Association between serum perfluoroalkyl substances and COPD risk: population evidence and in vitro validation.

BACKGROUND: Per- and polyfluoroalkyl substances are characterized by environmental persistence, bioaccumulation potential, and multi-organ toxicity. Given their ubiquitous presence in the environment and human serum, concerns regarding respiratory health risks are growing, particularly due to scarce evidence on environmental etiologies of chronic obstructive pulmonary disease in never-smokers. This study aimed to investigate the mechanistic role of these substances in chronic obstructive pulmonary disease through a population-computational-experimental paradigm, with specific focus on lipid-me

Localized Tracheobronchial Transthyretin Amyloidosis Diagnosed After Hemoptysis: A Case Report.

Pulmonary involvement in transthyretin amyloidosis (ATTR) is rare, and tracheobronchial involvement is particularly uncommon. We report a case of wild-type ATTR amyloidosis presenting with hemoptysis in a 66-year-old woman with dyspnea. Chest computed tomography revealed bronchial wall thickening and infiltrative opacities. Bronchoscopy showed irregular mucosal lesions extending from the trachea to both bronchi. Histopathological examination of bronchial biopsy specimens showed Congo red-positive deposits, and immunohistochemistry confirmed transthyretin, establishing the diagnosis of ATTR amy

Airway Stenting for Benign and Malignant Central Airway Obstruction: A Case Series.

Central airway obstruction (CAO) is a life-threatening condition caused by malignant or benign processes involving the trachea and main bronchi. Bronchoscopic airway stenting is an established intervention to restore airway patency, relieve symptoms, and stabilize patients, particularly in urgent or critical settings. However, its clinical role varies according to underlying etiology and disease context. We report a single-center case series of seven adult patients with symptomatic CAO who underwent bronchoscopic airway stenting. Six cases were associated with malignant disease, and one involv

Enhancing the translational potential of the ovine pulmonary adenocarcinoma model.

Ovine pulmonary adenocarcinoma (OPA) is a transmissible lung cancer of sheep caused by Jaagsiekte sheep retrovirus (JSRV), resulting in substantial animal welfare concerns and economic losses worldwide. We previously developed a bronchoscopic JSRV catheter instillation technique targeting the cardiac lung lobe that reliably reproduced the spectrum of OPA lesions observed in naturally occurring disease. Beyond its value for investigating JSRV pathogenesis, this experimental system provides a large animal platform for studying human lung cancer. Here, we sought to enhance its translational utili

Vanishing Bronchus Syndrome After Lung Transplantation: Clinical Characteristics, Airway Colonization, and Stent Outcomes.

BACKGROUND: Airway complications remain a significant cause of morbidity after lung transplantation. Among them, vanishing bronchus syndrome (VBS) represents a severe form of airway injury characterized by progressive distal bronchial stenosis. Although ischemia has been implicated in its pathogenesis, the role of airway colonization with multidrug-resistant organisms remains incompletely understood. MATERIALS AND METHODS: We conducted a retrospective, single-center study of lung transplant recipients who developed VBS between January 2001 and November 2025. Clinical characteristics, bronchos

Impact of Neoadjuvant Immunotherapy Combined with Chemotherapy on Surgical Margin Strategy in Central Non-Small-Cell Lung Cancer.

BACKGROUND: The CheckMate 816 trial showed that neoadjuvant immunotherapy combined with chemotherapy improved pathological complete response and tumor downstaging compared with chemotherapy alone. Whether this permits less extensive resection, such as reduced margins, without compromising survival remains unclear. METHODS: We included 142 patients with central-type non-small-cell lung cancer (NSCLC) who received neoadjuvant chemoimmunotherapy at three hospitals between 2017 and 2022. All were initially considered candidates for sleeve lobectomy (SLT) based on pretreatment findings. After trea

3D CT-Based quantification of airway structural changes during asthma exacerbations: a retrospective observational study.

BACKGROUND: Traditionally, mucus plugs have been evaluated using two-dimensional computed tomography (CT) images through expert visual assessment, a process that requires substantial experience and specialized knowledge of airway anatomy. In the present study, we propose a novel approach employing three-dimensional CT (3D-CT) with semi-automated quantification, which can be performed in routine clinical practice without requiring expert readers. We aimed to demonstrate its feasibility and clinical relevance for assessing mucus plug burden and exacerbation severity in asthma. METHODS: This exp

Social interdependencies: the deep evolutionary roots of morality and normativity.

The conducting airways of the respiratory system play a crucial role in filtering, humidifying, and directing air into the lungs. Among the specialized cell types within these airways, airway serous cells are notable for their secretion of watery, protein-rich fluids and enzymes, which contribute to maintaining airway surface liquid homeostasis and defending against pathogens. However, the distribution and abundance of serous cells across different species in the conducting airways remain poorly understood. In this study, we addressed this gap by investigating the spatial distribution of the a

Selective KIT-inhibition Supports Mast Cells as Key Drivers of Allergic Asthma Pathogenesis.

BACKGROUND: Asthma is a chronic respiratory disease characterized by airway inflammation, airway hyperresponsiveness (AHR) and airway remodeling. Mast cell numbers are increased in asthmatic airways and contribute to bronchoconstriction and inflammation; yet their role in asthma development remains elusive. OBJECTIVE: To investigate mast cell contributions to allergic asthma by pharmacologically targeting KIT, a central regulator of mast cell survival and activation, using the selective inhibitor THB001. METHODS: THB001 effects were assessed in human lung mast cells (HLMCs), isolated human b

Impact of temperature, pH, and L-tryptophan supplementation on indole-3-acetic acid biosynthesis in Bacillus subtilis strain ESU181.

