Chandipura Virus: An Emerging Neurological Threat Transmitted by Sandflies in India.

BACKGROUND: Chandipura virus (CHPV), a neurotropic, negative-sense RNA virus belonging to the genus Vesiculovirus (family Rhabdoviridae), has emerged as a significant cause of acute encephalitis syndrome (AES) in India, predominantly affecting children. Since the major outbreaks in 2003-2004 and the recent resurgence in 2024, CHPV has re-emerged as a high-fatality, climate-sensitive arboviral threat transmitted primarily by phlebotomine sandflies. The absence of specific antivirals or licensed vaccines underscores the need for comprehensive understanding and preparedness. METHODS: This narrat

Pre-Clinical Immunogenicity and Protective Efficacy of a Recombinant Subunit West Nile Virus Vaccine Formulated with Aluminum Hydroxide Adjuvant.

West Nile Virus (WNV) is endemic throughout the world and is a leading cause of mosquito-borne severe and/or fatal encephalitic disease. Although several promising WNV vaccines have been developed, none have been approved for human use. Studies were conducted to evaluate the immunogenicity, protective efficacy, and safety of a recombinant subunit WNV candidate vaccine (WN-80E) formulated with aluminum hydroxide (Alhydrogel™) adjuvant and/or in combination with a semi-synthetic saponin (GPI-0100) adjuvant in mice and hamsters, and for toxicology, immunogenicity and safety in rabbits. Adult mice

Insight into cytotoxic and molecular mechanisms of bee venom and its nanoemulsion against West Nile virus in vitro.

West Nile virus (WNV) causes West Nile fever and is primarily transmitted by Culex spp. mosquitoes. The primary hosts of WNV are birds, while humans and horses can also develop illness following infection. Currently, no specific antiviral therapy exists for WNV, and vaccination remains the only effective preventive measure. This study aimed to evaluate the efficiency of the crude bee venom (BV) and its nanoemulsion (NE) on WNV by determining in vitro Vero cells cytotoxicity and viral titer by RT-qPCR. We also conducted BV peptides-envelope glycoprotein of WNV interactions by molecular docking

Incorporating vertical transmission into mechanistic modeling of West Nile virus for optimized control in Germany.

West Nile virus is an arbovirus transmitted by infected Culex mosquitoes to birds, horses, and humans. Since its first recorded case in Germany in 2018, WNV has been endemic in several parts of Germany. Globalization, ecosystem modification and climate change have been identified as significant drivers of the arbovirus spread. However, vertical transmission of pathogens from adult female to their progeny might alter the transmission dynamics. In this study, we designed a process-based epidemic model incorporating climatic variables and epidemiological parameters that replicated mosquito popula

Circulating miRNAs as Biomarkers of Tick-Borne Encephalitis Severity: Association with Cytokine Profile in Febrile, Meningeal, and Encephalitic Forms.

Tick-borne encephalitis (TBE) is a neuroinvasive flavivirus infection with a wide spectrum of clinical manifestations whose molecular mechanisms remain insufficiently characterized. MiRNAs regulate pro-inflammatory cytokine production; therefore, changes in their profiles across different forms of TBE may determine the nature of the cytokine response. The aim of this work was to identify the specific features of the circulating miRNA and cytokine profiles in different clinical forms of TBE, as well as to analyze the correlations between them. The study included patients with febrile, meningeal

Reassessing Regional Tick-Borne Encephalitis Endemicity in Poland Through Seroprevalence Analysis in Blood Donors, 2021-2022.

TBEV is a major cause of viral central nervous system infections in Europe, with heterogeneous geographical distribution and substantial underdiagnosis in low-incidence regions. This study aimed to evaluate the validity of regional TBE risk classification in Poland by combining surveillance-based incidence data with serological markers of TBEV exposure. Plasma samples from 5541 blood donors residing in nine regions were tested by anti-TBEV IgG ELISA, followed by confirmatory VNT, IFA, and anti-NS1 IgG ELISA to differentiate infection-induced from vaccine-induced antibodies. Regions were classi

DNA Vaccine Encoding the prM-E Polyprotein of the Tick-Borne Encephalitis Virus.

We evaluated immunogenicity of the experimental DNA-vaccine construct pVAX-prM-E encoding the polyprotein prM-E of the tick-borne encephalitis virus (strain Sofjin). It was shown that pVAX-prM-E administered to mice using a needle-free jet injector induced the production of high levels of virus-neutralizing antibodies. The level of humoral response to pVAX-prM-E was comparable to that induced by administration of the commercial Tick-E-Vac vaccine. ELISpot analysis of the T-cell response showed that the obtained construct induces high levels of IFNγ in response to stimulation with a pool of spe

Tembusu virus-vectored chimeric vaccine induces potent neutralizing antibody response against West Nile virus in ducks.

West Nile virus (WNV) is a zoonotic, mosquito-borne flavivirus with birds as its primary reservoir host. Infection of domestic ducks leads to disease, decreased egg production, and mortality, highlighting the need for an effective WNV vaccine for ducks. In this study, several live-attenuated chimeric vaccine candidates were engineered with WNV pre-Membrane (prM) and Envelope (E) genes in the backbone of the duck Tembusu virus (TMUV) vaccine strain FX2010-180P. Two mutations in E (D120N and S156P) were introduced to alter glycosylation and reduce virulence. In mice, the vaccine candidates 180P/

A replication-defective tick-borne encephalitis virus generated by NS1 trans-complementation confers robust protective efficacy.

