Antibodies targeting a conserved cryptic epitope at the influenza hemagglutinin head-stem interface via distinct binding modes.

Influenza A virus continues to pose pandemic threats, as demonstrated by the recent global spread of highly pathogenic H5N1 strains. Consequently, the development and characterization of vaccines targeting zoonotic influenza subtypes have become a major focus of the field. In this study, we characterize two broadly reactive antibodies previously isolated from H5N1 vaccinees. Although they have distinct immunoglobulin germline gene usage and binding modes, cryo-EM analysis shows that both antibodies share a highly conserved cryptic epitope at the interface of the hemagglutinin (HA) head and ste

Report of the National Influenza Surveillance Scheme, 2022-2023.

This report describes influenza surveillance activities in Australia in 2022 to 2023. Data were extracted from several sources constituting the National Influenza Surveillance Scheme. Following the relaxation of public health and social measures which had been applied in Australia in response to the coronavirus disease 2019 (COVID-19) pandemic in 2020 and 2021, the influenza seasons in 2022 and 2023 were characterised by high activity overall, with 1.4 and 1.8 times the number of laboratory-confirmed influenza notifications than those of the 2015-2019 pre-pandemic five-year mean, respectively.

Co-occurrence of viral respiratory pathogens: prevalence, seasonal distribution, and symptomatology in outpatients in Bavaria, Germany, from April 2025 - March 2026.

PURPOSE: The Bavarian Influenza Sentinel (BIS + C) monitors the circulation of respiratory viruses in outpatients in Bavaria, Germany. After the Covid-19 pandemic, the molecular surveillance program was expanded for improved insight into the rate of viral infections. Only limited information on viral co-infections is yet available, so this study provides an insight into their co-occurrence, seasonal distribution, and symptomatic outcome. METHODS: From calendar week 15/2025 to 14/2026, general practitioners and pediatricians collected swab samples from outpatients with acute respiratory diseas

Epidemiology and lagged climatic drivers of pediatric viral lower respiratory tract infections in Tianjin, China (2023-2025).

BACKGROUND: Following the lifting of pandemic-era restrictions, China experienced a sharp resurgence in pediatric respiratory infections; however, post-pandemic epidemiological and climatic data on lower respiratory tract infections (LRTIs) from the temperate continental climate of northern China remain scarce. METHODS: In this retrospective, single-center surveillance study, we characterized the post-pandemic epidemiology of six respiratory viruses in children hospitalized with LRTI in Tianjin and assessed their pathogen-specific, lagged climatic associations. Children aged 0-18 years hospit

Cereblon promotes influenza A virus replication through AMPK ubiquitination.

Influenza A and B viruses (IAV and IBV) continually threaten global health, with IAV posing a risk of emerging pandemics. Rapid viral evolution makes current treatments less effective, highlighting the urgent need for broad-spectrum antivirals. Targeting host factors essential for viral replication may offer a highly promising broad-spectrum antiviral strategy. In this context, cereblon (CRBN), a substrate adaptor of the CRL4 E3 ubiquitin ligase complex, is found to promote both IAV and IBV replication as a key pro-viral host factor. Mechanistically, CRBN targets and degrades AMP-activated pro

Pattern recognition receptors and divergent disease severity in zoonotic viral infections.

Zoonotic viruses continue to emerge unpredictably, cross species barriers, and inflict major health, economic, and ecological burdens. Recent outbreaks of SARS-CoV-2, H5N1 avian influenza virus (AIV), monkeypox, and Zika virus underscore the unpredictability of viral emergence and the urgent need to understand host/pathogen interactions. This review focuses on how innate immune sensing via pattern recognition receptors (PRRs) shapes disease severity across different species. While PRRs initiate antiviral defenses (interferons, ISGs), inflammatory cytokines, and programmed cell death, host outc

[Vaccination against respiratory infections in older adults: Covid-19, respiratory syncytial virus, influenza and pneumococcus].

Population aging increases the risk of severe respiratory infections and their complications in older adults, largely because of immunosenescence. Beyond pneumonia, these infections can lead to cardiac and respiratory decompensation, acute cardiovascular events, loss of functional independence, and institutionalization. Vaccination remains the cornerstone of prevention. This review provides an update on four vaccine-preventable respiratory infections that are particularly common or severe in older adults: coronavirus disease 2019 (COVID-19), respiratory syncytial virus (RSV) infection, seasona

Influenza virus research in an era of emerging respiratory threats: host-virus interactions, surveillance technologies, antiviral management and resistance, and vaccine preparedness.

Seasonal epidemics, zoonotic spillover, antiviral resistance, and pandemic risk continue to make influenza a major challenge for respiratory preparedness. The recent spread of highly pathogenic avian influenza A(H5N1) in mammalian hosts has made it increasingly important to consider molecular adaptation, surveillance, treatment, and vaccination as connected parts of preparedness rather than as separate issues. This review focuses on four interconnected areas: host-virus interactions, surveillance technologies, antiviral management and resistance, and vaccine preparedness. Evidence on mammalian

A novel antiviral drug candidate acts by binding to the hemagglutinin stem.

