Role of olfactory receptor Olfr544 in neuronal cells: Insights into neurite outgrowth and CREB signaling.

Olfactory receptors (ORs) are expressed beyond the nasal epithelium and implicated in brain function, yet their roles remain unclear. This study investigated the function of the OR Olfr544, which has been reported to be highly expressed near amyloid plaques in Alzheimer's disease model mice, using the mouse neuroblastoma N2a cell line. We found that its agonist azelaic acid (AZA) promotes neurite outgrowth and activates the cAMP-PKA-CREB signaling cascade in an Olfr544-dependent manner. Transcriptome analysis of Olfr544-knockout cells revealed altered expression of genes involved in neuroactiv

Imaging nanoscopic co-assemblies of α-synuclein and Tau in human samples.

α-synuclein (αS), amyloid-β (Aβ), and Tau aggregates are hallmarks of many neurodegenerative diseases, yet the prevalence and nature of their nanoscopic co-assemblies remain poorly understood. Here, we adapt a single-molecule pull-down (SiMPull) assay to profile αS, Aβ, and Tau co-assemblies across post-mortem brain tissue homogenates, soluble brain fractions, human serum, cerebrospinal fluid, and saliva. We show that αS-Tau co-assemblies are abundant across all sample matrices, accounting for ~10-70% of total αS aggregates in both Parkinson's disease (PD) and control samples. Notably, αS-Tau

Meprin β is specifically inhibited by its cognate synthetic propeptide.

Meprin β is a zinc-dependent metalloprotease of the astacin family highly expressed in kidney and intestine. Dysregulation of meprin β expression and activity was observed in several pathological conditions such as Alzheimer's disease, cancer, fibrosis, and inflammatory bowel disease. Therefore, inhibition of meprin β could be a promising therapeutic option to treat pathological conditions influenced by this protease. However, available small-molecule compounds such as the broad-spectrum metalloproteinases inhibitors marimastat or actinonin lack specificity. Hence, more selective meprin β inhi

Exercise promotes lipophagy and mitigates cognitive impairment in Alzheimer's disease through downregulating microglial TMEM9.

BACKGROUND: Autophagy-lysosomal dysfunction contributes to lipid droplet (LD) accumulation, which drives pathological progression in Alzheimer's disease (AD). Exercise is reported to restore autophagy-lysosomal function and attenuate cognitive impairment in AD, but its regulation of LD deposition remains unknown. METHODS: 5×FAD transgenic mice underwent a three-month exercise intervention. Cognitive and emotional behaviors were assessed using the Morris water maze, novel object recognition, and open field test. LD deposition was evaluated by oil red O and BODIPY staining. De novo formation of familial Alzheimer's disease-relevant Aβ strains occurs exclusively in the human brain.

In familial Alzheimer's disease (FAD), mutations in the amyloid precursor protein gene (APP) accelerate the aggregation of Aβ peptide, leading to deposition of amyloid plaques within the brain. Conformational variants of Aβ aggregates ("Aβ strains") possess distinct pathological properties and may contribute to disease heterogeneity. While the Swedish APP mutation produces wild-type (WT) Aβ, the Arctic APP mutation generates E22G-mutant Aβ. To clarify whether different FAD mutations are sufficient to specify the formation of distinct Aβ strains, we performed Aβ seeding studies in AppNL-F knock

Real-World Patient Eligibility for Alzheimer's Disease Anti-Amyloid Therapy in Seven Swedish Memory Clinics.

BACKGROUND: Anti-amyloid therapies lecanemab and donanemab are the first potentially disease-modifying treatments for Alzheimer's disease (AD) approved in Europe. We estimated real-world eligibility for these treatments in a large multicenter memory clinic patient cohort in Sweden (MemClin). METHODS: MemClin included patients with a first diagnostic visit between 2016 and 2022 at seven specialist memory clinics in Stockholm, Sweden. To estimate eligibility for anti-amyloid therapy, we applied key inclusion/exclusion criteria from the lecanemab and donanemab appropriate use recommendations. El

Atherosclerotic cardiovascular disease risk is associated with AT(N)V neuroimaging biomarkers and cognitive impairment in the health and aging brain study-health disparities cohort.

