OBJECTIVES: To characterize the clinical, epidemiological, and laboratory spectrum of acute viral-associated neurological syndromes in an arbovirus-endemic region of northeastern Brazil.
METHODS: We conducted a prospective, hospital-based cohort (March 2020 to August 2025) including patients with suspected viral neurological syndromes. Systematic testing combined qRT-PCR and serology for arboviruses (CHIKV, DENV, ZIKV, OROV) with multiplex PCR for classical neurotropic viruses. Clinical, imaging, EEG, and ENMG data were collected using standardized electronic forms. Temporal and seasonal tren

Venezuelan equine encephalitis virus (VEEV), a neuroinvasive alphavirus, can cause significant neurological deficits in humans. Viral infections, including VEEV, have been linked to neurological diseases such as Parkinson's and Alzheimer's, though mechanisms remain unclear. Currently, not only are there no therapeutic options for VEEV available, but there is also limited information on the host responses following infection that contribute to neurological sequelae. To fill this gap in knowledge, longitudinal neuropathological, behavioral, and single-cell transcriptomic changes were examined in

West Nile virus (WNV) is transmitted by mosquitoes to humans in many regions and can cause neuroinvasive disease and death. To better understand the local burden of disease caused by WNV and risk factors for disease severity, we analyzed cases from across the Johns Hopkins Health System Corporation. We identified 86 patients who met confirmed/probable case definitions for WNV infection from 2013 to 2024. Seventy-one patients (83%) had West Nile neuroinvasive disease (WNND), and nine (10%) died. Encephalitis (n = 47) was the most common complication and was associated with increased age, alcoho

Many West Nile virus (WNV) forecasting frameworks incorporate entomological or avian surveillance data, which may be unavailable in some regions. We introduce a novel data-parsimonious probabilistic model to predict both the timing of outbreak onset and the seasonal severity of WNV spillover. Our approach combines a temperature-driven compartmental model of WNV with nonparametric kernel density estimation methods to construct a joint probability density function and a Poisson rate surface as function of mosquito abundance and normalized cumulative temperature. Calibrated on human incidence rec

Recently a novel neurotropic tick-borne flavivirus (TBFV) was isolated from Alpine chamois with encephalitis and from attached ticks in the Austrian federal state of Salzburg and the Italian provinces of Piedmont and Lombardy, preliminarily characterized, and provisionally named Alpine chamois encephalitis virus (ACEV). However, several aspects of this newly described virus-including its host range and epidemiology-remain unknown. Therefore, surveillance was conducted in 2023 and 2024 in the Orobic Alps (Northern Italy) to investigate the circulation of this virus in wild and domestic ruminant

Mosquito-borne arboviruses pose a growing public health concern in Canada, particularly in the context of climate change and increased global travel. This study aimed to quantify the burden of endemic and non-endemic mosquito-transmitted arboviral diseases in Canada by examining hospitalization trends from 2002 to 2023. Using administrative hospital data from the Canadian Institute for Health Information (CIHI) and national West Nile virus (WNV) surveillance data, we analyzed patient demographics, temporal and spatial patterns, and disease classification. Hospitalizations were classified as en

Roadside stormwater catch basins are frequently treated with larvicides in metropolitan areas because they are ideal larval growth sites for West Nile virus (WNV) vector species Culex pipiens Linnaeus and Cx. restuans (Theobald). Despite the wide-spread implementation of catch basin treatments in integrated vector management programs, there is little entomological evidence to suggest catch basin larvicide treatments reduce adult mosquito abundance and thereby impact WNV risk. We evaluated the impact of treating catch basins at three of four sites (700-1000 basins each) with a formulation of Ly

Snowshoe hare virus (SSHV) is an arbovirus in the California serogroup known to circulate throughout Canada and northern latitudes of the United States. The clinical spectrum of SSHV infection ranges from asymptomatic or mild febrile illness to neuroinvasive disease; neuroinvasive disease occurs more often in children and young adults. We describe a cluster of confirmed and probable SSHV meningoencephalitis cases in 3 children from Whistler, British Columbia, Canada, in the summer of 2024. We highlight the shared epidemiology, clinical manifestations, serologic diagnostic methods, and outcomes

Tick-borne encephalitis (TBE) is an emerging public health concern in Sweden, with a steadily increasing incidence over the past two decades. Climate change, expanding tick populations, and increased human exposure contribute to the growing burden of disease. Although TBE vaccination is highly effective, it is not included in Sweden's national immunization program, and data on vaccination coverage among high-risk occupational groups remain unknown. We conducted a nationwide cross-sectional Knowledge, Attitude, and Practice (KAP) survey in 2024 to assess TBE vaccination coverage among major out

BACKGROUND: West Nile virus (WNV), a mosquito-borne flavivirus, has circulated in the United States since 1999. In 2025, Florida was home to 24 million people, with projected increases in population and urbanization in a changing climate. The southern house mosquito, Culex quinquefasciatus, is found in every county, and is a major vector for WNV. Describing shifting WNV transmission risk is important to inform public health and vector control planning.
MATERIALS AND METHODS: Using published estimates of thermal suitability of WNV transmission by Cx. quinquefasciatus, with climate models and p

