Oropouche virus NSs protein suppresses host transcription by targeting the RNA polymerase II RPB1 protein.

UNLABELLED: Oropouche fever is a debilitating disease caused by Oropouche virus (OROV), an arthropod-borne member of the Peribunyaviridae family. Despite its public health significance, the molecular mechanisms driving OROV pathogenesis remain poorly understood. In other bunyaviruses, the nonstructural NSs protein encoded by the small (S) genome segment acts as a major virulence factor. In this study, infection with the OROV MD023 strain led to nuclear accumulation of NSs and redistribution of nucleophosmin 1 (NPM1) from the nucleolus. OROV infection suppressed nascent RNA synthesis and result

A DNA vaccine candidate provides protection against Rift Valley Fever virus in sheep under natural field conditions.

Rift Valley Fever virus (RVFV) is a mosquito-borne zoonotic pathogen, that causes significant morbidity and mortality in livestock, including high abortion rates in pregnant animals and elevated case fatality in neonates, representing a major threat to both animal and human health. Vaccination is the most effective countermeasure to reduce RVFV's impact. In this study, we designed a veterinary DNA vaccine encoding a consensus RVFV glycoprotein precursor (GPC), optimized for expression in sheep. The construct was evaluated for immunogenicity in mice and sheep and for protective efficacy in shee

Climate change and the emergence of Rift Valley fever virus: a pathological and environmental study of sheep jaundice in Egyptian slaughterhouses.

BACKGROUND: The emergence of Rift Valley Fever Virus (RVFV) and its link to sheep jaundice in Egypt highlights the growing impact of climate change on the epidemiology of vector-borne diseases. Shifting climatic patterns, such as rising temperatures and altered rainfall, have expanded mosquito habitats, enhancing RVFV transmission risks. These environmental shifts create ideal breeding conditions for the mosquitoes, increasing virus's transmission risk to both livestock and humans. Jaundice, a severe symptom resulting from RVFV infection, not only threatens livestock health but also poses sign

Development of a Rapid Diagnostic Test to Distinguish between Emerging Viruses That Cause Hemorrhagic Fever.

Viruses that cause the clinical syndrome referred to as viral hemorrhagic fever (VHF) are responsible for numerous infectious disease outbreaks. High-priority emerging viruses include orthoebolaviruses, orthomarburgviruses, Lassa virus, Crimean-Congo hemorrhagic fever virus, Rift Valley fever virus (RVFV), dengue virus (DENV), and yellow fever virus (YFV). Many of these viruses cause a similar clinical presentation in infected humans and have an overlapping geographic distribution with a risk of coemergence. As such, an antigen rapid diagnostic test to distinguish between these viruses would b

Rift Valley fever: A zoonotic disease with global potential.

An arthropod-borne zoonotic disease called "Rift Valley fever (RVF)" spreads widely among ruminant animals and humans. RVF is caused by the RVF Virus (RVFV), a round-enveloped RNA virus belonging to the genus Phlebovirus and family Bunyaviridae. RVF is found exclusively in African nations, and it is primarily associated with high rainfall and dense vector mosquito populations. The virus moves from its initial replication site to vital organs, such as the brain, liver, and spleen, after infection. These organs either recover due to both general and particular host responses, or they are affecte

Altered histone modifications in Aedes aegypti following Rift Valley fever virus exposure.

When arthropod-borne viruses (arboviruses) are delivered to vector mosquitoes in an infectious bloodmeal, viral components interact with host proteins to hijack cells and initiate replication. The extent to which arbovirus infection alters mosquito host transcriptional and genomic regulatory processes is currently unknown. We hypothesized that histone modifications would be altered in mosquitoes exposed to Rift Valley fever virus (RVFV MP12, Phlebovirus riftense, family Phleboviridae). We interrogated transcriptome and chromatin landscapes in Aedes aegypti midguts by performing Cleavage Under

Modeling sparse Rift Valley fever incidence data: a Bayesian perspective on zero-inflated self-exciting and autoregressive models.

BACKGROUND: Rift Valley fever (RVF) is a mosquito-borne zoonotic disease for which predictive modeling is often hindered by sparse data, particularly the high frequency of zero counts in both human and livestock surveillance systems. While zero-inflated models are commonly used for sparse data, several temporal count modelling frameworks exist, including less common self-exciting models that assume an initial case increases the likelihood of subsequent cases. METHODS: This study compares three zero-inflated Bayesian models: the negative binomial (ZINB) with autoregressive temporal random effe

Local drivers of Rift Valley fever outbreaks in Mauritania: A one health approach combining ecological, vector, host and livestock movement data.

Rift Valley fever (RVF) is a vector-borne zoonotic disease with recurrent epidemic and epizootic outbreaks in Mauritania caused by the RVF virus (RVFV). In recent years, outbreaks have occurred with increasingly shorter inter-epidemic periods. The primary objective of this study was to utilise a high-resolution spatiotemporal model and identify the drivers and ecological suitability for RVFV infections, as well as areas for RVF outbreaks and emergence in humans and animals, respectively, in Mauritania. We used geolocated data from 2019 to 2023 for modelling, including human RVF cases confirmed

Rift Valley fever seroprevalence in ruminants in Dhobley town, Lower Juba region, Somalia, in 2021.

