Monitoring Chlamydia trachomatis Development in Polarized Endocervical Epithelial Cells.

Understanding Chlamydia trachomatis infection requires physiologically relevant models tailored to various research questions. One of the main sites of C. trachomatis infection is the columnar epithelium of the endocervix; therefore, a model that accurately mimics the environment of endocervical epithelium is essential. Polarized cell models provide a valuable solution, offering a cost-effective yet biologically relevant alternative to conventional 2D culture systems and more complex and costly 3D or animal models. Immortalized human endocervical epithelial cells (A2EN cells) form a polarized

Chlamydia trachomatis Infection in a Microphysiologic Cell Culture Model.

Chlamydia trachomatis is an obligate intracellular bacterium that infects the columnar epithelium of the human endocervix. While conventional two-dimensional cell cultures and animal models have been instrumental in advancing our understanding of C. trachomatis biology, they are limited in capturing the multicellularity, architecture, and physiological microenvironment of the human cervix. This chapter describes the use of a three-dimensional (3D) microphysiologic model to study Chlamydia trachomatis infection. The model is inexpensively made without specialized equipment and is designed to re

A Conditional Knockout Mutant Approach to Study Essential Genes in Chlamydia in Cell Culture and In Vivo.

Identification and characterization of essential Chlamydia-specific factors are vital for the generation of effective therapeutics and vaccines. Despite delays in the development of genetic tools for Chlamydia, significant advancements in Chlamydia genetic manipulation have been made over the last decade and a half, now allowing for the targeted interruption or deletion of chromosomal genes. However, because of the obligate intracellular nature of the bacteria and its unique biphasic developmental cycle, it remains challenging to inactivate essential genes and study the function of their encod

Chlamydia trachomatis co-infection and cervical cancer risk in HPV-positive women: a cross-sectional and histological case-control study.

OBJECTIVES: To characterise Chlamydia trachomatis (CT)-HPV co-infection and test whether CT positivity is associated with cervical cancer in HPV-positive women. METHODS: We conducted a two-component retrospective study at two southern Chinese tertiary hospitals (January 2022-June 2025). Component 1 comprised 21,205 women tested for CT and HPV. Component 2 comprised 100 HPV-positive invasive cervical cancers and 100 HPV-positive non-neoplastic controls, with CT detected by pgp3-PCR on tissue. RESULTS: CT positivity was 3.36%, HPV 21.27%, co-infection 1.29%. CT was associated with HPV positivi

Chlamydia and gonorrhoea prevalence in abortion and prenatal care: a cross-sectional study in a Portuguese centre.

BACKGROUND AND OBJECTIVE: Infections caused by C. trachomatis (CT) and Neisseria gonorrhoeae (NG) are increasing in Europe. However, differences in surveillance systems limit accurate epidemiological data on representative populations, such as women receiving prenatal care. This study aimed to estimate the prevalence of urogenital CT and NG among pregnant women in two distinct pregnancy-related care contexts at a Portuguese center. METHODS: A cross-sectional prevalence study was conducted at a Portuguese tertiary care center, including pregnant women undergoing induced abortion (up to 10 week

Maternal genitourinary tract infections and spontaneous preterm labor: A narrative review of pathogens, pathways, and prevention.

Preterm birth is the leading cause of death among children aged under 5 years of age worldwide. Maternal genitourinary tract infections (GUTIs) are among the few modifiable risk factors for spontaneous preterm labor (sPTL), yet the gap between association and effective intervention remains unresolved. This narrative review consolidates recent evidence on GUTIs and sPTL, integrating pathogens, ascending and hematogenous pathways, and prevention. MEDLINE, Embase, the Cochrane Library, and Scopus were searched (January 2000-January 2026); evidence was synthesized narratively and stratified by pre

Prevalence and behavioral factors associated with penile and rectal HPV infection among men who have sex with men in Kenya.

BACKGROUND: High-risk human papillomavirus (hrHPV) is a leading cause of anogenital cancers and disproportionately affects men who have sex with men (MSM), particularly in low- and middle-income countries. Few studies in sub-Saharan Africa have concurrently examined penile and rectal hrHPV infection and their site-specific behavioral and biological correlates. We assessed the prevalence, genotype distribution, and behavioral factors associated with penile and rectal hrHPV among MSM in Kisumu, Kenya. METHODS: We conducted a cross-sectional analysis of baseline data from MSM aged 18-35 years. C

Syphilis Screening in Pregnancy During Emergency Department Gonorrhea and Chlamydia Testing.

INTRODUCTION: Congenital syphilis is rising in the United States, especially among infants born to women with limited prenatal care. Emergency departments (ED) offer opportunities for early detection, particularly among patients evaluated for urogenital sexually transmitted infection (STI) concerns. In October 2019, our health system implemented an ED STI order set that promoted HIV co-testing and included a preselected option for syphilis testing. In this study, we evaluated change in syphilis testing during the post-implementation period among ED encounters with both pregnancy and gonorrhea/

Pathogen-targeted quick multiplex PCR identification of Chlamydia psittaci and Haemophilus influenzae coinfection in pneumonia: a case report.

This study describes a rare case of pneumonia induced by coinfection with Chlamydia psittaci (C. psittaci) and Haemophilus influenzae (H. influenzae). The patient had a history of chronic nephritis and long-term pigeon breeding. The initial manifestation was slurred speech, followed by rapid progression to high fever and respiratory failure. Chest computed tomography revealed bilateral interstitial pneumonia, predominantly in the right lung. Pathogen-targeted quick multiplex polymerase chain reaction (pt-qPCR) confirmed the pathogens as C. psittaci and H. influenzae, and metagenomic next-gener

PROBIT TIME-TO-EVENT REGRESSION FOR MISCLASSIFIED GROUP TESTING DATA.

Group testing has been used extensively to reduce screening costs in epidemiological studies involving low-prevalence diseases. This testing strategy involves combining specimens (e.g., blood, urine, swabs, etc.) from several individuals to form a pool and then testing the pooled specimen for infection. When the endpoint of interest is a time-to-event outcome, for example, the time until infection or disease, and pools are measured only once, the resulting data are called group-tested current status data (Petito and Jewell, 2016). In this paper, we propose a new type of regression analysis for
