Blood treatment of Lyme borreliae demonstrates the mechanism of CspZ-mediated complement evasion to promote systemic infection in vertebrate hosts.

Lyme disease, caused by the spirochete Borrelia burgdorferi, is the most common vector-borne disease in the United States and Europe. The spirochetes are transmitted from mammalian and avian reservoir hosts to humans via ticks. Following tick bites, spirochetes colonize the host skin and then disseminate haematogenously to various organs, a process that requires this pathogen to evade host complement, an innate immune defence system. CspZ, a spirochete surface protein, facilitates resistance to complement-mediated killing in vitro by binding to the complement regulator, factor H (FH). Low expr

Antigenic variation in the Lyme spirochete: detailed functional assessment of recombinational switching at vlsE in the JD1 strain of Borrelia burgdorferi.

Borrelia burgdorferi is a causative agent of Lyme disease and establishes long-term infection in mammalian hosts. Persistence is promoted by the VlsE antigenic variation system, which generates combinatorial diversity of VlsE through unidirectional, segmental gene conversion from an array of silent cassettes. Here we explore the variants generated by the vls system of strain JD1, which has divergent sequence and structural elements from the type strain B31, the only B. burgdorferi strain in which recombinational switching at vlsE has been studied in detail. We first completed the sequencing of

Mapping tweets to a known disease epidemiology; a case study of Lyme disease in the United Kingdom and Republic of Ireland.

BACKGROUND: Analysis of social media is an emerging method with potential as a tool for disease surveillance. Twitter may offer a route for surveillance by using tweeting habits as a proxy for disease incidence. Previous work has focused on temporal patterns and have proven to be successful. However, the identification of geographical patterns has been limited by a combination of Twitter's data collection policies and by exploring diseases that have a high prevalence and high levels of awareness with the public. We propose that, by performing a restricted geographical search strategy on a dise

The economic burden of Lyme disease and the cost-effectiveness of Lyme disease interventions: A scoping review.

INTRODUCTION: While Lyme disease (LD) is mostly treatable, misdiagnosed or untreated LD can result in debilitating sequelae and excessive healthcare usage. The objective of this review was to characterize the body of literature on the economic burden of Lyme disease (LD) and the cost-effectiveness of LD interventions, such as antibiotic treatment and vaccination. METHODS: We followed Joanna Briggs Institute scoping review methodologies. We systematically searched terms related to LD, economic evaluations, costs, and cost-effectiveness in Medline, Embase, PsycInfo, Cochrane Library, and the gr

Different Competence as a Lyme Borreliosis Causative Agent Reservoir Found in Two Thrush Species: The Blackbird (Turdus merula) and the Song Thrush (Turdus philomelos).

Passerine birds are significantly involved in the dissemination of Borreliella spp. bacteria (formerly Borrelia burgdorferi sensu lato complex), the causative agent of most common and important tick-spread disease Lyme borreliosis. Among several dozen investigated passerine bird species, thrushes (Turdidae) have been reported as a relatively good pathogen reservoirs and disseminators. The principal aim of the study was to identify the differences in Borreliella spp. reservoir competence between two widespread and showing similar behavior thrush species. A total of 157 Ixodes ricinus ticks (19

Blood Smears Have Poor Sensitivity for Confirming Borrelia miyamotoi Disease.

Borrelia miyamotoi disease (BMD) is a newly recognized borreliosis that is cotransmitted by ticks wherever Lyme disease is zoonotic. Unlike Borrelia burgdorferisensu lato, the agent of Lyme disease, B. miyamotoi is closely related to relapsing fever spirochetes, such as Borrelia hermsii Some authors have suggested that the disease caused by B. miyamotoi should be considered a hard-tick-transmitted relapsing fever, and thus, the main mode of confirming a diagnosis for that infection, microscopy to analyze a blood smear, may have clinical utility. To determine whether blood smears may detect B.

A luminescence-based assay for evaluating bactericidal antibody to Borrelia burgdorferi in vaccinated horses' serum.

BACKGROUND: Current serological tests cannot discriminate between bactericidal Borrelia burgdorferi antibodies from others that are merely a response to Borrelia antigenic stimulation. OBJECTIVE: To develop a sensitive and convenient luminescence-based serum bactericidal assay (L-SBA) to identify serum borreliacidal activity. STUDY DESIGN: Prospective validation study and method comparison. METHODS: Serum samples were obtained either from archives of the Animal Health Diagnostic Center at Cornell University (N = 7) or from a vaccination trial (N = 238). Lyme disease: Insight from social sciences.

