Assessing the Vector Competence for Chikungunya Virus in Mosquitoes.

Chikungunya virus (CHIKV) is a human pathogenic, mosquito-borne virus (arbovirus), which is causing epidemic outbreaks among human populations in Africa, Asia, Europe, and South- and Central America including the Caribbean. The development of novel approaches to prevent mosquito transmission of the virus in the field requires a detailed study of CHIKV interactions with its mosquito vectors in the laboratory. In this chapter, we describe how to prepare Aedes aegypti mosquitoes for the infection with freshly cultivated CHIKV. We also describe quantitative and qualitative viral detection assays i

A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

Wesselsbron virus at the livestock-human interface: A practical guide to disease recognition and diagnosis.

Wesselsbron virus (WSLV) is a mosquito-borne Orthoflavivirus of the yellow fever virus (YFV) serocomplex, widely distributed across sub-Saharan Africa. It is neglected, and its true impact is likely underestimated. WSLV causes abortion, congenital malformations, and neonatal mortality in sheep, the primary recognized vertebrate host, but infections have also been documented in goats, cattle, camels, wildlife, and humans. The virus is maintained in complex cycles predominantly involving Aedes mosquitoes and incompletely defined domestic and wild vertebrate hosts. Direct infection via the placen

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri

Early-warning surveillance of West Nile and Usutu viruses in wetland and larval habitat waters and in mosquito excreta using digital PCR.

West Nile virus (WNV) and Usutu virus (USUV) are mainly avian mosquito-borne pathogens, increasingly cause human and equine disease in temperate regions. Mostly asymptomatic, current surveillance in France is reactive, based on clinical and veterinary reports, delaying signals for vector control and preventative interventions. We implemented in Southern France, in 2024 and 2025, a proactive surveillance methodology combining molecular xenomonitoring of mosquito excreta and environmental water detection of viral RNA in wetlands and mosquito larval habitat, through a multiplex reverse transcript

Evidence of West Nile Virus Infections in Wild Boars (Sus scrofa) in the Netherlands, 2018-2021.

INTRODUCTION: Orthoflaviviruses are emerging in Europe and have potential significant impact on animal and public health. Usutu virus (USUV) and West Nile virus (WNV) have been first detected in the Netherlands in 2016 and 2020, respectively. Wild boars (Sus scrofa) have been suggested as an interesting population to detect the circulation of orthoflaviviruses. We aim to provide epidemiological insights of USUV and WNV infections in wild boars in the Netherlands. We further explore the interpretation of serological results, taking into account cross-reactivity patterns. METHODS: A total of 15

Seroprevalence and durability of West Nile virus (WNV) neutralizing antibody responses in asymptomatic adults.

OBJECTIVE: West Nile virus (WNV) is a mosquito-borne pathogen of growing public health concern that is endemic in Israel and has been responsible for intermittent outbreaks. The seroprevalence and durability of functional antibody responses in asymptomatic WNV-infected populations remain unclear. METHODS: We screened a large cohort of asymptomatic healthcare workers (HCWs) for WNV IgG and neutralizing antibody titers. Predictors of seropositivity and neutralization levels were assessed using multivariable regression. Among seropositive individuals, longitudinal samples, collected up to 44 mon

Imported mosquito-borne infectious diseases in travellers and migrants: Global patterns, clinical overlap, and prevention gaps-an umbrella review of 21 systematic reviews.

BACKGROUND: Imported mosquito-borne infectious diseases (MBIDs) pose public health challenges in nonendemic regions, but cross-disease evidence remains fragmented. We conducted an umbrella review of systematic reviews to identify gaps and inform clinical practice and research priorities. METHODS: Five databases (PubMed, Embase, Scopus, Web of Science, and the Cochrane Library) were searched from inception to December 3, 2025, for systematic reviews on nine imported MBIDs, including chikungunya, dengue, Rift Valley fever, yellow fever, Zika, lymphatic filariasis, malaria, Japanese encephalitis

Unveiling the antiviral mechanism of A4S7: a synthetic antimicrobial peptide targeting dengue and other mosquito-borne enveloped virus infections.

Dengue virus (DENV), an enveloped mosquito-borne flavivirus, is the causative agent of dengue and prevalent in tropical and subtropical regions. Severe cases can progress to dengue hemorrhagic fever or shock syndrome, posing significant public health challenges. In the absence of specific antiviral drugs, antimicrobial peptides (AMPs) have emerged as promising therapeutic agents against DENV due to their broad-spectrum antibacterial and antiviral properties. This study investigated the synthetic AMP A4S7, which effectively inhibited DENV-2 RNA and protein expression and reduced viral progeny p

Longitudinal dynamics and lineage cross-reactivity of the human antibody response to West Nile virus.

