Characterization of bacterial microbiome and molecular detection of rickettsiosis in free-living ticks (Amblyomma sculptum Berlese, 1888).

Amblyomma sculptum is a major tick species in southeastern Brazil and an important vector in the epidemiology of Brazilian Spotted Fever (BSF). This study characterized the bacterial microbiome of free-living A. sculptum ticks in a BSF-endemic area, focusing on differences among developmental stages and sexes, and investigated rickettsial agents using 16S rRNA gene (V3-V4) sequencing. A total of 154 ticks were collected and analysed as 13 pooled samples grouped by stage and sex. Sequencing identified a diverse bacterial community of 180 genera, dominated by Sphingomonas, Nocardioides, Actinomy

Apathy in the elderly: an atypical manifestation of spotted fever rickettsioses.

BACKGROUND: Apathy denotes a lack of motivation, enthusiasm, feelings, or concern. Decreased daily functions and cognitive impairment are other features. In the elderly, these manifestations are overlooked and attributed to ageing or associated comorbidities. Spotted fever group rickettsial (SFG) infections are endemic in the Central Hills of Sri Lanka. We observed occult SFG infection as a treatable cause of apathy in the elderly. METHODS: We studied patients with apathy admitted to the Teaching Hospital, Peradeniya (THP), Sri Lanka, during 2018 & 2019. The diagnosis was confirmed by indirect

Active Surveillance for Heartland virus in North Carolina: Clinical and Genomic Epidemiology.

BACKGROUND: Heartland virus (HRTV) is an emerging tick-borne virus capable of causing severe illness and death. The burden of disease is likely underestimated due to limited seroprevalence studies, lack of commercially available diagnostic tests, and an overlapping clinical syndrome with more commonly diagnosed bacterial diseases such as spotted fever group rickettsiosis or ehrlichiosis. METHODS: Active surveillance for Heartland virus disease was conducted at a large academic center from March to September 2024. Enrolled subjects included those who had testing sent for Ehrlichia polymerase c

Ecologic and Sociodemographic Factors Associated with Seroprevalence of Rickettsia in Yucatan, Mexico.

BACKGROUND: Rickettsioses disproportionately affect vulnerable populations and are frequently misdiagnosed as other febrile illnesses in Yucatan, the Mexican state with the greatest diversity of Rickettsia spp. Although significant seroprevalence has been reported in rural communities, the last population-based study was conducted over two decades ago, despite environmental and social changes that have likely increased transmission risk. This study aimed to estimate the seroprevalence of spotted fever group (SFG) and typhus group (TG) of Rickettsia in an endemic area of southeastern Mexico. M

Effects of exposure to Rickettsia bellii on reservoir competence of guinea pigs for Rickettsia rickettsii.

BACKGROUND: Amblyomma sculptum is the main vector implicated in the transmission of Rickettsia rickettsii in southeastern Brazil, where capybaras are known to be the main amplifying hosts of this pathogen. Interestingly, many areas inhabited by large populations of capybaras and A. sculptum remain free of R. rickettsii, suggesting that other drivers may be involved in the pathogen perpetuation. A. sculptum frequently occurs in sympatry with Amblyomma dubitatum ticks, sharing the same hosts with them. Considering that A. dubitatum ticks are frequently infected with Rickettsia bellii, a possible

Subclinical Cardiac Disturbances After Rickettsia spp. Infection in an Endemic Region of Mexico.

Background: Rickettsial diseases are endemic in southeastern Mexico, yet their potential subclinical cardiac effects remain poorly understood. Although severe spotted fever and typhus group infections may cause myocarditis and arrhythmias, limited evidence exists regarding cardiac alterations in individuals previously diagnosed with rickettsiosis who later show Rickettsia spp. IgG seropositivity. Methods: This follow-up observational study was conducted at a tertiary referral hospital in the Yucatan Peninsula. From an initial cohort of 390 patients evaluated for suspected rickettsial disease,

Development and Evaluation of a Dual-Target One-Step Nested PCR for the Detection of Spotted Fever Group Rickettsia spp. in Ticks.

Spotted fever group (SFG) rickettsioses are tick-borne infectious diseases caused by more than 30 Rickettsia species. As ticks may harbor and transmit multiple pathogens during a single blood meal, sensitive and specific molecular detection methods are essential for early diagnosis. Conventional nested PCR is commonly used but is time-consuming and prone to cross-contamination due to multiple amplification steps. This study evaluated a dual-target one-step nested PCR assay developed as a rapid alternative to conventional nested PCR for SFG Rickettsia detection. Gene-specific primers targeting

Japanese Spotted Fever With Marked Serological Cross-Reactivity to Rickettsia typhi Confirmed by Sequence-Based Molecular Analysis: A Case Report.

We report a case of Japanese spotted fever (JSF) showing marked serological cross-reactivity with Rickettsia typhi, which made serological differentiation difficult. An 80-year-old man presented with fever and disseminated erythematous macules. Laboratory finding suggested a rickettsial infection. Minocycline treatment led to clinical improvement. Real-time polymerase chain reaction screening was positive for spotted fever group rickettsiae. Serological testing using paired sera demonstrated comparable increases in antibody titers against R. japonica and R. typhi in the convalescent phase, pre

Molecular identification of Rickettsia conorii subsp. israelensis and other tick-borne pathogens in dog ticks from the Upper East region of Ghana.

Ticks continue to facilitate the spread of pathogens that affect both humans and domestic animals. Domestic dogs interact with humans and other domestic animals, playing a crucial role in the spread of ticks and tick-borne pathogens. This study examined the diversity of tick species infesting domestic dogs and the occurrence of tick-borne pathogens in the Upper East region. Domestic dogs were randomly selected and examined for tick infestation. The sampled ticks were morphologically identified, pooled and screened for tick-borne pathogens using polymerase chain reaction (PCR) and Sanger sequen

Rickettsiae of the spotted fever group in dogs of the Americas: A systematic review and meta-analysis.

Sentinel animals are defined as sensitive indicators of environmental hazards that provide an early warning system for public health interventions. Spotted fever group rickettsioses are zoonotic tick-borne diseases that are considered endemic in different American countries with significant rates of morbidity and lethality, mainly when involving species such as Rickettsia rickettsii and Rickettsia parkeri. Dogs have been recognized as good sentinels for these rickettsioses, since they are hosts to different species and stages of ticks, and they sustain a good immunological response after infec

A real-time PCR assay for detection of Rickettsia rickettsii to aid in vivo infection monitoring.

Rocky Mountain spotted fever (RMSF) is an important and potentially fatal tick-borne disease impacting people and dogs. RMSF, caused by Rickettsia rickettsii, is frequently reported in parts of the North, Central and South American countries. The treatment for RMSF is limited to tetracyclines, specifically to doxycycline. If untreated, the disease can progress rapidly to fatal outcomes, particularly in children. In recent years, fatality rates ranging from 30% to 80% have been reported throughout South America, Mexico, and in parts of Indian reservations in southwest regions of the USA. To aid