BACKGROUND: Cardiovascular risk is increasingly recognized as relevant to cognitive aging and dementia-related brain changes, yet its relationship to multimodal neuroimaging biomarkers of amyloid, tau, neurodegeneration, and vascular injury (AT(N)-V) remains incompletely understood in racially and ethnically diverse cohorts. We examined whether ten-year atherosclerotic cardiovascular disease (ASCVD) risk was associated with AT(N)-V neuroimaging biomarkers and cognitive domain scores. METHODS: This cross-sectional study included 1,626 Health and Aging Brain Study-Health Disparities (HABS-HD) p

An integrated human forebrain organoid reveals microglia-mediated CD8+ T cell recruitment and neuroimmune dysfunction in Alzheimer's disease pathology.

Genetic evidence implicates immune dysfunction in Alzheimer's disease (AD), yet the neuroimmune mechanisms operating in human tissue remain poorly defined. Here, we establish a modular human forebrain organoid platform that systematically integrates human induced pluripotent stem cell-derived microglia and CD8+ T cells to reconstitute multicellular AD pathology. This system enables functional interrogation of both innate and adaptive immune components in a human-relevant context. Using this platform, we demonstrate that microglia mediate amyloid-β clearance and neuronal maturation but also dri

Amyloid-Induced Network Resilience and Collapse in Alzheimer's Disease: Insights From Computational Modeling.

Alzheimer's disease is marked by progressive synaptic loss and cognitive decline, with amyloid-beta accumulation playing a central pathological role. Although the molecular effects of amyloid-beta are well-characterized, the cascade from local synaptic dysfunction to large-scale network collapse remains poorly understood. We present a biophysically grounded computational model that integrates small-world network topology, Izhikevich neuron dynamics, short-term plasticity, and amyloid-beta-induced synaptic pruning to investigate how amyloid burden degrades functional brain connectivity over tim

Investigating Structural Heterogeneities of Amyloid Aggregates with Spatially Resolved Infrared Spectroscopy.

The aggregation of proteins into fibrillar amyloids is central to the pathology of neurodegenerative disorders. However, the relationship between the structure of amyloid aggregates and disease progression remains incompletely understood, in part because amyloid systems are intrinsically heterogeneous: structurally distinct species coexist at every stage of aggregation, and conventional ensemble-averaged methods cannot always resolve them. This Account describes how infrared spectroscopy can be coupled to different imaging modalities to resolve this heterogeneity at multiple length scales: fro

[Astrocytes as a key structural and functional component in the development of mixed dementia].

Mixed dementia, characterized by a combination of Alzheimer's disease and cerebrovascular disease (CVD), is prevalent among the elderly population. Accumulated evidence suggests that CVD plays an active role in the neurodegenerative process, enhancing the amyloid and tau pathways and contributing to the progressive disintegration of the brain's functional and structural networks. This review presents data indicating that astrocytes serve as a critical integrative component, linking the pathologies associated with beta-amyloid and tau protein, dysfunction of the neurovascular unit, blood-brain

Comment on "High-intensity interval training modulates APP expression and reduces neuronal apoptosis in a β-amyloid-induced model of Alzheimer's disease: hippocampal evidence".

Godinho et al. reported that high-intensity interval training (HIIT) modulated molecular and cellular markers associated with neuroprotection in a β-amyloid-induced model of Alzheimer's disease. However, the causal relationship between reduced APP expression and neuronal apoptosis, the translation of molecular adaptations into cognitive outcomes, and the discrepancy between FNDC5 mRNA and protein expression remain unclear. This Letter highlights these three considerations and discusses potential mechanisms that warrant further investigation.

An interstitial fluid-like NICHE-chip for neural stem cell niche modeling and nanoscale neurotrophic delivery.

