BACKGROUND: Influenza virus-associated encephalitis is a rare but potentially threatening complication of influenza. Although it has been reported in various populations, its pathophysiological, clinical, radiological and neuropsychological features remain largely unknown. Further data are therefore needed to develop appropriate management strategies.
CASE PRESENTATION: We describe four French patients (two adults and two children) presenting with neurologic impairment after confirmed influenza, leading to the diagnosis of influenza virus-associated encephalitis. All of them were assessed usi

INTRODUCTION: Orthoflaviviruses are emerging in Europe and have potential significant impact on animal and public health. Usutu virus (USUV) and West Nile virus (WNV) have been first detected in the Netherlands in 2016 and 2020, respectively. Wild boars (Sus scrofa) have been suggested as an interesting population to detect the circulation of orthoflaviviruses. We aim to provide epidemiological insights of USUV and WNV infections in wild boars in the Netherlands. We further explore the interpretation of serological results, taking into account cross-reactivity patterns.
METHODS: A total of 15

T cell exhaustion remains a barrier to adoptive cell therapies. Although γc cytokines are used to expand T cells, their impact on T cell states remains incompletely defined. Using an in vitro model of repetitive TCR stimulation, we found that high-dose IL-2 or IL-15 promoted exhausted CD8⁺ T cell (TEX) differentiation, with increased inhibitory receptor expression and loss of stem-like features. During chronic LCMV Clone 13 infection, high IL2Rβ expression marked virus-specific CD8⁺ T cells with features of terminally exhausted TEX cells, whereas IL2Rβ deficiency favored progenitor-like popula

OBJECTIVE: West Nile virus (WNV) is a mosquito-borne pathogen of growing public health concern that is endemic in Israel and has been responsible for intermittent outbreaks. The seroprevalence and durability of functional antibody responses in asymptomatic WNV-infected populations remain unclear.
METHODS: We screened a large cohort of asymptomatic healthcare workers (HCWs) for WNV IgG and neutralizing antibody titers. Predictors of seropositivity and neutralization levels were assessed using multivariable regression. Among seropositive individuals, longitudinal samples, collected up to 44 mon

A 29-year-old woman presented with fever, malar rash, and alopecia. Laboratory evaluation demonstrated a high-titre antinuclear antibody of 1:5120, hypocomplementaemia, and positivity for anti-double-stranded DNA and anti-Smith antibodies, leading to a diagnosis of systemic lupus erythematosus. During the clinical course, she developed progressive peripheral neuropathy with inability to walk. Intravenous methylprednisolone pulse therapy and intravenous immunoglobulin were administered, but neurological improvement was limited, and intravenous cyclophosphamide therapy was initiated. Central nervous system (CNS) infections may cause sudden unexpected death when diagnosis is delayed or missed. We report a fatal case of Streptococcus pyogenes meningoencephalitis and systematically review postmortem investigations of fatal bacterial CNS infections. A 57-year-old woman was found dead at home after untreated otalgia, hearing loss, purulent otorrhea, low-grade fever, asthenia, and vomiting. Autopsy showed diffuse cerebral edema, subdural empyema involving the posterior fossa, and cerebellar tonsillar herniation. Histology demonstrated acute suppurative meningoencephalitis with v

Varicella-zoster virus (VZV) encephalitis is a rare but severe condition associated with significant morbidity and mortality. Its clinical presentation is often nonspecific and may overlap with other neurological conditions, including complications of influenza infection. We report a case of a 76-year-old man with confirmed influenza A infection who developed progressive neurological deterioration. Notably, no cutaneous manifestations suggestive of herpes zoster were observed. Cerebrospinal fluid analysis revealed lymphocytic pleocytosis, and polymerase chain reaction confirmed the presence of

BACKGROUND: Cryptococcal meningoencephalitis is a serious opportunistic infection, classically associated with advanced immunosuppression. The increasing use of immunomodulatory drugs, such as leflunomide, has broadened the at-risk population.
CASE REPORT: A 66-year-old woman with rheumatoid arthritis, on long-term leflunomide therapy, presented with progressive cognitive decline, behavioral changes, and myoclonus over a six-week period. The initial presentation mimicked rapidly progressive dementia. Cerebrospinal fluid analysis showed elevated opening pressure, hypoglycorrhachia, and hyperpr

