Knowledge mapping of risk mitigation measures against vector-borne diseases.

A comprehensive synthesis of current evidence on surveillance, prevention and control measures for 25 selected vector-borne diseases (VBDs) affecting animals in the EU is presented here. The assessment integrates evidence from systematic literature reviews, modelling studies, field investigations and expert judgement. The aim is mapping the availability and effectiveness of risk mitigation measures (RMMs), including surveillance, movement restrictions, vaccination, culling, medicinal treatments, on-farm biosecurity and vector control. Of the 25 VBDs considered, 13 are listed under the EU Anima

Live-attenuated Rift Valley fever virus vaccination leads to durable, protective immunity independent of sex in C57BL/6 mice.

BACKGROUND: Rift Valley fever virus (RVFV) is a zoonotic arbovirus endemic to Africa and the Arabian Peninsula that can cause disease in humans and livestock. There are currently no licensed vaccines for human use, but several candidates are at the clinical trial stage including DDVaxTM, a preparation of live-attenuated RVFV lacking the nonstructural proteins NSs and NSm (ΔNSsΔNSm). Multiple pre-clinical studies have shown that ΔNSsΔNSm is safe and immunogenic. METHODS: The longitudinal study described in this manuscript examined the additional features of duration, sex and dose in immunogeni

Plasmodium falciparum Malaria and Arbovirus Co-Exposure in the Boende Health Zone, Northwestern Democratic Republic of the Congo.

Background: Malaria remains hyperendemic in the Democratic Republic of the Congo, while arboviral infections are increasingly reported but remain under-surveilled, particularly in remote regions. Overlapping ecological niches and non-specific clinical presentations complicate case management and surveillance. Methods: A cross-sectional door-to-door survey was conducted in December 2023 in Inkanamongo village (Lokolia Health Area, Boende Health Zone, Tshuapa Province). Blood samples were collected from 379 adults; malaria infection was assessed by using HRP2-based rapid diagnostic tests, and ar

Knowledge, Attitudes and Practices Regarding Rift Valley Fever Among Livestock Traders in the Alaotra Mangoro Region, Madagascar.

Rift Valley fever (RVF) is a viral zoonosis endemic in Madagascar, threatening human and animal health as well as the economy. Trade-related livestock movements are a major factor in the spread of RVF virus. While previous RVF research in Madagascar has focused on farmers or general ecology, this study is the first to specifically target livestock traders, the primary drivers for long-distance viral spread, in the Alaotra Mangoro endemic hotspot. This study aimed to assess the level of knowledge, prevailing attitudes and current practices regarding RVF among people engaged in livestock trade i

Immunopharmacologsical and structural insights into Rift Valley fever virus envelope glycoproteins: an immunoinformatics approach to multi-epitope vaccine design.

Rift Valley fever (RVF) is a clinically zoonotic pathogen associated with severe systemic and neurological complications, for which no approved vaccine is currently available. This study aimed to design a novel chimeric multi-epitope vaccine candidate targeting the RVFV Envelope polyprotein (Gn and Gc) via an integrative immunoinformatic and structural modeling approach, targeting viral envelope glycoproteins which play a crucial role in host cell entry and immune recognition. The epitopes used in the vaccine assembly were selected on the basis of antigenicity, allergenicity, and toxicity, the

Biochemical Basis for LRP1 Interaction With Rift Valley Fever Virus Glycoprotein and Its Role in Viral Entry.

The LDL receptor related protein 1 (LRP1) is a host entry factor for Rift Valley fever virus (RVFV), a negative sense RNA virus in the Bunyaviricetes class within the Phenuiviridae family. Previous studies revealed that RVFV glycoproteins (Gn and Gc; GnGc) primarily interact with LRP1 clusters II (CLII) and IV (CLIV) to promote viral entry. However, the mechanism of RVFV GnGc binding to LRP1 is not fully characterized. In this study, we have successfully mapped the roles of individual LRP1 ligand binding regions, known as CR domains, and identified CR25 to be sufficient for high affinity RVFV

Maternal vaccination protects dams and prevents in utero transmission of Rift Valley fever virus in rats.

UNLABELLED: In utero transmission and spontaneous fetal death are hallmarks of Rift Valley fever virus (RVFV) infection in pregnant animals. Compounding evidence indicates pregnant individuals are particularly vulnerable to RVFV, as in utero transmission and increased rates of miscarriage have been reported in people infected during pregnancy. Further, human placenta explants are permissive to RVFV infection. Viable vaccine candidates intended for veterinary or human use must protect vulnerable populations, including pregnant individuals and fetuses. Using a pregnant rat model, we show that ma

Favipiravir tissue distribution and inhibitory quotients in preclinical models: towards a pipeline for evidence-based antiviral repurposing.

OBJECTIVES: Favipiravir (T-705) shows potent in vitro activity but inconsistent in vivo efficacy. We assessed how plasma exposure and tissue distribution shape antiviral coverage using a tissue-based PK/PD framework. METHODS: Favipiravir and its hydroxylated metabolite (M1) were quantified in plasma and multiple organs of Syrian golden hamsters after single and repeated dosing and compared with a non-human primate dataset. We derived tissue penetration factors (TPFs), metabolic ratios (MR) and inhibitory quotients (IQ) against published EC50 values for representative RNA viruses and integrate

Study of patients with acute undifferentiated fever identifies dengue as a growing threat to public health in Mali.

The circulation of arboviruses in sub-Saharan African countries remains poorly documented. The associated health burden may be underestimated and masked by the significance of malaria. Here, we have investigated acute undifferentiated fevers for arboviral infections in Mali (2016-2024). To estimate the proportion of patients with arboviral infection, and in particular dengue. A retrospective (2016-2022) and a prospective (2023-2024) studies were conducted in patients from health centers and hospitals of Mali (mainly in the Bamako region) selected by health professionals. Studies included patie

Climate change and the growing threat of bunyaviruses.

Anthropogenic climate change and its implications for infectious diseases have been central to the scientific and public health discourse for more than three decades. In 2026, we have now entered a climate adaptation phase, with accumulating evidence demonstrating that ongoing climatic variability is directly reshaping the dynamics of multiple vector-borne viruses, including bunyaviruses. Bunyaviruses comprise a diverse group of segmented, negative-sense RNA viruses that include significant human and veterinary pathogens, such as Rift Valley fever virus, Crimean-Congo hemorrhagic fever virus,
