Recombinant Detoxified Holotoxin as a Potent Candidate Vaccine Against Botulism.

Vaccination may represent a suitable strategy for preventing botulism. The recombinant expression of toxin functional domains can induce effective immune responses against botulism. This study aims to develop a safe and effective recombinant detoxified full-length BoNTA vaccine. In this study, we engineered and mutated the toxin activity-related sites on the basis of the full-length BoNTA protein and constructed three detoxified full-length toxin mutants. They were recombinant expressed and purified in Escherichia coli, and the BoNTA/M4 mutant was determined to have the highest safety, with a

Effectiveness of a Bivalent Recombinant Vaccine on the Production of Neutralizing Antibodies Against BoNT/C, BoNT/D, BoNT/CD e BoNT/DC in Bovines.

Background/Objectives. Bovine botulism, although relatively rare, presents significant economic losses due to high mortality rates and restrictions on livestock product trade. Vaccination remains the most effective strategy for preventing botulism-related mortality. This study evaluated the efficacy of a bivalent recombinant vaccine targeting the C-terminal portion of the heavy chain (Hc) of botulinum neurotoxin serotype C (BoNT/C) (Hc BoNT/C) and botulinum neurotoxin serotype D (BoNT/D) (Hc BoNT/D) in inducing neutralizing antibodies against these toxins and their mosaic variants BoNT/CD and

Effects of botulinum neurotoxin-associated proteins on cellular organelle dynamics in NRK-52E cells.

Clostridium botulinum produces seven distinct serotypes (A-G) of botulinum neurotoxin (BoNT), a potent biological toxin that affects the nervous system. In its natural state, BoNT forms a progenitor toxin complex with neurotoxin-associated proteins (NAPs), substantially enhancing its stability and oral toxicity. Recent studies have shown that NAPs in certain serotypes induce vacuolation and death in various epithelial cell types. In this study, we examined the effects of the serotype D NAPs complex (NAPs/D) on intracellular organelles in NRK-52E cells, a rat kidney cell line. NRK-52E cells wer

Validation of an Endopep-suspension immunoassay for the diagnostics of human botulism.

Botulism is a potentially life-threatening disease caused by botulinum neurotoxin (BoNT)-producing bacteria of the genus Clostridium. Laboratory detection of BoNTs in patients’ samples is essential to confirm clinical diagnoses and to identify the causative BoNT serotype. The current ’gold standard’ method for BoNT detection is the mouse bioassay (MBA), a highly stressful animal experiment. A viable animal experiment replacement method must demonstrate high sensitivity, specificity, reproducibility and robustness, as well as comprehensive BoNT subtype detection, and be widely accepted in the f

Botulinum neurotoxin A mutants with enhanced ganglioside binding show improved potency and altered ganglioside selectivity.

Botulinum neurotoxins are the causative agents of botulism, a lethal paralytic disease, but are also one of the most commonly used therapeutics for the treatment of numerous neuromuscular conditions. These toxins recognise motor nerve terminals with high specificity and affinity by using a dual binding mechanism involving gangliosides and protein receptors. The initial recognition of gangliosides is crucial for the toxins' potency. In this study, we employed a synaptosome-binding screening strategy to identify BoNT/A mutants with enhanced ganglioside-binding which translated into improved pote

A Systematic Approach to Diagnosing and Managing a Patient With Excessive Gingival Display and a Hypermobile Lip.

This complex case involved a patient with lip hypermobility and a "gummy smile" who was self-conscious of her aging ceramic restorations. She also lacked a comfortable bite position and desired a restful position for her jaw. The patient was opposed to surgical correction of her lip and temporary botulism toxin injections to decrease the mobility. A treatment plan was produced utilizing a systematic treatment approach that accommodated the patient's treatment preferences and successfully reduced her gingival display, created a comfortable bite, and enhanced her smile, thus achieving all of the

Equine botulism.

Botulism is a severe and often fatal disease in equine patients worldwide. Clostridium botulinum is a ubiquitous soil organism which produces a potent neurotoxin resulting in neuromuscular blockade and flaccid paralysis in affected animals. Definitive diagnosis is often impractical or impossible, leading to diagnosis and treatment based on clinical experience and presenting signs. Delay in case identification and treatment results in rapid deterioration of the patient. Treatment of recumbent equine botulism cases presents challenges due to patient size and requires intensive nursing care. This

Single Tri-Epitopic Antibodies (TeAbs) to Botulinum Neurotoxin Serotypes B, E, and F Recapitulate the Full Potency of a Combination of Three Monoclonal Antibodies in Toxin Neutralization.

Recombinant monoclonal antibody (mAb) botulinum neurotoxin (BoNT) antitoxins, consisting of three mAbs that bind non-overlapping epitopes, are highly potent. However, the three-mAb mixtures pose unique development and manufacturing challenges. Combining even more mAbs to create multivalent antitoxin drugs multiplies those challenges. We previously reported that a single tri-epitopic IgG1-based mAb (TeAb) containing the variable domains of the three parental BoNT/A mAbs and an Fc was as potent as the combination of three IgGs in the mouse neutralization assay (MNA). Here, we extended the tri-ep