Due to the high pandemic risk of influenza and increasing drug resistance, novel antivirals are urgently needed. Influenza hemagglutinin (HA) fusion inhibitors, which block the initial steps of viral infection and promote neutralization, have gradually gained recognition as valuable targets in modern drug development. However, with only one approved HA-targeting drug, umifenovir, which possesses poor pharmacokinetic properties, the development of new HA fusion inhibitors remains a key objective. In this study, we identified a novel benzothiophene-based class of small-molecule inhibitors of inf

Vaccinations in pregnancy: An update for a changing time.

Maternal immunization is one of the most effective public health interventions to reduce infectious morbidity and mortality in pregnant women, fetuses, and newborns. Despite robust evidence supporting vaccine safety and effectiveness in pregnancy, global uptake has declined following the COVID-19 pandemic, contributing to resurgent outbreaks of vaccine-preventable diseases. While national immunization schedules vary, there is broad international consensus recommending influenza and tetanus-diphtheria-acellular pertussis (Tdap) vaccination during pregnancy, with increasing adoption of maternal

From molecular evolution to the school health room: the role of community-based public health nurses (helsesykepleiere) in one health communication on cross-species influenza transmission in Norway.

Highly pathogenic avian influenza (HPAI) A(H5N1) clade 2.3.4.4b viruses have undergone an unprecedented expansion of host range since 2021, moving from wild birds and poultry into dairy cattle, domestic cats, and other mammals, with sporadic human infections in Europe and North America (Food and Agriculture Organization et al., 2025a; Rolfes et al., 2025). This expansion is underpinned by well-characterised molecular mechanisms of influenza A virus evolution - antigenic drift, gene segment reassortment, and receptor-binding adaptation - that shape the frequency and direction of cross-species s

Towards broader-spectrum direct-acting antivirals against RNA viruses.

Small molecule antiviral drugs have transformed the management of infections such as those caused by HIV, hepatitis B virus, hepatitis C virus, herpesviruses and influenza viruses. Effective antiviral therapeutics remain unavailable for many other often life-threatening viral infections, many of which are caused by RNA viruses. The development of potent, safe and orally available antivirals is therefore essential both to address current unmet medical needs and to strengthen epidemic and pandemic preparedness. In this Review, we discuss the rationale for building a diversified portfolio of broa

Distinct influenza interfaces in cattle: repeated spillovers of highly pathogenic avian influenza A(H5N1) from wild birds and enzootic circulation of influenza D virus.

Cattle now occupy two epidemiologically distinct influenza interfaces. This Mini Review comparatively synthesizes epidemiological, pathological, experimental-transmission, genomic, and surveillance evidence for highly pathogenic avian influenza A(H5N1) and influenza D virus (IDV), asking how source ecology, tissue tropism, shedding, transmission, persistence, and zoonotic evidence differ between the two viruses. Genetically distinct H5N1 viruses have spilled over repeatedly from wild birds into United States dairy cattle. In lactating cows, infection is prominent in mammary alveolar epithelium

A digitally immune-optimized influenza vaccine broadly neutralizes swine and human H1N1 influenza viruses and protects from heterologous challenge.

UNLABELLED: Influenza A virus (IAV) zoonotic transmission and continuous evolution in multiple host species heighten the risk of emerging novel strains at the human-animal interface. Digitally immune-optimized hemagglutinin (HA), neuraminidase (NA), and matrix-2 (M2) vaccine antigens were engineered and combined as a single DNA expression construct (DVX-H1N1) to maximize immune responses to human seasonal, pandemic, and zoonotic H1N1 IAVs. Immunization of mice with DVX antigens induced broad neutralizing antibody responses to human- and swine-origin H1N1 isolates. The DVX-H1N1 candidate was th

Triazavirin And Its Derivatives Act As Influenza A Virus Fusion Inhibitors.

PURPOSE: The influenza A virus still remains one of the most dangerous pathogens possessing a high potential for the emergence of pandemic strains. However, there are only a few antivirals which are still effective against this virus. So-called viral fusion inhibitors are considered as a relatively new class of such drugs with a high therapeutic potential. METHODS: We combined a system for monitoring the fusion of individual virions with lipid membranes, molecular dynamics simulations and measurements of affinity of charged molecules to lipid membranes. This allowed us to determine the mechan

A divergent betacoronavirus with a functional furin cleavage site in South American bats.

Bats are natural reservoirs for a wide range of RNA viruses. Members of the genus Betacoronavirus, including Severe Acute Respiratory Syndrome virus 2 (SARS-CoV-2) and Middle East Respiratory Syndrome virus (MERS-CoV), have attracted particular attention due to their recent zoonotic emergence. However, much of the known diversity of betacoronaviruses is based on data from Asia, Africa, and Europe, with limited genomic information available from the Americas. Herein, we report the complete genome of a betacoronavirus identified from a Pteronotus parnellii bat sampled in Brazil. Phylogenetic ana

Trends in respiratory virus positivity across urban and rural hospitals in Ontario over three seasons.

OBJECTIVES: Respiratory viruses are a global health burden. While influenza has been a large focus of surveillance, non-influenza viruses remain understudied in Canada, leaving gaps in understanding their seasonal patterns and epidemiology. The coronavirus disease 2019 pandemic further disrupted typical virus seasonality, highlighting the urgent need for expanded surveillance to track evolving trends. METHODS: A multicenter retrospective analysis of molecular testing data for 15 respiratory viruses was conducted on tests performed over three consecutive respiratory virus seasons. Hospital-lev
