Tumor necrosis factor-α-stimulated gene 6 promotes hematoma clearance after intracerebral hemorrhage in a mouse model.

The prognosis for patients who experience intracerebral hemorrhage is poor because of a lack of effective treatments. Tumor necrosis factor-α–stimulated gene 6 (TSG6) is a secreted glycoprotein that exerts anti-inflammatory effects in various inflammatory diseases. We previously showed that adipose-derived stem cells can inhibit inflammation by upregulating TSG6 secretion in an in vitro model of intracerebral hemorrhage. However, the direct effects of TSG6 on hematoma clearance in vivo remain largely unknown. The aim of this study was to determine how TSG6 affects hematoma absorption in mice s

Effect of admission blood glucose on early mortality in patients with pontine hemorrhage.

Elevated admission blood glucose (ABG) is increasingly recognized as a prognostic marker in intracerebral hemorrhage, but its specific role in pontine hemorrhage (PH) remains underexplored. This retrospective study analyzed data from 247 PH patients admitted to two tertiary hospitals between 2012 and 2016 to evaluate the association between ABG and 30-day mortality. Receiver operating characteristic analysis identified 8.69 mmol/L as the optimal hyperglycemia threshold predictive of mortality. Survival analysis revealed significantly lower 30-day survival rates in hyperglycemic patients compar

Regulation of Hindbrain Vascular Development by rps20 in Zebrafish.

During aging, the brain vasculature undergoes significant deterioration characterized by increased arterial tortuosity, compromised blood-brain barrier integrity, and reduced cerebral blood flow, all of which contribute to various neurological disorders. Thus, understanding the mechanisms underlying aging-related cerebrovascular defects is critical for developing strategies to alleviate aging-associated neurological diseases. In this study, we investigated the role of aging-related genes in brain vascular development using zebrafish as an in vivo model. By thoroughly analyzing scRNA-seq datase

Overexpression of BDNF and uPA Combined with the Suppression of Von Hippel-Lindau Tumor Suppressor Enhances the Neuroprotective Activity of the Secretome of Human Mesenchymal Stromal Cells in the Model of Intracerebral Hemorrhage.

Nerve tissue damage is an unsolved problem in modern neurology and neurosurgery, which prompts the need to search for approaches to stimulate neuroprotection and regeneration of neural tissue. Earlier we have shown that the secretome of human mesenchymal stromal cells (MSCs) stimulates rat survival, reduces the severity of neurological deficits, and decreases the volume of brain damage in a hemorrhagic stroke model. A significant disadvantage of using the MSC secretome is the need to concentrate it (at least 5-10 fold) to achieve appreciable pharmacological activity. This increases the cost of

Diagnosis and etiology of poststroke depression: A review.

Following an acute stroke, patients often encounter a range of impairments affecting their physical, cognitive, verbal, and social capabilities. Poststroke depression (PSD) has been identified as a significant consequence of stroke and serves as a crucial predictor of patient outcomes. The diagnosis of PSD presents a challenge, as the physical manifestations following a stroke can overlap with certain depressive symptoms, leading to a potential underdiagnosis of this condition. This review employs a narrative approach to synthesize diverse findings within this domain. PSD is categorized as an

Comparison of brainstem hematoma volume evaluation techniques with a novel quantitative approach.

This study systematically compares various methods for calculating brainstem hematoma volume, and introduces a novel estimation method based on the maximum cross-sectional area. First, we assessed the accuracy of the Tada formula and 3D Slicer software for quantifying brainstem hematomas of different sizes and morphologies. The results revealed significant differences among methods, with the traditional Tada formula frequently overestimating hematoma volumes. We therefore propose an improved algorithm that substantially increases both the accuracy and reliability of hematoma volume assessment.

3D cell-laden scaffold printed with brain acellular matrix bioink.

BACKGROUND: Intracerebral hemorrhage (ICH) is a severe neurological disorder characterized by bleeding within the brain tissue, typically associated with factors such as hypertension, cerebrovascular disease, and trauma. The transplantation of human umbilical cord-derived mesenchymal stem cells (hUCMSCs) has demonstrated promising effects in restoring neurological function in ICH rats; however, limited retention of these cells significantly impedes their efficacy. To address this limitation, we developed a bioink composed of decellularized extracellular matrix (dECM) and hUCMSCs, which was syn

Clinical characteristics of hypertrophic olivary degeneration following brainstem or cerebellar hemorrhage.

Patients who have experienced bleeding in the posterior circulation of the brain often develop Hypertrophic Olivary Degeneration (HOD). This condition can lead to new neurological problems several months after the initial hemorrhage, potentially worsening the overall outcome for these patients. However, its pathogenesis and prognosis remain inconclusive. The research included 214 patients diagnosed with brainstem or cerebellar hemorrhage, of which 36 developed secondary HOD. The study aimed to analyze the clinical data of these patients, investigate the risk factors associated with HOD develop

Intraventricular continuous BDNF administration ameliorates neuroinflammation and enhances neurogenesis against rodent intracerebral hemorrhage model.

