Bridging spatial scales in respiratory viral infection.

Respiratory viruses spread within the host through a combination of local expansion and occasional long-range dissemination that seeds new infection foci. We present LEAP (Lotka-Euler Airway Pathogen model), an analytically tractable within-host model that captures this two-scale process. The model takes the form of an age-dependent branching process and yields a closed-form expression for the exponential growth rate during early infection. Using empirical data to parameterize the model, we find that productive dissemination requires only a small number of successful long-range seeding events

Highly Pathogenic Avian Influenza H5N1 in Missouri: Current Situation and Pandemic Potential.

Highly pathogenic avian influenza (HPAI) H5N1, which has caused severe disease in poultry since 1996, has become panzootic in wild birds and recently has spread to dairy cattle and other mammalian species in the United States. Although few human cases have been reported, devastating HPAI H5N1 poultry outbreaks and massive dieoffs in wild birds and in some mammals raise concern about pandemic potential. Surveillance, early detection, public health interventions, farm biosecurity measures, and next-generation vaccines may reduce the risk of a future HPAI H5N1 pandemic.

Prevalence of respiratory viruses among pilgrims in Saudi Arabia during the Hajj 2026: a cross-sectional study.

BACKGROUND: Hajj is one of the world's largest annual mass gatherings and provides favorable conditions for respiratory virus transmission because of intense crowding and extensive international travel. Contemporary post-pandemic molecular surveillance data remain limited. This study investigated the prevalence, determinants, and characteristics of respiratory virus detections among pilgrims during the 2026 Hajj season. METHODS: A cross-sectional study was conducted among adult domestic and international pilgrims over an extended peri-Hajj period (before, during and after Hajj) in the year 20

Nationwide survey of testing practices for respiratory viruses in Australia in the post COVID-19 pandemic era.

BACKGROUND: In the wake of the coronavirus disease 2019 (COVID-19) pandemic, there have been dramatic changes in the testing protocols for respiratory illness. Laboratory identification of respiratory pathogens causing clinical symptoms has become more important in the post-COVID-19 era, both for epidemiological and clinical reasons. This is particularly the case for respiratory syncytial virus (RSV) where a number of preventatives are at the point of clinical translation. The laboratory diagnosis is one of the critical elements in understanding the disease burden of respiratory pathogens and

The Risk of an H5N1 Influenza Pandemic.

The last 110 years have seen four influenza A pandemics, where viruses have emerged from mammals or birds with the ability to transmit and cause disease in human populations. Since 2021, a high-pathogenicity strain of avian influenza, H5N1, has caused a panzootic (pandemic of animals), with global spread in wild birds, resulting in frequent spillovers into farmed poultry and into mammals, including mammal species not previously known to be affected by influenza A. This has created increased opportunity for reassortment, mutation, human infection, and potential adaptation to transmit. The risk

Nosocomial Respiratory Syncytial Virus in the Post-Pandemic Era: Evolving Epidemiology and Integrated Prevention Strategies.

BACKGROUND/OBJECTIVES: Pandemic non-pharmaceutical interventions virtually eliminated respiratory syncytial virus (RSV) circulation in 2020-2021, and their relaxation was followed by unprecedented off-season resurgences. This narrative review, informed by a structured literature search, synthesizes current evidence on healthcare-associated RSV (HA-RSV), including transmission dynamics, vulnerable inpatient populations, and integrated prevention strategies, in the post-pandemic era. METHODS: We searched PubMed/MEDLINE and Europe PMC (January 2000-October 2025) using two complementary strategie

RSV Molecular Epidemiology Pre- and Post-COVID-19 Pandemic Periods in Seattle, Washington, USA.

BACKGROUND: Globally, RSV declined to historically low levels following stay-at-home orders in 2020 and re-emerged as restrictions lifted, with large inter-seasonal outbreaks and other shifts in epidemiology. The effect of the pandemic restrictions on genomic diversity has not been well studied. METHODS: Individuals with respiratory symptoms were enrolled into community-based surveillance studies and provided nasal swabs over three respiratory virus seasons from 2019 to 2022 in the Seattle metropolitan area in Washington State, USA. Samples were tested for RSV by RT-qPCR, with whole genome se

Taxonomy-Guided Discovery of Antiviral Resources in Korean Peucedanum L. (Apiaceae).

