West Nile Virus in Europe: Epidemiology, Vector Ecology, Environmental Drivers, and the Role of Equine Sentinel Surveillance in a One Health Framework.

West Nile virus (WNV) is a mosquito-borne flavivirus that remains an important public and veterinary health concern across Europe. Periodic outbreaks affecting humans, horses, and wildlife highlight the complex ecological interactions underlying viral circulation. This narrative review aims to synthesize current knowledge regarding WNV epidemiology, transmission dynamics, and surveillance strategies in Europe, with particular attention to the Romanian context. Available surveillance data indicate recurrent seasonal transmission in several European regions; however, reported case numbers may be

Reframing West Nile Virus in Latin America: From Enzootic Evidence to Human Risk-Surveillance Gaps and One Health Actions.

West Nile virus (WNV) is a mosquito-borne flavivirus with one of the widest global distributions. Since its discovery in Uganda in 1937, it has become a major zoonotic pathogen, and after its introduction into the United States in 1999, it spread rapidly across the Americas, becoming the leading cause of neuroinvasive arboviral disease. Its expansion illustrates a remarkable ecological adaptability, further intensified by climate change. In Latin America and the Caribbean, WNV circulation has been consistently documented in birds, horses, and mosquitoes; however, confirmed human cases remain d

Spatiotemporal and Demographic Patterns of West Nile Neuroinvasive Disease in Vojvodina, Serbia, 2012-2025.

West Nile neuroinvasive disease (WNND) causes substantial morbidity in endemic regions, yet data on its burden in Serbia remain limited. We conducted a retrospective, population-based study of WNND cases reported in Vojvodina Province, Serbia, from 2012 to 2025. Incidence and mortality trends were analysed by year, residence, age, sex, and week of symptom onset. Multivariable logistic regression was used to identify predictors of fatal outcome. Of 1337 suspected cases, 557 (41.66%) met the WNND case definition (530 confirmed, 27 probable cases) and 98.9% were autochthonous. The mean annual inc

Viral etiology of adult encephalitis in Egypt: the role of West Nile Virus.

BACKGROUND: West Nile Virus (WNV) is a neurotropic virus in the Orthoflavivirus genus of the Flaviviridae family that typically causes asymptomatic infections. Despite the high seroprevalence of WNV in Egypt, no documented cases of neurological involvement were previously recorded. This study aimed to document the causes of encephalitis, including WNV, in Egypt. METHODS: Data were extracted retrospectively from laboratory records of adult patients suspected of encephalitis who underwent cerebrospinal fluid (CSF) and serum examinations during the study period (2018–2024). Eligible records were

Relative proportion and contrasting host preference of Culex pipiens biotypes across Europe.

BACKGROUND: The primary vector of West Nile virus (WNV) in Europe, Culex pipiens, has two morphologically identical but behaviorally and genetically distinct biotypes, here referred to as Pipiens and Molestus. Pipiens and Molestus, and their hybrids, are differentially distributed across Europe and display variable patterns of blood-feeding on birds and humans across the continent, but whether host choice correlates with host preference is unclear. METHODS: Samples of mosquitoes were collected and subsequently biotyped using real-time PCR, following which the relative proportions of each biot

Venezuelan equine encephalitis virus infection causes chronic neurobehavioral outcomes, cellular remodeling, and hippocampal single-cell transcriptomic changes.

Venezuelan equine encephalitis virus (VEEV), a neuroinvasive alphavirus, can cause significant neurological deficits in humans. Viral infections, including VEEV, have been linked to neurological diseases such as Parkinson's and Alzheimer's, though mechanisms remain unclear. Currently, not only are there no therapeutic options for VEEV available, but there is also limited information on the host responses following infection that contribute to neurological sequelae. To fill this gap in knowledge, longitudinal neuropathological, behavioral, and single-cell transcriptomic changes were examined in

West Nile Morbidity and Mortality in a Mid-Atlantic Health Care System, 2013-2024.

West Nile virus (WNV) is transmitted by mosquitoes to humans in many regions and can cause neuroinvasive disease and death. To better understand the local burden of disease caused by WNV and risk factors for disease severity, we analyzed cases from across the Johns Hopkins Health System Corporation. We identified 86 patients who met confirmed/probable case definitions for WNV infection from 2013 to 2024. Seventy-one patients (83%) had West Nile neuroinvasive disease (WNND), and nine (10%) died. Encephalitis (n = 47) was the most common complication and was associated with increased age, alcoho

A data-parsimonious model for long-term risk assessments of West Nile virus spillover.

Many West Nile virus (WNV) forecasting frameworks incorporate entomological or avian surveillance data, which may be unavailable in some regions. We introduce a novel data-parsimonious probabilistic model to predict both the timing of outbreak onset and the seasonal severity of WNV spillover. Our approach combines a temperature-driven compartmental model of WNV with nonparametric kernel density estimation methods to construct a joint probability density function and a Poisson rate surface as function of mosquito abundance and normalized cumulative temperature. Calibrated on human incidence rec

Emerging neurotropic tick-borne flavivirus infections in goats and wild ungulates in the Italian Alps.