The bovine respiratory syncytial virus (BRSV) known as Bovine orthopneumovirus according to the international classification is one of the most important etiological agents of respiratory diseases in calves. At present, rapid and reliable methods to detect and measure the concentrations of this pathogen are needed. The objectives of the survey are developing the real-time polymerase chain reaction (PCR) to identify and quantify the BRSV RNA and, based on it, determining the number of the virus genomes in the respiratory tract of sick animals during the disease outbreaks. Airway epithelial heterogeneity and mucus plugging in asthmatic bronchioles.

RATIONALE: Bronchiolar dysfunction is associated with asthma exacerbations and poor symptom control. However, the molecular pathophysiology of asthmatic bronchiolar disease is poorly defined. OBJECTIVES: Test the hypothesis that asthmatic bronchioles exhibit disturbances in epithelial biology that produce MUC5AC-dominated mucus plugs. METHODS: Peripheral lung tissues from severe asthmatics, fatal asthmatics (FA), and controls were evaluated with histology, RNA in situ hybridization, and immunohistochemistry. Isolated bronchiolar and bronchial basal cell responses to IL13 were compared in cul

Digital twin-based bronchoscopy simulator improves training performance and skill retention of novices: a randomised controlled study.

RATIONALE: Conventional bronchoscopy training often does not ensure lasting skill retention or adaptability to different anatomies, limiting real-world impact. This study used a digital-twin bronchoscopy simulator with various CT-derived bronchial tree models to better train novices. OBJECTIVES: To explore training with various anatomically diverse bronchial tree models in novices' bronchoscopy performance. METHODS: 60 bronchoscopy-naive participants were randomly assigned to three groups (n=20 each): control (written instruction only), anatomic-uniformity (trained on one standard bronchial

Does mechanobiology drive respiratory disease? Biomechanical induction of mucus hypersecretion in human bronchial organoids using a photocontrolled biomaterial gel.

Respiratory diseases such as COPD, IPF and severe asthma are major causes of death globally, characterized by chronic inflammation and by fibrotic biomechanical remodelling of the lung ECM. However, present treatments focus on relieving inflammation and symptoms and do not address the mechanobiological aspect. This is in great part because the role of mechanobiology in disease progression and aetiology is not well-understood, indicating a need for new investigatory models. Here we introduce a combined biomaterial and 3D-organoid model, based on a hybrid biomaterial-matrix double-network gel, w

The Evaluation of the Inner Diameter of the Airway in Asthma Recovery by using HRCT: A Retrospective Observational Cohort Study.

BACKGROUND: Although airway size changes occur in patients with chronic asthma, HRCT has not yet been used to assess changes in the inner diameter of the airways. OBJECTIVE: This study aimed to evaluate the airway diameter in asthma recovery by using HRCT. METHODS: Thirty patients with asthma were recruited and underwent HRCT examination in acute exacerbation and stable phase, respectively. The inner diameter of the airway (Din) was measured from the bilateral main bronchi to all 18 segmental bronchi during acute exacerbation and the stable phase. RESULTS: The inner diameter of the airway r

Histopathologic Findings in Three Dogs With Metastatic Carcinoma Presenting With Bronchovascular Bundle Thickening on Computed Tomography.

Carcinoma metastasis to the lungs can occur via multiple pathways, most notably via hematogenous, lymphangitic, or aerogenous routes. Computed tomographic (CT) characteristics of these metastatic pathways can vary, with only sparse reports of bronchovascular bundle thickening (BVBT) associated with metastatic carcinoma. In this retrospective cross-sectional study, three dogs with metastatic carcinoma and BVBT on CT that had corresponding histopathological sections at the site of BVBT were reviewed. These cases showed expansion of the peribronchial/peribronchiolar interstitium with islands of n

Evidence of T2/T3 Endotype Overlap in Mild-to-Severe Asthma with Chronic Rhinosinusitis: A Pilot Study.

RATIONALE: Asthma and chronic rhinosinusitis (CRS) often cooccur and share heterogenous inflammatory processes resulting in frequent exacerbations and poor clinical control. OBJECTIVES: This observational cross-sectional study aims to characterize asthma with comorbid CRS and to understand whether CRS could influence asthma severity focusing on the bronchial immune-inflammatory and remodeling response. METHODS: We assessed the number of inflammatory cells and the expression of type 2 (T2), T3 and remodeling biomarkers in the bronchial mucosa of 47 patients with mild-to-severe asthma who had

A Case of Bronchial Dieulafoy's Disease Presenting as an Endobronchial Polypoid Lesion That Regressed After Bronchial Artery Embolisation.

A 37-year-old woman presented to the hospital with a history of a small amount of hemoptysis. Bronchoscopy revealed a polypoid lesion in the left main bronchus. Due to oozing bleeding from the lesion, biopsy was deferred to avoid fatal haemorrhage risk. Bronchial angiography revealed an abnormal tortuous branch with pulmonary artery shunting. Bronchial artery embolisation with coils was successfully performed without complications. Bronchial Dieulafoy's disease was diagnosed. Follow-up bronchoscopy approximately 4 months post-embolisation showed regression of the polypoid lesion, with residual