Neural stem cells (NSCs) reside in anatomically confined niches, where three-dimensional (3D) geometry and interstitial fluid (ISF) flow collectively regulate NSC behavior. However, in vitro platforms that simultaneously recapitulate size-controlled 3D architecture, ISF-like dynamics, and nanoscale therapeutic delivery remain limited. Here, we present a 3D printing-based platform with multi-sized microwells, termed the Neural-Inspired Culture for a Hydrodynamic Environment (NICHE)-chip, which incorporates an osmosis-driven flow system to generate ISF-like flow. The NICHE-chip enabled stable fo

CX3CL1 (fractalkine)-CX3CR1 interface drives CD8⁺ T cell-induced neurodegeneration in human brain organoids.

Immune-neuron interactions play a critical role in neuroinflammatory and neurodegenerative diseases, such as Alzheimer's disease (AD), but how CD8⁺ T cells contribute to brain pathology remains poorly understood. Amyloid-β (Aβ), a hallmark of AD, is known to trigger neuronal stress and inflammation. Here, we established a human cortical organoid (hCO) model to investigate CD8⁺ T cell responses to Aβ-induced neurodegenerative stress. Aβ exposure increased neuronal expression of CX3CL1 (fractalkine), which engages its receptor CX3CR1 on CD8⁺ T cells. Transwell migration assays showed enhanced re

A natural neuronal receptor-derived MRI probe targets β-amyloid oligomers for the diagnosis of Alzheimer's disease.

Early-to-mid detection of Alzheimer's disease (AD) remains challenging because clinically used amyloid positron emission tomography (PET) probes primarily visualize fibrillar β-amyloid (Aβ) deposits and require radioactive isotopes. Magnetic resonance imaging (MRI) offers a radiation-free alternative, but its application in molecular AD diagnosis is limited by insufficient blood-brain barrier (BBB) penetration and poor specificity toward early-to-mid Aβ species. Here, we developed a BBB-crossing liposomal MRI probe, Lipo@D1D2-Gd, by integrating a natural Aβ oligomer-binding receptor domain wit

Cuproptosis: New Mechanisms Linking Endothelial Dysfunction to Neurodegenerative Diseases and Therapeutic Prospects.

Cuproptosis is a copper-dependent form of regulated cell death driven by disrupted copper homeostasis and mitochondrial metabolic reprogramming. It is distinct from other cell death pathways and has been increasingly linked to vascular endothelial dysfunction and neurodegenerative diseases such as Alzheimer's and Parkinson's disease. This article comprehensively reviews the core molecular mechanisms of cuproptosis and its emerging roles in endothelial dysfunction and neurodegeneration, synthesizing recent findings from cellular, animal, and human studies. Cuproptosis exacerbates vascular patho

CD8+ t cell-mediated regulation of synaptic integrity in Alzheimer's disease.

Alzheimer's disease (AD) has traditionally been interpreted through amyloid, tau, and innate immune frameworks, with the complement cascade long regarded as the principal driver of aberrant synaptic pruning. Synaptic loss is increasingly recognized as one of the strongest correlates of cognitive decline. This review examines an emerging CD8+ T cell-synapse axis that extends beyond complement-centric models. We discuss how brain-infiltrating CD8+ T cells intersect with synaptic integrity through MHC-I-dependent and MHC-I-independent mechanisms, including IFN-γ-mediated microglial reprogramming

Extracellular vesicles for Alzheimer's disease therapy: mechanisms, engineering strategies, and clinical translation.

Alzheimer's disease (AD), a progressive neurodegenerative disorder, remains a major global health challenge owing to its complex pathogenesis and the presence of the blood-brain barrier (BBB), which substantially limits the delivery of effective therapeutics to the brain. Extracellular vesicles (EVs), which exhibit favorable biocompatibility, low immunogenicity, and the potential to cross or bypass the BBB under specific conditions, have emerged as promising therapeutic agents and delivery platforms for AD. This review focuses on the therapeutic potential of EV-based interventions in AD and su
