Detection and genetic diversity of Wolbachia and its associated prophage WO in mosquito populations from Ethiopia.

Arboviral diseases transmitted by mosquitoes, including dengue, Chikungunya, Zika, yellow fever, West Nile virus (WNV), and Rift Valley fever (RVF), pose a significant public health challenge globally, particularly impacting populations in low and middle-income countries. Conventional mosquito control methods, which primarily rely on insecticides, face critical challenges, including the development of insecticide resistance and environmental concerns. In this context, Wolbachia, an endosymbiotic bacterium, presents an alternative strategy due to its ability to manipulate mosquito reproduction

JAK inhibitors remove innate immune barriers facilitating viral propagation.

Janus kinase (JAK) inhibitors are small-molecule therapeutics that reduce inflammation in autoimmune and inflammatory diseases by modulating the JAK-STAT pathway. While effective in alleviating immune-mediated conditions, JAK inhibitors can impair antiviral defences by suppressing interferon (IFN) responses, potentially increasing susceptibility to viral infections. This study investigates the pro-viral mechanism of JAK inhibitors, focusing on baricitinib, across various cell lines, organoids, and viral strains, including a recombinant Rift Valley fever virus, influenza A virus, SARS-CoV-2, an

Site-directed tyrosinase conjugation on engineered ferritin retains immune recognition.

Ferritin (Ft) nanoparticles are promising scaffolds for antigen display in vaccine design due to their stability, defined architecture, and biocompatibility. Enzymatic methods, such as tyrosinase catalysis, enable covalent antigen conjugation by oxidizing tyrosine residues into o-quinones that react with accessible cysteine thiols. Here, we engineered Pyrococcus furiosus ferritin (PfFt) by introducing single cysteines at defined positions (K8C, D33C, and E92C) to enable site-specific bioconjugation. All PfFt variants retained their quaternary nanoparticle structure, as confirmed by mass spectr

Incidence rate of Rift Valley fever exposure in humans and livestock from a longitudinal study in Northern Kenya.

Unravelling the mechanisms of Rift Valley fever virus (RVFV) maintenance in endemic areas during interepidemic periods is critical for enhancing early detection and response. Unfortunately, data on key epidemiological parameters, such as incidence rates, which are crucial for risk assessments and designing targeted interventions, are almost nonexistent. We conducted a longitudinal study of 1,938 pastoral livestock and 814 livestock keepers in an endemic region of northern Kenya from March 2022 to May 2023 to estimate the incidence rate of RVFV exposure and determine risk factors for infection.

A Multi-Host Approach to Quantitatively Assess the Role of Dogs as Sentinels for Rift Valley Fever Virus (RVFV) Surveillance in Madagascar.

Sentinel animals may play a key role in the surveillance of arbovirus circulation, particularly in developing countries. This study aimed to assess the relevance of using dogs as sentinel animals for Rift Valley fever virus (RVFV) surveillance in Madagascar. Serological surveys were conducted on 513 dogs and 135 cattle in the Ifanadiana district, southeastern Madagascar. In addition, 486 human dry blood samples available from the same area were used. Antibodies against RVFV were detected in 23 of 513 dogs, in 86 of 486 humans, and in 33 of 135 cattle. Serocatalytic models fitted to age-stratif

Seasonal variation in mosquito abundance and environmental predictors in semi-pastoral southern Kenya: implications for endemic Rift Valley fever.

BACKGROUND: Ecological variables that vary across time and space shape mosquito populations, creating microenvironments that can become disease transmission hotspots. Rift Valley fever virus (RVFV), a priority zoonotic arbovirus, thrives in diverse conditions and has been detected in over 50 mosquito species. This diversity complicates efforts to identify the key vectors involved in transmission and highlights the need to understand how environmental conditions shape mosquito abundance in high-risk landscapes. METHODS: This study investigated spatio-temporal variation in mosquito abundance ac

Exploring community-based reporting of livestock abortions for rift valley fever and brucellosis surveillance in Uganda: a pilot study.

Rift Valley fever (RVF) and brucellosis are serious zoonotic diseases with significant public health and economic consequences. In livestock, both diseases are associated with abortions that are not routinely reported as part of disease surveillance. We piloted a community-driven call centre initiative in Isingiro District, Uganda, from March to June 2023, aimed at facilitating the reporting of livestock abortions. The community call centre was promoted through stakeholder engagements, social media campaigns, and targeted sensitisation materials.Over three months, we received 53 alerts reporti

Molecular Characterization of Rift Valley Fever Virus From the 2025 Outbreak in Northern Senegal Reveals Lineage H Persistence and Key Polymerase Mutations.

Rift Valley fever virus (RVFV) is a mosquito-borne phlebovirus that causes severe febrile and hemorrhagic illness in humans. In September 2025, an outbreak in northern Senegal led to 119 confirmed infections and 15 deaths as of October 7, 2025. We performed rapid genomic sequencing to characterize the virus responsible for this epidemic. RNA from RT-qPCR-confirmed samples was sequenced using the Twist Comprehensive Viral Research Panel on an Illumina iSeq 100 platform. Consensus genomes were analyzed with and compared with all complete RVFV genomes in GenBank. Nine genomes were recovered, incl

Detection of Crimean-Congo hemorrhagic fever virus and Rift Valley fever virus antibodies in animal workers in Cameroon.