This study assesses the seroprevalence of Rift Valley fever (RVF) in ruminants in Dhobley, Somalia, following a 2021 outbreak in Kenya. Among 142 ruminants sampled, 4.9% were seropositive for RVF virus (RVFV) antibody, with IgM antibodies (1.4%) indicating recent exposure, though no cases were RT-PCR-positive. Unregulated livestock movement and limited surveillance pose significant risks for future outbreaks, underscoring the need for enhanced surveillance systems and One Health strategies.

Harnessing eDNA metabarcoding for mosquito surveillance: a pilot study in Ethiopia's Great Rift Valley.

BACKGROUND: Vector-borne diseases account for over 17% of all infectious diseases worldwide, with malaria and mosquito-borne arboviruses emerging as a significant and escalating threat. In Ethiopia, malaria and arboviruses such as Dengue and Yellow fever viruses pose substantial public health challenges; however, surveillance efforts are hindered by traditional methods that are costly and labor-intensive, yet unable to identify cryptic species. Environmental DNA (eDNA) metabarcoding presents a promising alternative, facilitating noninvasive and high-throughput detection of vector species direc

Rift Valley Fever Virus: An update on current status and future prospects.

Rift Valley fever is a mosquito-borne disease caused by the Rift Valley Fever Virus (RVFV) belonging to the genus Phlebovirus. This virus causes febrile or hemorrhagic illness in humans and ruminants, such as abortion, and death; especially in young sheep, cattle, and goats resulting in devastating epidemics in Africa and the Arabian Peninsula. The WHO has included this virus in Bluepoint's list of eight pathogens. This virus is a crucial health concern in the Kingdom of Saudi Arabia (KSA), as the Kingdom is regularly exposed to this virus from the original source of East African countries. A

When nothing happens: could Theileria parva-like endemic stability explain Rift Valley fever's cryptic persistence during the inter-epidemic periods?

The epidemiological pattern of Rift Valley fever (RVF) is marked by explosive epidemics followed by prolonged inter-epidemic periods (IEPs), during which the virus circulates at low levels without apparent outbreaks. This technical note proposes a conceptual framework comparing RVF maintenance during IEPs to Theileria parva endemic stability in cattle. Despite differences in pathogen and vector biology, both diseases display similar dynamics that sustain a balanced host-pathogen relationship. We identify three key features supporting this comparison: continuous low-level exposure through persi

A Single Dose of Live-Attenuated Rift Valley Fever Virus Vector Expressing Peste Des Petits Ruminants Virus (PPRV) H or F Antigens Induces Immunity in Sheep.

INTRODUCTION/BACKGROUND: Rift Valley fever virus (RVFV) and peste des petits ruminants virus (PPRV) are significant pathogens affecting small ruminants, causing substantial economic losses in the affected regions. The development of effective vaccines against both viruses is crucial for disease control. Recombinant viruses expressing heterologous antigens have shown promise as multivalent vaccine candidates. Unlike conventional PPRV vaccines, our recombinant RVFV-vectored vaccines offer a novel dual-protection strategy against RVF and PPR, combining safety, immunogenicity, and a DIVA strategy.

Rift Valley Fever Virus Transmission During an Unreported Outbreak Among People and Livestock in South-Central Tanzania.

Rift Valley fever (RVF) is a re-emerging vector-borne zoonotic disease that causes outbreaks in humans and animals across Africa. To better understand RVF at human-animal interfaces, a prospective longitudinal survey of people, livestock, and mosquitoes was conducted from 2016 to 2018, in two regions of Tanzania, with distinct climatic zones (Iringa and Morogoro). Molecular and serological tools for testing (RT-qPCR and IgM/IgG ELISA) for RVF virus (RVFV) were used to assess infection and exposure in people and animals. Mosquitoes were collected quarterly from 10 sentinel locations. In total,

Immune Evasion by the NSs Protein of Rift Valley Fever Virus: A Viral Houdini Act.

Rift Valley fever virus (RVFV) is a negative-sense arbovirus that causes several severe diseases, including hemorrhagic fever in ruminants and humans. There are currently no FDA-approved vaccines or therapeutics for RVFV. The viral nonstructural protein NSs acts like a molecular Harry Houdini, the world-famous escape artist, to help the virus evade the host's innate immune response and serves as the main virulence factor of RVFV. In this review, we discuss the molecular mechanisms by which NSs interacts with multiple factors to modulate host processes, evade the host immune response, and facil

Targeting the Rift Valley Fever Virus Polymerase: Resistance Mechanisms and Structural Insights.

Rift Valley fever virus (RVFV) is an arbovirus from the Phenuiviridae family that can cause severe disease in humans and livestock, with outbreaks resulting in substantial economic losses. Despite the availability of attenuated vaccines for animals, there is no approved preventive or therapeutic agent for human RVFV infections. Moreover, the safety and efficacy of the current veterinary vaccines remain uncertain. The RVFV L protein, a 250 kDa polymerase, plays a key role in viral replication and transcription, containing endonuclease, RNA-dependent RNA polymerase (RdRp), and cap-binding domain

A system to rapidly develop a bunyavirus pseudotyped virus neutralisation assay for pandemic preparedness.

Several Bunyavirales families have been listed as being of high pandemic or epidemic risk by the WHO R&D Blueprint. To support pandemic preparedness and the 100 Days Mission, along with rapid provision of vaccines and therapeutics, the development of tools to assess the immune response is required. Pseudotyped viruses (PV) have been shown to be a suitable alternative to authentic infectious virus to measure virus-neutralising activity, a key component of the immune response. They alleviate the need to acquire and amplify viral isolates and do not require high containment facilities. Generating