CD8 T cells and the process of T cell exhaustion are central topics in both basic immunology and translational research, including clinical applications. Here, we provide a comprehensive, step-by-step guide to investigate CD8 T cells in murine in vivo lymphocytic choriomeningitis virus (LCMV) infection models. We outline viral stock preparation, infection, and viral titer evaluation. We put an emphasis on flow cytometry as a method to investigate phenotypical and functional characteristics of CD8 T cell exhaustion, but the protocols provided will also facilitate imaging or transcriptomic analy

HIV Cerebrospinal Fluid (CSF) viral escape is a condition characterized by inadequate virological control within the CSF despite systemic antiretroviral therapy, often assessed through discordant CSF and plasma HIV viral loads. The clinical relevance of CSF viral escape remains poorly understood, but it is thought to contribute to a spectrum of neurological and neurocognitive manifestations. The present study details the case of a 51-year-old individual from São Paulo, Brazil, presenting with acute meningoencephalitis and seizures in the context of persistent low-level viremia. This summary report on vaccine preventable diseases in Australia is the latest in a series of multi-year surveillance reports that bring together the three most important national sources of routinely-collected data on VPDs (notifications, hospitalisations and deaths) and serve as a resource to support surveillance and control of VPDs in Australia. This report focuses on the four-year period 1 January 2019 to 31 December 2022 with longer term trends also presented. Two VPDs are included for the first time: coronavirus disease 2019 (COVID-19) and Japanese encephalitis virus (JEV) infection.

Oncogenic viruses are widely recognized for their pivotal role in malignant transformation; however, their capacity to induce neurological disorders has received comparatively less attention. Increasing evidence indicates that several oncogenic viruses, including human T-cell leukemia virus type 1 (HTLV-1), human papillomavirus (HPV), hepatitis B virus (HBV), hepatitis C virus (HCV), and Kaposi's sarcoma-associated herpesvirus (KSHV), are associated with a broad spectrum of neurological complications involving both the central and peripheral nervous systems. These manifestations arise through

West Nile virus (WNV) is an emerging/re-emerging flavivirus representing one of the major global causes of mosquito-borne encephalitis, yet no approved therapeutics or vaccines currently exist. To characterize the kinetics of WNV-specific immunity, we evaluated the humoral response in a longitudinal cohort of 55 WNV lineage 1-infected blood donors up to 1 year. We demonstrate that WNV infection elicits robust and durable binding and neutralizing antibodies. Using a biphasic decay model, we estimated the half-life of neutralizing antibodies to be 37 days during the rapid decay phase and 472 day

BACKGROUND: West Nile virus (WNV) is an emerging pathogen in Europe with an unprecedented northward expansion. In Romania, the virus has been endemic for at least three decades, and human infection cases are registered yearly, sometimes culminating in outbreak events. Bucharest, the capital city of Romania, is the largest urban area of the country, encompasses a multitude of mosquito and bird habitats, and has been continuously reported as an active hotspot for WNV transmission. The circulation of lineage 2 has been documented in Bucharest since 2010 in mosquito and human populations. Our stud

Several animal species, including horses and pigs, can be infected with Japanese encephalitis virus (JEV). Feral pigs (Sus scrofa) are considered an effective amplifying host for JEV in natural habitats. In this study, 265 dried blood spots were collected from feral pigs in four selected North Queensland local government areas, and antibodies against JEV were detected using a competitive enzyme-linked immunosorbent assay (cELISA). The results showed that 26.4% (70/265) of the feral pigs had antibodies specific to JEV. We found a significant difference in the seroprevalence of JEV in the Wet Tr

Inborn errors of type I interferon (IFN-I) immunity account for ∼7-8% of herpes simplex encephalitis (HSE) cases in young children, but the pathogenesis of HSE in adults is largely unknown. We report that IFN-I autoantibodies (auto-Abs) in serum/plasma samples drawn before, during, or after HSE in 16/339 Danish adults neutralized 0.1-10 ng/ml of IFN-I. Neutralization was IgG-mediated and was also detected in cerebrospinal fluid in several patients. Patients' serum impaired the ability of exogenous IFN-I to protect fibroblasts against herpes simplex virus (HSV)-1 infection in vitro. Among patie
