyclic GMP-AMP synthase (cGAS)/stimulator of interferon genes (STING) signaling pathway mediated by nuclear factor E2-related factor 2 (Nrf2) contributes to the pathogenesis of doxorubicin (DOX)-induced cardiotoxicity. Ling-Gui-Zhu-Gan decoction (LGZGD) ameliorated DOX-cardiotoxicity in cardiomyocytes in our previous studies. In this study, the therapeutic effects of LGZGD that protect against DOX-induced cardiotoxicity in C57 male mice and HL-1 cells, and the underlying mechanisms were investigated. LGZGD-L (2.15 g/kg), LGZGD-M (4.29 g/kg), and LGZGD-H (8.58

INTRODUCTION: Heterozygous UCHL1 variants have recently been associated with an autosomal dominant neurodegenerative disease characterized by spastic ataxia, optic atrophy and neuropathy.
METHODS: We describe two individuals from a single family who presented with optic atrophy and progressive vision loss, without demonstrable spasticity, ataxia or peripheral neuropathy.
RESULTS: Genetic testing revealed a novel pathogenic UCHL1 variant accounting for the two individuals' phenotype.
DISCUSSION: Our findings highlight the significant phenotypic variability related to heterozygous UCHL1-relat

Wolfram syndrome is a rare autosomal recessive disorder caused by pathogenic variants in the WFS1 gene, characterized by early-onset diabetes mellitus, optic atrophy, sensorineural hearing loss, arginine vasopressin deficiency, and progressive neurodegeneration. The condition selectively affects pancreatic β cells and neurons via chronic endoplasmic reticulum (ER) stress, and no proven disease-modifying therapy currently exists. Diabetes mellitus is typically the first manifestation, presenting at a mean age of 6 years as an insulin-dependent phenotype with preserved C-peptide and negative dia

Growth differentiation factor 15 (GDF15) is a cytokine induced in several tissues in response to stress. GDF15 suppresses food intake and increases energy expenditure via its actions on the glial-derived neurotrophic factor receptor α family-like specific receptor (GFRAL), located in the hindbrain. We recently showed that selective deletion of the mitochondrial fusion protein optic atrophy 1 (OPA1) in brown adipocytes (OPA1 BKO) leads to GDF15 secretion, partially mediating resistance to diet-induced obesity (DIO), and improving thermoregulation. To investigate whether GDF15 signaling through

A 29-year-old man presented with a headache and right-sided visual deficit. His neurological examination was normal except for bilateral optic atrophy. CEMRI revealed a uniformly contrast-enhancing multilobulated lesion at the sellar and suprasellar region. Prolactin levels were high, with the value of 12,800 ng/mL (normal range 3-5 ng/mL) according to hormonal analysis. The pituitary adenoma was removed transsphenoidally from the patient. Histological examination revealed an adenoma with spheroid amyloid deposits adjacent to prolactin-staining adenoma cells. The patient recovered from the sur

Autosomal dominant optic atrophy (ADOA) is an inherited optic neuropathy primarily caused by mutations in OPA1. We identified and defined a spontaneous nonhuman primate (NHP) model of ADOA using rhesus macaques heterozygous for a missense mutation (OPA1A8S). With ocular examinations, ophthalmic imaging, electroretinography, histopathology, immunohistochemistry, and transmission electron microscopy (TEM), we documented retinal nerve fiber layer (RNFL) thinning, retinal ganglion cell (RGC) loss and dysfunction, OPA1 mislocalization, and reduced axonal mitochondrial density in affected macaques.

INTRODUCTION: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have demonstrated significant weight-reducing effects and may offer benefits in idiopathic intracranial hypertension (IIH); however, recent concerns about the risk of non-arteritic anterior ischemic optic neuropathy (NAION) have emerged. Hence, this study was designed to examine the relationship between GLP-1 RA use and optic outcomes in IIH patients.
METHODS: This study used the TriNetX Global Collaborative Network to perform a retrospective propensity score-matched cohort study. It involved GLP-1 RA exposure status to class

BACKGROUND: Autoimmune nodopathy (AN) associated with anti-neurofascin-155 (NF155) antibodies is a distinct disorder characterized by treatment-resistant peripheral neuropathy. Although central nervous system (CNS) involvement is theorized due to the presence of NF155 in oligodendrocytes, definitive clinical reports remain limited.
CASE PRESENTATION: A 31-year-old male presented with a seven-year history of relapsing-remitting progressive sensorimotor neuropathy, tremor, and sensory ataxia. He was initially diagnosed with chronic inflammatory demyelinating polyneuropathy (CIDP) and demonstrat