Tick-borne encephalitis virus (TBEV) poses a severe threat to public health, causing neurological disorders with high morbidity and mortality in endemic regions. Therefore, developing safe and effective strategies for TBEV vaccines has long been a focus of attention. Herein, leveraging our well-established NS1 trans-complementation platform, we successfully constructed a high-titer replication-defective TBEV (TBEV-ΔNS1) using the BHK-21 cell line stably expressing Omsk hemorrhagic fever virus (OHFV) NS1 (designated BHKNS1). Retention of the NS1 deletion in TBEV-ΔNS1 was confirmed by continuous

Modeling the impact of host heterogeneity on the risk and dynamics of a West Nile virus epidemic.

West Nile virus (WNV) is a mosquito-borne arbovirus with significant ecological and public health implications. Its transmission cycle involves avian hosts and mosquito vectors. Many factors, including host diversity and mosquito population dynamics are known to shape epidemic patterns. In this study, we extend previous work by incorporating multiple host types, horizontal transmission among hosts, and mosquito feeding preference into a WNV model. Using a system of ordinary differential equations, we analyze the impact of variable host competence, host abundance, host community structure, and

Not all West Nile virus lineages behave alike: dose-response differences between lineages 1 and 2.

The globally distributed arbovirus West Nile virus (WNV) is expanding across Europe, with rising numbers of human cases and an increasingly broad geographic distribution. Transmitted primarily by Culex mosquitoes, WNV lineages 1 and 2 are pathogenic to humans, yet their differences in transmission dynamics remain unclear. As vertebrate hosts develop varying viremia, mosquitoes encounter a wide range of viral doses, which may influence transmission. Here, we investigated the dose-response differences between the lineages in Cx. pipiens biotype pipiens, Cx. torrentium, and Aedes albopictus. Mosq

Downregulation of thioredoxin-like 1 contributes to miconazole's antiviral activity against West Nile virus.

INTRODUCTION: West Nile virus (WNV)-associated neurological diseases pose a global public health burden, yet no approved antiviral treatments are available. This is primarily due to the challenges of crossing the blood-brain barrier and the lengthy, costly process of drug development. METHODS: High-throughput screening of a blood-brain barrier-penetrating FDA-approved compound library was performed to identify anti-WNV candidates in a neuronal cell infection model. Antiviral efficacy was further evaluated across different target cells and a range of flaviviruses, as well as in a mouse model o

Finite-time stability of the West Nile Virus model with Brownian motion.

West Nile virus disease is an acute infectious illness caused by infection with the West Nile virus, which was first isolated from the blood of a febrile patient in the West Nile region of Uganda in 1937. In recent decades, West Nile fever has continued to expand across endemic regions globally, with no specific antiviral treatment or vaccine currently available for prevention. This study examines the finite-time stability of a stochastic West Nile virus model. Initially, the boundedness, existence and uniqueness of the solution are established. Furthermore, by constructing a Lyapunov function

Bovine lactoferrin confers antiviral effect against La Crosse virus by interfering with virus-heparan sulfate proteoglycan binding.

La Crosse virus (LACV) is a mosquito-borne orthobunyavirus within the California serogroup that primarily causes neuroinvasive disease in pediatric populations in the upper Midwestern, mid-Atlantic, and southeastern states of USA. Despite its substantial impact on public health, no vaccine or effective antiviral treatment has been developed so far. In this study, we investigated the role of the cell-surface heparan sulfate proteoglycan (HSPG) in LACV cell attachment and assessed the in vitro antiviral potential of bovine lactoferrin (bLf), which interacts with both the cell surface molecules a

Increasing Seroprevalence In Horses Evidences Widespread Circulation Of West Nile Virus In Albania.

West Nile virus (WNV) is a zoonotic mosquito-borne flavivirus that causes West Nile disease, an emerging and re-emerging infection of major global importance. Horses are an important sentinel species for monitoring WNV circulation. Since viraemia in horses is brief and viral loads in the blood are low, serological assays are the primary tools used to monitor WNV exposure. This study aimed to estimate the seroprevalence of WNV antibodies in horses in Albania. Between 2017 and 2020, 509 serum samples were collected from apparently healthy, unvaccinated horses across different regions of the coun

Serological evidence of hidden circulation of three arthropod-borne diseases (Rift Valley fever, West Nile fever, and Crimean-Congo haemorrhagic fever) in dromedary camels from the Eastern Algerian Sahara.

Several arthropod vectors, including ticks, sand flies, and mosquitoes, play a prominent role in the introduction and spread of numerous viral diseases. Despite Algeria being at high risk of arbovirus emergence, data on the circulation of arboviruses remain limited, except for some reports on West Nile virus (WNV). Therefore, the aim of this study was to investigate the seroprevalence of antibodies against three arboviruses in several regions of the Algerian Sahara in order to document previous viral exposure. Serum samples collected from dromedaries between May and November 2023 were tested f

Molecular and Serological Screening of West Nile and Usutu Virus in Birds and Humans: A Sentinel Study in Finland.

BACKGROUND: The northward expansion of mosquito-borne viruses such as West Nile virus (WNV) and Usutu virus (USUV) in Europe, driven partly by climate-related changes affecting vectors and hosts, poses an increasing threat to human and animal health. Reliable diagnostic tools are therefore essential for early detection and effective surveillance. OBJECTIVES: This study aimed to develop and evaluate sensitive molecular assays for WNV and USUV, apply them to screen wild birds and human patients in Finland, assess bird exposure through serology, and evaluate national preparedness for emerging ar