OBJECTIVE: Brain-derived neurotrophic factor (BDNF) is a pivotal growth factor for neuronal survival; however, its precise role following intracerebral hemorrhage (ICH) remains poorly understood. This study sought to investigate whether intraventricular administration of BDNF could enhance neurogenesis and ameliorate neuroinflammation, resulting in improvement of neurological outcomes in a rat ICH model. METHODS: Male Sprague-Dawley rats were utilized to create an acute ICH model by collagenase infusion into the internal capsule. An intraventricular minipump was subcutaneously implanted, with

Single-Cell Sequencing of Human Neural Organoids in a Model of Intracerebral Hemorrhage Reveals Temporally Dynamic Responses to Blood.

BACKGROUND: Intracerebral hemorrhage leads to significant morbidity and mortality due to primary mechanical and secondary neurotoxic injury to brain parenchyma. Timing of surgical evacuation to ensure optimal outcomes is controversial, with recent evidence suggesting early intervention improves functional outcome. Here, we characterize the impact of blood-induced secondary injury on diverse brain cell types in a scalable organoid model of intracerebral hemorrhage. METHODS: Human neural organoids consisting of excitatory neurons, inhibitory neurons, neural progenitor cells, astrocytes, endothe

Salvage medullary perforating artery revascularization using the muscular branch of the occipital artery following occipital artery-posterior inferior cerebellar artery bypass for trapping a ruptured vertebral artery dissecting aneurysm.

Vertebral artery dissecting aneurysms (VADAs) presenting with subarachnoid hemorrhage (SAH) are relatively rare. The re-rupture and mortality rates during the acute phase of SAH remain significantly high. Endovascular coil trapping is one of the definitive treatments to prevent re-rupture; however, it carries a potential risk of medullary infarction derived from occlusion of the medullary perforating arteries (Mperf). Here we present a case with poor-grade SAH resulting from a ruptured left VADA affecting the posterior inferior cerebellar artery (PICA). After obtaining informed consent, open s

Trends in Incidence of Intracerebral Hemorrhage and Its Bleeding Sites from 1985 to 2017 in Japanese Rural Communities.

INTRODUCTION: Evaluating long-term trends for the incidence of intracerebral hemorrhage is a priority for primary prevention. It is also important to assess the trends in the proportions of bleeding sites because the pathogenesis, prognosis, and operative procedures differ among them. METHODS: A prospective community-wide stroke registry in two rural Japanese communities (Ikawa Town and Kyowa Town, with populations of approximately 5,000 and 15,000, respectively) was conducted. The age-adjusted incidence of intracerebral hemorrhage from 1985 to 2017 was calculated by the direct method using t

Human amniotic mesenchymal stem cell-derived exosome inhibits ferroptosis through the suppression of JNK/MAPK-mediated EGR1 expression in spontaneous intracerebral hemorrhage.

INTRODUCTION: Spontaneous intracerebral hemorrhage (ICH) represents an essential part of stroke, wherein ferroptosis is identified as a critical factor leading to the pathogenesis. Symptomatical treatments are mainly adopted which tend to leave severe neurological deficits. Recently, human amniotic mesenchymal stem cell (hAMSC) therapy has developed rapidly in the field of regenerative medicine, with its secreted exosomes (hAMSC-Exos) regarded as effective alternatives owing to the ability to penetrate the blood-brain barrier. Therefore, we aim to explore the therapeutic efficacy as well as mo

Adipose-derived stem cell exosomes promote neurological recovery after subarachnoid hemorrhage via AMPK/mTOR-mediated TFEB activation.

BACKGROUND: Subarachnoid hemorrhage (SAH) is a common neurologically devastating disorder, and existing treatment options remain very limited. Increasing evidence has confirmed the neuroprotective effects of exosomes derived from mesenchymal stem cells (MSC). Our work investigates that adipose-derived stem cell exosomes (ADSC-Exos) exert therapeutic effects against SAH through autophagy-related pathways. METHODS: An intracerebral perforation method was used to establish a rat SAH model. Then we injected PBS containing 50 µg, 100 µg, or 200 µg of exosomes. Rat neurological deficits and brain w

The impact of intraventricular hemorrhage on brainstem auditory function in preterm babies.

OBJECTIVE: Whether intraventricular hemorrhage (IVH) in preterm babies affects the brainstem auditory pathway is undetermined. This study investigated the functional status of the pathway in preterm babies with IVH to assess the effect of IVH on the pathway. METHODS: Brainstem auditory evoked response (BAER) was studied at term equivalent age in 83 preterm babies with or without IVH and 35 normal term controls, and between different groups of babies to detect any abnormality in preterm IVH babies. RESULTS: Compared to the normal term group, the preterm IVH group manifested an elevated BAER t

Induced pluripotent stem cell-derived neural progenitor cells coculturing promotes mice cortical astrocytes antiapoptosis via nuclear factor erythroid 2-related factor 2/heme oxygenase-1 axis in vitro.

BACKGROUND: Intracerebral hemorrhage (ICH) induces secondary brain injury, driven in part by oxidative stress caused by hemin, a toxic hemoglobin breakdown product. Cortical astrocytes, critical for maintaining redox homeostasis, are vulnerable to hemin-induced oxidative damage, exacerbating neuronal injury. Induced pluripotent stem cell-derived neural progenitor cells (iPSC-NPCs) have shown therapeutic potential in brain injury models by promoting neural repair, but their ability to protect astrocytes against hemin toxicity remains unexplored. We hypothesized that iPSC-NPCs coculture mitigate
