Pathogen trends and paradigm shifts of respiratory infections in children: a 5-year retrospective study from Perugia.

BACKGROUND: Respiratory infections are one of the most common causes of morbidity and mortality among children. After the COVID-19 pandemic, the epidemiology of respiratory tract infections has changed and has been better traced by more frequent and less expensive molecular and antigenic tests. The main aim of this retrospective study was to evaluate the epidemiology of respiratory infections among children hospitalized in Perugia during the epidemic seasons of 2018-2019 and 2023-2024, comparing clinical severity, hospitalization length, diagnostic and therapeutic interventions. METHODS: We r

Selective Wettability-Driven Evaporation-Enhanced Redox Cycling for Robust and Ultrasensitive Detection of Viral Particles.

Timely detection of viral infections, particularly in settings outside centralized laboratories, is essential for controlling outbreaks. Small electrochemical biosensors are appealing because they are fast, require microliter sample volumes, and can be integrated into portable devices. However, shrinking the sensor area often weakens the signal, making it difficult to detect low viral loads. We previously showed that combining redox cycling with controlled droplet evaporation can boost the signal. Yet, this method faces signal variability due to user-dependent droplet placement. Here, we intro

Case Report: Re-expansion pulmonary edema following a pneumothorax drainage in a patient with H1N1 and Mycoplasma pneumoniae co-infection.

BACKGROUND: Re-expansion pulmonary edema (RPE) represents a rare but potentially fatal complication that can occur subsequent to pneumothorax drainage or pleural effusion. Currently, there is a limited understanding of its underlying pathogenesis and associated risk factors. Co-infection with Mycoplasma pneumoniae and influenza A (H1N1) virus, although rare, may exacerbate lung injury and complicate clinical prognoses. CASE PRESENTATION: Herein, we report a case of a 20-year-old male with no prior significant medical history. The patient presented with fever and chest tightness and was subseq

Therapeutic Efficacy of the Supersulfide Donor NAC-S2 in Influenza Virus Pneumonia via Suppression of Excessive Inflammatory Responses.

Influenza pneumonia is characterized by excessive inflammatory responses that contribute to severe lung injury and mortality. Supersulfides, endogenously produced cysteine-derived persulfides and polysulfides, exert potent antioxidant, anti-ferroptotic, and anti-inflammatory activities; however, their therapeutic potential after disease onset remains unclear. Here, we investigated the efficacy of N-acetylcysteine tetrasulfide (NAC-S2), a highly water-soluble and cell-permeable supersulfide donor, in a mouse model of influenza A virus (IAV)-induced pneumonia. Subcutaneous administration of NAC-

[Progress in research of influence of gene polymorphisms on vaccine immune response].

To introduce the recent progress in the research of gene polymorphisms and differences in vaccine immune responses, this paper systematically summarizes current findings of the associations between human leukocyte antigen (HLA) polymorphisms and other key immunoregulatory gene variations with vaccine responses across different domains, including COVID-19 and influenza vaccines. Furthermore, it discusses the impact of different genotypes on antibody production, immune protection, and the risk for breakthrough infections. To address the challenges posed by genetic polymorphisms, this paper furth

The "New Flu": Ontological Insecurity and Pandemic Sense-Making in the US South.

Pandemics are inherently disruptive, evoking uncomfortable and unsustainable emotions. We explore the emotional landscape of the COVID-19 pandemic and its normalization among vaccine hesitant residents in Oklahoma. Ontological insecurity reported at the pandemic's outset stemmed from the absence of a clear and resonant cultural script that left individuals to construct narratives in which COVID-19 came to be understood as the "New Flu." Within a backdrop of institutional distrust, participants enacted this narrative through individual health practices. The "New Flu" became a symbol that normal

HEDGES co-prevents both SARS-CoV-2 and pandemic influenza infection in mice by rapid, durable co-production of twelve different anti-pandemic monoclonal antibodies.

Despite all currently available anti-pandemic monoclonal-antibodies (mAbs) and vaccines, subsequently emerging pandemic-infections will likely become more pan-resistant-, -transmissible and/or -lethal. We have created HEDGES generation-2, a significantly more-combinatorial, -synergistic version of our generation-1 HEDGES DNA vector-based platform. We previously published that one safe intravenous injection of a HEDGES generation-1 DNA vector encoding one of three different FDA-approved mAbs produced durable therapeutic serum mAb levels as well as critical therapeutic endpoints in immunocompete

Guizhi Gegen Decoction alleviates influenza-associated intestinal injury by balancing lung-gut ILC2 distribution via the S1P/S1PR1 axis.

ETHNOPHARMACOLOGICAL RELEVANCE: Influenza virus primarily affects the respiratory system but frequently induces gastrointestinal complications as well. Guizhi Gegen Decoction (GGD), a classical traditional Chinese herbal formula, has been used to treat influenza patients presenting with gastrointestinal symptoms such as vomiting and diarrhea, yet its underlying pharmacological mechanisms remain to be clarified. AIM OF THE STUDY: This study aimed to elucidate the molecular mechanisms by which GGD ameliorates influenza-associated intestinal injury through the regulation of lung-gut ILC2 homeost

Short-course intranasal LAT9997 therapy limits post-influenza bacterial pneumonia by recalibrating lung immunity.

Secondary bacterial pneumonia causes substantial morbidity and mortality following influenza A virus (IAV) infection, yet no therapies exist that address the underlying immune dysfunction and epithelial damage that predispose to bacterial superinfection. Here, we demonstrate that short-course intranasal treatment with LAT9997, a synthetic peptide derived from human growth hormone, protects against severe IAV infection and subsequent Streptococcus pneumoniae superinfection. Significantly, just two intranasal doses administered on days 1 and 2 post-IAV infection, with no further treatment, impro

Fatal H5N1 Highly Pathogenic Avian Influenza with Retrograde Neuroinvasion in a Free-Ranging Leopard Cat (Prionailurus bengalensis) During a Wild Bird Outbreak in South Korea.

Highly pathogenic avian influenza (HPAI) H5N1 clade 2.3.4.4b viruses spread efficiently via migratory wild birds and increasingly infect mammals. The leopard cat (Prionailurus bengalensis) is an endangered mesopredator in South Korea that frequents wetland-forest ecotones and overlaps with wild waterbirds, placing it at risk of exposure. We describe a fatal HPAI H5N1 infection in a free-ranging leopard cat detected through national wildlife surveillance during a period of widespread H5N1 activity in wild birds along the East Asian-Australasian Flyway. The animal showed acute neurological and r