Venezuelan equine encephalitis virus (VEEV) causes encephalitis in humans and equids, and there are no vaccines or therapeutics for humans. In recent years, non-coding RNAs have emerged as critical regulatory factors affecting different cellular pathways. Specifically, long non-coding RNAs (lncRNAs) have been identified as regulators of antiviral pathways; however, their role in VEEV infection has not been assessed. Here, we show differential expression of several lncRNAs in primary mouse target cells infected with a vaccine strain of VEEV (TC-83) but not a pathogenic strain (TrD). Among the d

Venezuelan equine encephalitis virus (VEEV) is a mosquito-borne RNA virus that causes low mortality but high morbidity in humans. In addition to natural outbreaks, there is potential for exposure to VEEV via aerosolized virus particles. Currently there are no FDA-licensed vaccines or antiviral therapies for VEEV. We previously demonstrated that high titer anti-VEEV human antibody preparations derived from the plasma of hyperimmunized transchromosomic (Tc) bovines provided significant protection to mice from subcutaneous and aerosol challenge with virulent VEEV (Gardner et al., J. Virol., 91:e0

West Nile virus (WNV) is a re-emerging mosquito-borne RNA virus of the Flaviviridae family that continues to pose a significant public health threat. It is maintained in nature through a transmission cycle between wild birds, the main hosts, and vector mosquitoes, primarily Culex spp. In Italy, WNV infections have been reported since 2008, with seasonal outbreaks occurring almost annually. This surveillance-based descriptive epidemiological analysis summarizes data on confirmed human infections between 2018 and 2023, analysing annual incidence rates, 95% confidence intervals, temporal trends,

Tick-borne encephalitis (TBE) is an arboviral disease in Europe, with increasing incidence in northeastern Italy. Transmission of the virus via Ixodes ticks is influenced by seasonal dynamics and environmental factors. This study aimed to describe TBE epidemiology in the Veneto Region, assess spatiotemporal patterns, and examine associations with environmental determinants, including possible impacts of climate change. We conducted a population-based study using hospital discharge records to identify TBE cases from 2007 to 2024. Temporal trends and spatial patterns of age-standardized incidenc

Tick-borne encephalitis virus (TBEV) is a medically important flavivirus that causes severe neurological diseases in humans. The assembly of flaviviruses is initiated by the interaction between capsid (C) proteins and viral genomic RNA, yet the molecular determinants that govern RNA encapsidation remain unclear. In this study, we established a TBEV virus-like particle (VLP) system to analyse viral factors that influence viral RNA incorporation independently of productive infection. Using a reporter-containing TBEV minigenome, we investigated the contribution of UTRs to the incorporation of RNA

West Nile virus (WNV) and Usutu virus (USUV) are neurotropic orthoflaviviruses of the Flaviviridae family, transmitted primarily by Culex mosquitoes and maintained in enzootic cycles involving birds. While WNV is a well-established human pathogen causing hundreds of neuroinvasive cases annually in Europe, USUV has emerged more recently, with fewer documented human infections but increasing evidence of neurovirulence. The viral nonstructural protein 1 (NS1) plays a central role in orthoflavivirus pathogenesis by modulating host immune responses, disrupting endothelial barrier integrity, and fac

UNLABELLED: Powassan virus (POWV) is a tick-borne orthoflavivirus that can cause severe neuroinvasive disease. There are no approved vaccines or therapeutics, and the incidence of POWV infection in humans is rising. Here, we isolated and characterized a panel of human monoclonal antibodies (mAbs) from a convalescent donor using single B-cell sorting with POWV EDIII and full-length E ectodomain as antigen bait. Binding and neutralization assays with both POWV reporter virus particles and an authentic strain identified multiple neutralizing mAbs that target distinct epitopes across the E glycopr

Infections caused by arboviruses, a diverse group of viral pathogens transmitted by biting arthropod vectors, mainly mosquitoes, ticks, and midges, can cause a range of illnesses in humans, from mild, influenza-like symptoms to severe neurological complications including encephalitis and viral hemorrhagic fever. According to 2024 World Health Organization statistics, vector-borne diseases collectively account for over 700,000 human deaths annually, with mosquito-borne infections such as dengue, chikungunya, Zika, and yellow fever constituting a growing and significant proportion of this burden

BACKGROUND: Louse flies (Diptera: Hippoboscidae), commonly known as keds or flat flies, are a family of insects that are obligate ectoparasites of birds and mammals, with 216 species documented worldwide. Approximately 75% of the species in the Hippoboscidae family infest birds, while the remaining 25% infest various mammals. Despite the diversity within this family, understanding of their prevalence, vector competencies, and geographic distribution is still quite limited.
METHODS: From August 2023 to December 2024, louse flies were collected from wild raptors brought for rehabilitation to th

UNLABELLED: Zika viral replication vesicles (VRVs) were previously observed to cluster asymmetrically on one side of the nucleus in infected Huh7, A549, and h-NPCs. West Nile virus (WNV) infections in these cells also induce asymmetrically clustered VRVs. The sequential cell responses leading to asymmetric clustering of the WNV and ZIKV VRVs were investigated. Both infections activated the cellular unfolded protein response (UPR), as indicated by increased expression of GRP78, the master regulator of UPR, and activation of the PERK branch of the UPR (p-PERK-p-eIF2α-GADD34). Increased microtubu