This article is a selective literature review of social science works published on Lyme disease that draws on other articles published on similar health hazards. These works present Lyme borreliosis as an "archetypal" example of modern infectious risks. It is an "invisible" risk resulting from interactions between human activities, ecosystems, and pathogens. To tackle this risk, health authorities promote individual-based prevention measures. Perceptions of the general population should thus be better understood: different from the perceptions of experts, the general population's perceptions a

Potential effects of blood meal host on bacterial community composition in Ixodes scapularis nymphs.

Tick microbiomes may play an important role in pathogen transmission. However, the drivers of microbiome variation are poorly understood, and this limitation has impeded mechanistic understanding of the functions of microbial communities for pathogen acquisition. The goal of this research was to characterize the role of the blood meal host in structuring the microbiome of Ixodes scapularis, the primary vector of Lyme disease in the eastern United States, and to determine if ticks that fed from different host species harbor distinct bacterial communities. We performed high-throughput 16S rDNA a

Neuroborreliosis in a horse with common variable immunodeficiency.

Common variable immunodeficiency (CVID) is a rare condition in adult horses characterized by hypogammaglobulinemia and increased susceptibility to parasitic and bacterial infections, including recurrent respiratory diseases, septicemia, and meningitis. Lyme disease is often included as a differential diagnosis in CVID horses with signs of meningitis; however, the Borrelia burgdorferi organism has not been demonstrated previously within central nervous system tissues of CVID horses with neurologic disease, to our knowledge. We report herein a case of neuroborreliosis in a CVID horse, confirmed

Outer surface protein polymorphisms linked to host-spirochete association in Lyme borreliae.

Lyme borreliosis is caused by multiple species of the spirochete bacteria Borrelia burgdorferi sensu lato. The spirochetes are transmitted by ticks to vertebrate hosts, including small- and medium-sized mammals, birds, reptiles, and humans. Strain-to-strain variation in host-specific infectivity has been documented, but the molecular basis that drives this differentiation is still unclear. Spirochetes possess the ability to evade host immune responses and colonize host tissues to establish infection in vertebrate hosts. In turn, hosts have developed distinct levels of immune responses when inv

Lyme Disease Emergence after Invasion of the Blacklegged Tick, Ixodes scapularis, Ontario, Canada, 2010-2016.

Analysis of surveillance data for 2010-2016 in eastern Ontario, Canada, demonstrates the rapid northward spread of Ixodes scapularis ticks and Borrelia burgdorferi, followed by increasing human Lyme disease incidence. Most spread occurred during 2011-2013. Continued monitoring is essential to identify emerging risk areas in this region.

Limitations of diagnostic tests for bacterial infections.

Lyme disease diagnosis is currently based on serology - an indirect diagnostic method - as laboratory cultures are fastidious. The only direct diagnostic method that can be useful with some specimens (cutaneous biopsies or aspiration fluid) is PCR. We aimed to detail the main limitations of serology and PCR testing in the diagnosis of bacterial infections. Limitations are supported by examples from the recent history of microbiology. The main limitation of bacterial serology is the presence of numerous cross-reactions due to many genes that are common to various bacterial species. Some serolog

Mitral Valve Endocarditis: A Rare Manifestation of Lyme Disease.

Valvular involvement in Lyme disease is rare. Confirmation of Borrelia species as the causative agent with polymerase chain reaction was done in a few cases in Europe and the United States. We describe a case of mitral regurgitation with a preoperative diagnosis of myxomatous mitral valve degeneration. During surgery, the surgeon suspected infective rather than degenerative etiology; the results of tissue cultures were negative. However, universal bacterial polymerase chain reaction on explanted valve tissue detected Borrelia burgdorferi DNA. If a surgeon suspects infective endocarditis at the

Prevalence of Borrelia burgdorferi, Anaplasma spp., Ehrlichia spp. and Dirofilaria immitis in Canadian dogs, 2008 to 2015: a repeat cross-sectional study.

BACKGROUND: Vector-borne pathogens are emerging concerns in multiple regions of Canada. Determining regional prevalence of canine vector-borne pathogens and documenting change will improve clinician awareness, enable targeted prevention, enhance diagnosis and ideally reduce the risk of disease. Study objectives were to: (i) estimate the prevalence of positive canine vector-borne test results from samples submitted in Canada; (ii) assess change in prevalence over time, from baseline (2008) to 2015; and (iii) estimate the prevalence of pathogen co-infections. METHODS: This repeat cross-sectiona