West Nile virus (WNV) is an emerging/re-emerging flavivirus representing one of the major global causes of mosquito-borne encephalitis, yet no approved therapeutics or vaccines currently exist. To characterize the kinetics of WNV-specific immunity, we evaluated the humoral response in a longitudinal cohort of 55 WNV lineage 1-infected blood donors up to 1 year. We demonstrate that WNV infection elicits robust and durable binding and neutralizing antibodies. Using a biphasic decay model, we estimated the half-life of neutralizing antibodies to be 37 days during the rapid decay phase and 472 day

Genetic diversity of West Nile virus lineage 2a circulating in Culex pipiens vector populations from Bucharest city during the 2024 and 2025 transmission seasons.

BACKGROUND: West Nile virus (WNV) is an emerging pathogen in Europe with an unprecedented northward expansion. In Romania, the virus has been endemic for at least three decades, and human infection cases are registered yearly, sometimes culminating in outbreak events. Bucharest, the capital city of Romania, is the largest urban area of the country, encompasses a multitude of mosquito and bird habitats, and has been continuously reported as an active hotspot for WNV transmission. The circulation of lineage 2 has been documented in Bucharest since 2010 in mosquito and human populations. Our stud

Isolation and molecular characterization of West Nile Virus with evidence of vertical transmission in the Coastal region, Kenya.

West Nile virus (WNV) is a mosquito-borne flavivirus of global public health importance, maintained in an enzootic cycle between birds and mosquitoes, with humans and other mammals as incidental hosts. Although WNV has been documented in Kenya, surveillance remains inconsistent, geographically fragmented, and largely dependent on serological methods. This limits understanding of circulating lineages, vector diversity, and transmission dynamics, particularly in coastal regions. This study aimed to address these gaps through systematic arboviral surveillance of mosquito populations across four e

Role of Cellular Mechanisms in Dengue Pathogenesis: Focus on Immune Cells Interactions.

Dengue is an arbovirus mosquito-borne disease that occurs after an infection with dengue virus. Dengue virus releases E-proteins, which act as binding proteins and enter the host cell after infection. It triggers several cellular reactions and activates the immune system; however, the mechanisms are still poorly understood. Our goal is to find out how these cellular interactions participate in the activation of immune cells and participate in dengue pathogenesis. Once dengue infects the host cell, it follows these steps: (1) dengue virus releases M- protein into the skin of the host, and it in

Dengue Fever: Viral, Environmental, and Human Factors Driving Expansion and Pandemic Risk.

Dengue fever (DF), caused by the dengue virus (DENV), is the fastest-spreading mosquito-borne viral disease globally and presents significant public health challenges. This comprehensive review explores the multifaceted factors influencing DENV expansion and control. We examine viral factors, including serotypes, sub-genotypes, genetic mutations, recombination, and immune evasion mechanisms, and their role in enhancing dengue's adaptability and virulence. The influence of environmental factors, such as climate change, global warming, and unplanned urbanization on mosquito vector distribution,

In Vivo Risk Assessment of Yellow Fever Virus Transmission Through Breastfeeding, and Mechanistic Insights.

BACKGROUND: Yellow fever virus (YFV), a mosquito-borne orthoflavivirus, remains a significant public health concern, especially in regions with low vaccine coverage. Since 2010, yellow fever vaccination is not recommended for breastfeeding women due to reported cases of vaccine strain transmission through breast milk causing neonatal meningoencephalitis. However, breastfeeding transmission efficiency of the vaccine strains remains unknown, and wild-type strain transmission has been suggested following viral RNA detection in milk. Obtaining direct evidence of breastfeeding-related transmission

Platforms to Evaluate Flavivirus-Induced Immunoinflammatory Responses and Their Pathogenesis.

Mosquito-borne diseases induced by viruses belonging to the Flavivirus genus such as Dengue, Zika, and Yellow fever have proven to cause widespread outbreaks over time. These viruses are prevalent mainly in tropical and subtropical regions and it is estimated that more than 100 countries are affected by one or more of these pathogens. The diseases may have mild clinical manifestations in common, such as fever, joint pain, headaches, and diarrhea; however, they can progress to serious reactions and, sometimes, trigger significant neurological malformations and post-infection sequelae, as in inf