BORCS5 encodes a subunit of the BLOC-One-Related Complex (BORC), which is known to promote anterograde movement and fusion of lysosomes. We identified 16 individuals from 9 families with bi-allelic BORCS5 variants, revealing a spectrum of neurodevelopmental and neurodegenerative phenotypes. Carriers of homozygous protein-truncating variants (PTVs), resulting in complete loss of BORCS5, presented with prenatally lethal arthrogryposis multiplex congenita, brain malformations, and neuropathological evidence of neuroaxonal dystrophy. Individuals with missense or splice-site variants presented diff

Demyelinating optic neuritis (DON) is an autoimmune inflammatory demyelinating disease that affects the optic nerve. In recent years, with the rapid advancement of biotechnology, novel targeted biological agents have demonstrated unique advantages in the treatment of DON, and the number of approved targeted biologics for clinical use has gradually increased. However, the risks of infections and adverse effects caused by the long-term use of biologics cannot be ignored. How to use various biologics reasonably, effectively, and safely presents a new challenge that clinical physicians must addres

BACKGROUND: Mitochondrial structural abnormalities and impaired energy metabolism are recognized hallmarks of diabetic cardiomyopathy (DCM). GULP1 (PTB-domain engulfment adapter protein 1, also known as CED-6) is a cytoplasmic adaptor protein containing a phosphotyrosine-binding domain. GULP1 has been implicated in metabolic disorders, particularly in type 2 diabetes. However, its potential involvement in cardiovascular homeostasis remains unclear. This study aimed to investigate whether GULP1 attenuates DCM by preserving mitochondrial architecture and bioenergetic function, and to explore the

Leber hereditary optic neuropathy (LHON) is a mitochondrial optic neuropathy that typically causes severe bilateral central vision loss. Although several natural-history studies have reported visual outcomes, long-term trajectories beyond 5 years remain incompletely quantified, particularly in Asian cohorts. We performed a single-center retrospective cohort study at a tertiary eye hospital in Japan. Among 174 genetically confirmed LHON patients, we identified 27 patients (53 eyes) who presented within 6 months of onset and were followed continuously for ≥10 years. Best-corrected visual acuity

Cerebellar ataxia, mental retardation, and disequilibrium syndrome (CAMRQ)-related disorders are rare, nonprogressive, autosomal recessive conditions primarily characterized by cerebellar ataxia, hypotonia, intellectual disability, delayed ambulation, and, in some cases, quadrupedal locomotion. Pathogenic variants in four disease genes, VLDLR, CA8, WRD81, and ATP8A2, have been linked to these disorders, with cases reported across various ethnic groups and geographic regions. However, no reports of CAMRQ1 (OMIM #224050) have been previously made from Africa. In this study, we report the first A

Hereditary optic neuropathies are characterized by bilateral visual loss due to the degeneration of retinal ganglion cells, resulting in optic nerve degeneration and atrophy. Although the genetic origin of the main isolated and syndromic hereditary optic neuropathies has been characterized, the clinical phenotypes exhibit significant and poorly understood variability in both penetrance and expressivity. Additionally, the genetic and environmental factors that influence the onset of these optic neuropathies remain poorly understood, with limited biomarkers to predict disease progression or as r

Hereditary optic neuropathies comprise a genetically heterogeneous group of disorders caused by pathogenic variants in mitochondrial and nuclear genes. Despite increasing diagnostic yields, many patients remain without a molecular diagnosis. We report a novel candidate heterozygous variant in the PHB1 (Prohibitin 1) gene in a large family affected by autosomal dominant optic atrophy. A three-generation family with slowly progressive visual acuity loss due to optic neuropathy and an apparent autosomal dominant pattern was clinically characterized and recruited for genetic counseling. BACKGROUND: The current mainstays of treatment for type 1 retinopathy of prematurity (ROP) are laser therapy and anti-VEGF intravitreal injections (IVI) or a combination of the two. Data comparing treatment outcomes between these modalities are heterogenous and limited by short-term follow-up. There is limited data on visual acuity after treatment. The purpose of this study was to investigate visual acuity, structural outcomes, and refractive error (RE) between primary laser and primary IVI in a contemporary cohort treated for ROP.
METHODS: The medical records of patients treated for type 1 R