Recently a novel neurotropic tick-borne flavivirus (TBFV) was isolated from Alpine chamois with encephalitis and from attached ticks in the Austrian federal state of Salzburg and the Italian provinces of Piedmont and Lombardy, preliminarily characterized, and provisionally named Alpine chamois encephalitis virus (ACEV). However, several aspects of this newly described virus-including its host range and epidemiology-remain unknown. Therefore, surveillance was conducted in 2023 and 2024 in the Orobic Alps (Northern Italy) to investigate the circulation of this virus in wild and domestic ruminant

Hospitalizations associated with endemic and non-endemic mosquito-borne arboviruses in Canada, 2002-2023.

Mosquito-borne arboviruses pose a growing public health concern in Canada, particularly in the context of climate change and increased global travel. This study aimed to quantify the burden of endemic and non-endemic mosquito-transmitted arboviral diseases in Canada by examining hospitalization trends from 2002 to 2023. Using administrative hospital data from the Canadian Institute for Health Information (CIHI) and national West Nile virus (WNV) surveillance data, we analyzed patient demographics, temporal and spatial patterns, and disease classification. Hospitalizations were classified as en

Catch basin larvicide treatments impact adult mosquito West Nile virus vector species in metropolitan Milwaukee, WI, U.S.A.

Roadside stormwater catch basins are frequently treated with larvicides in metropolitan areas because they are ideal larval growth sites for West Nile virus (WNV) vector species Culex pipiens Linnaeus and Cx. restuans (Theobald). Despite the wide-spread implementation of catch basin treatments in integrated vector management programs, there is little entomological evidence to suggest catch basin larvicide treatments reduce adult mosquito abundance and thereby impact WNV risk. We evaluated the impact of treating catch basins at three of four sites (700-1000 basins each) with a formulation of Ly

Tick-borne encephalitis vaccination coverage among outdoor workers in Sweden: a nationwide survey.

Tick-borne encephalitis (TBE) is an emerging public health concern in Sweden, with a steadily increasing incidence over the past two decades. Climate change, expanding tick populations, and increased human exposure contribute to the growing burden of disease. Although TBE vaccination is highly effective, it is not included in Sweden's national immunization program, and data on vaccination coverage among high-risk occupational groups remain unknown. We conducted a nationwide cross-sectional Knowledge, Attitude, and Practice (KAP) survey in 2024 to assess TBE vaccination coverage among major out

Management Implications of Shifting Culex quinquefasciatus West Nile Virus Transmission Suitability in Florida.

BACKGROUND: West Nile virus (WNV), a mosquito-borne flavivirus, has circulated in the United States since 1999. In 2025, Florida was home to 24 million people, with projected increases in population and urbanization in a changing climate. The southern house mosquito, Culex quinquefasciatus, is found in every county, and is a major vector for WNV. Describing shifting WNV transmission risk is important to inform public health and vector control planning. MATERIALS AND METHODS: Using published estimates of thermal suitability of WNV transmission by Cx. quinquefasciatus, with climate models and p

Identification of host lncRNAs that impact Venezuelan equine encephalitis virus TC-83 replication.

Venezuelan equine encephalitis virus (VEEV) causes encephalitis in humans and equids, and there are no vaccines or therapeutics for humans. In recent years, non-coding RNAs have emerged as critical regulatory factors affecting different cellular pathways. Specifically, long non-coding RNAs (lncRNAs) have been identified as regulators of antiviral pathways; however, their role in VEEV infection has not been assessed. Here, we show differential expression of several lncRNAs in primary mouse target cells infected with a vaccine strain of VEEV (TC-83) but not a pathogenic strain (TrD). Among the d

A single dose of purified human antibody from transchromosomic bovines mitigates aerosolized Venezuelan equine encephalitis virus disease in cynomolgus macaques.

Venezuelan equine encephalitis virus (VEEV) is a mosquito-borne RNA virus that causes low mortality but high morbidity in humans. In addition to natural outbreaks, there is potential for exposure to VEEV via aerosolized virus particles. Currently there are no FDA-licensed vaccines or antiviral therapies for VEEV. We previously demonstrated that high titer anti-VEEV human antibody preparations derived from the plasma of hyperimmunized transchromosomic (Tc) bovines provided significant protection to mice from subcutaneous and aerosol challenge with virulent VEEV (Gardner et al., J. Virol., 91:e0

Descriptive Epidemiological Analysis of West Nile Virus Human Infections in Italy: A Surveillance-based Study (2018-2023).

West Nile virus (WNV) is a re-emerging mosquito-borne RNA virus of the Flaviviridae family that continues to pose a significant public health threat. It is maintained in nature through a transmission cycle between wild birds, the main hosts, and vector mosquitoes, primarily Culex spp. In Italy, WNV infections have been reported since 2008, with seasonal outbreaks occurring almost annually. This surveillance-based descriptive epidemiological analysis summarizes data on confirmed human infections between 2018 and 2023, analysing annual incidence rates, 95% confidence intervals, temporal trends,

Co-release of tick-borne encephalitis virus RNA and structural proteins via virus-like particles and extracellular vesicles.

Tick-borne encephalitis virus (TBEV) is a medically important flavivirus that causes severe neurological diseases in humans. The assembly of flaviviruses is initiated by the interaction between capsid (C) proteins and viral genomic RNA, yet the molecular determinants that govern RNA encapsidation remain unclear. In this study, we established a TBEV virus-like particle (VLP) system to analyse viral factors that influence viral RNA incorporation independently of productive infection. Using a reporter-containing TBEV minigenome, we investigated the contribution of UTRs to the incorporation of RNA