The genus Peucedanum (Apiaceae) contains diverse coumarins and related secondary metabolites with a wide range of pharmacological activities; however, the antiviral potential of Korean taxa remains largely unexplored. In this study, we evaluated the cytotoxicity and anti-influenza activity of aerial-part extracts from Korean Peucedanum taxa and the allied species Sillaphyton podagraria. Twenty-six ethanol extracts, comprising 24 accessions representing eight identified taxa and two unidentified Peucedanum samples, were screened against influenza A/Puerto Rico/8/34 (H1N1) virus in MDCK cells. Zoonotic Viral Spillover: From Reservoir Ecology and Viral Evolution to Pandemic Prevention.

Zoonotic viral spillover represents a major global health challenge arising from complex interactions among viral evolution, reservoir ecology, host biology, environmental disruption, and human activities. Successful emergence requires viruses to overcome successive ecological, cellular, and immunological barriers, including receptor compatibility, cellular tropism, replication efficiency, and innate immune restriction. Reservoir hosts such as bats, rodents, birds, and livestock contribute to viral maintenance and diversification through distinct ecological and physiological characteristics, w

Viral Respiratory Infections: COVID-19.

On March 11, 2020, the World Health Organization declared COVID-19 a pandemic. By late 2024, COVID-19 had become endemic, with regional surges caused by new variants and declining population immunity. Case fatality rates have decreased because of vaccination and the emergence of less virulent variants, but long COVID persists as a public health concern. Because of structural issues, including those related to employment, housing, health care access, and mistrust, minority and low-income individuals are at higher risk for COVID-19 infection and mortality. Preventive strategies include shared de

Viral Respiratory Infections: Influenza.

The influenza virus is a highly contagious respiratory pathogen that causes yearly seasonal epidemics and occasional pandemics, leading to significant health and economic consequences. It spreads through droplets, aerosols, and surfaces. Infected individuals are contagious before symptoms appear, with peak infectivity early in the illness. In the United States, the 2024-2025 influenza season was the most severe since 2017-2018, with an estimated 47 to 82 million cases, up to 1.3 million hospitalizations, and up to 130,000 deaths. The risk of adverse outcomes is higher for vulnerable groups, in

Avian influenza A virus polymerase gene leads to temperature-resistant genome replication.

Influenza A viruses (IAV) pose a persistent pandemic threat. Successful cross-species transmission from waterfowl to mammals requires host adaptation for efficient replication and spread. Avian IAVs are adapted to the temperature of the avian gut, approximately 41°C, while human IAVs replicate at 33°C in the upper respiratory tract. PB1, the catalytic core of the viral heterotrimeric RNA-dependent RNA polymerase (RdRp), is essential for viral RNA synthesis. Three pandemic IAV strains (1918, 1957, and 1968) carried an avian-origin PB1 gene, suggesting a selective advantage. Fever, a critical an

Variant escape site mapping and characterization of SARS-CoV-2 nucleocapsid rapid antigen test antibodies.

Rapid antigen tests (RATs) were a critical tool for diagnosing severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection during the height of the pandemic, but their performance depends on antibodies that remain robust to viral evolution. Since most commercial RATs target the viral nucleocapsid protein, mutations in this domain can compromise binding and test performance. To systematically define this risk, we used deep mutational scanning (DMS) to map escape mutations for 82 antibodies across 43 commercially available tests, generating the most comprehensive escape database of SA

Respiratory pathogen epidemiology in children under 5 years hospitalized with acute respiratory infections in Qingdao, China: a retrospective, single-center, repeated cross-sectional surveillance study.

BACKGROUND: During the coronavirus disease 2019 (COVID-19) pandemic, non-pharmaceutical interventions (NPIs) reduced the transmission of severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2) while simultaneously affecting the prevalence of respiratory pathogens to varying degrees. Currently, epidemiological characteristics vary across different regions and among different pathogens, and there is a lack of continuous surveillance data following the outbreak. This study aims to analyze the epidemiological characteristics of six respiratory pathogens in hospitalized children under the age

Antibodies targeting a conserved cryptic epitope at the influenza hemagglutinin head-stem interface via distinct binding modes.

Influenza A virus continues to pose pandemic threats, as demonstrated by the recent global spread of highly pathogenic H5N1 strains. Consequently, the development and characterization of vaccines targeting zoonotic influenza subtypes have become a major focus of the field. In this study, we characterize two broadly reactive antibodies previously isolated from H5N1 vaccinees. Although they have distinct immunoglobulin germline gene usage and binding modes, cryo-EM analysis shows that both antibodies share a highly conserved cryptic epitope at the interface of the hemagglutinin (HA) head and ste