INTRODUCTION: Crimean-Congo hemorrhagic fever virus (CCHFV) and Rift Valley fever virus (RVFV) are emerging arboviruses primarily affecting domestic animals. Research has demonstrated their endemicity in various parts of the world, including Africa. Despite the high seroprevalence of CCHFV infection recently reported among cattle in Cameroon, the epidemiological status of animal workers who frequently interact with these animals remains poorly understood. This study investigates the seroprevalence of CCHFV and RVFV infections, along with associated factors, among animal workers in Cameroon. M

Cryo-EM structure of the Rift Valley fever virus envelope protein in complex with a potent neutralization antibody.

Entry of Rift Valley fever virus (RVFV) into host cells is mediated by the viral glycoproteins Gn and Gc. Structural details and assembly mechanism of Gn and Gc on the surface of RVFV remain unclear. Here, we stabilized the GnGc with the neutralizing monoclonal antibody RVFV-140 and determined a near-atomic resolution structure of the GnGc hexamer in complex with the fragment antigen binding (Fab) domain of RVFV-140 (Fab140). Our structure showed that RVFV-140 recognizes a ternary epitope and crosslinks two adjacent Gn heads within the hexamer, thus preventing the prefusion to postfusion trans

ZnNPs and senps co-adjuvants deliver superior and long-lasting protection: first report in inactivated Rift Valley fever vaccines for sheep.

Rift Valley Fever (RVF) poses a significant threat to livestock and human health, necessitating effective and safe vaccines. This study evaluated the immunogenicity and safety of novel inactivated RVF vaccines in sheep, enhanced with zinc oxide nanoparticles (ZnNPs) and selenium nanoparticles (SeNPs) as co-adjuvants, compared to an aluminum hydroxide gel (ALHV) adjuvanted vaccine alone. Our findings demonstrate that nanoparticle-formulated ALHV elicited a markedly faster onset of protection, achieving protective neutralizing antibody titers within 3–4 days post-vaccination, significantly accel

Seroprevalence of Crimean-Congo Hemorrhagic Fever and Rift Valley Fever Viruses Among Ruminants in Nigeria: A Descriptive Epizootiological Analysis.

Crimean-Congo hemorrhagic fever (CCHF) and Rift Valley fever (RVF) are major zoonotic diseases, spread by arthropods, with livestock serving as amplifying hosts. Despite Nigeria's large ruminant population and robust cross-border animal trade, data on the seroprevalence of the viral agents causing these diseases remain limited. A longitudinal serological survey was conducted in five major livestock markets across Nigeria. A total of 3450 animals (cattle, sheep, and goats) were tested for Crimean-Congo hemorrhagic fever virus (CCHFV) and Rift Valley fever virus (RVFV) antibodies using ELISA. Da

LRP1 Interacts with the Rift Valley Fever Virus Glycoprotein Gn via a Calcium-Dependent Multivalent Electrostatic Mechanism.

The Rift Valley fever virus (RVFV) is a highly pathogenic, mosquito-borne zoonotic virus that poses a significant risk to livestock, human health, and global public health security. Although RVFV is classified by the World Health Organization (WHO) as a priority pathogen with epidemic potential, no licensed vaccines or effective antiviral therapies are currently available. A limited understanding of the molecular mechanisms of RVFV entry has hindered therapeutic development. Here, we elucidate the molecular basis by which the RVFV envelope glycoprotein Gn recognizes its receptor, low-density l

Efficient Expression of Oropouche Virus Nonstructural Proteins NSs and NSm.

Oropouche fever, a mosquito- or midge-borne emerging zoonotic disease endemic to South and Central America, manifests as a dengue-like acute febrile illness with occasional occurrences of meningitis or meningoencephalitis. The causative agent, Oropouche virus (OROV), belongs to the genus Orthobunyavirus within the family Peribunyaviridae. Its tripartite negative-sense RNA genome comprises small (S), medium (M), and large (L) segments, encoding structural N, Gn/Gc, and L proteins, respectively. Additionally, the S- and M-segments encode nonstructural proteins: NSs and NSm, which may act as viru

An orally available 4'-fluorouridine prodrug inhibits SFTSV and LCMV infection.

Bunyaviruses, a subset of segmented negative-sense RNA viruses, include pathogenic species capable of zoonotic transmission to humans via arthropod vectors and rodent hosts. Pathogenic bunyavirus infections can cause severe hemorrhagic fevers and other life-threatening diseases, posing threats to human health and social stability; however, therapeutic strategies for treating bunyavirus infections remain limited. Here, we report that VV251 hydrochloride salt (VV251), an optimized oral prodrug derivative of 4'-fluorouridine (EIDD-2794), exhibits potent efficacy against severe fever with thromboc

Evaluating the potential of anti-dsRNA antibodies as an alternative viral sensing tool in encephalitides of different species.

Although laboratory methods have advanced, the cause of many encephalitides is still unknown. Molecular methods like multiplex PCR and microarrays are considered to be often less sensitive than Next Generation Sequencing, whereas the latter is time-consuming and costly. These analyses require appropriate tissue preparations and are more difficult to perform on formalin-fixed, paraffin-embedded (FFPE) tissues. Anti-double-stranded RNA (dsRNA) antibodies could potentially identify virus infections independently of the viral genome and can be applied to FFPE material. This study examined the appl
