Ianalumab for the Treatment of Patients with Sjögren's Disease.

BACKGROUND: Sjögren's disease is a complex, systemic autoimmune disease with no approved targeted therapies. Ianalumab is an IgG1 monoclonal antibody with a dual mechanism of action, depleting B cells through antibody-dependent cellular cytotoxicity and inhibiting B-cell activation and survival through blockade of B-cell activating factor receptor. METHODS: We conducted two 52-week, double-blind, randomized, placebo-controlled trials involving patients with active Sjögren's disease. Patients in the NEPTUNUS-1 trial were assigned in a 1:1 ratio to receive subcutaneous ianalumab (300 mg monthly

Hollow Mesoporous Polydopamine Nanocarriers Loaded With Curcumin Nanoparticles Modulate Macrophage Polarization and Support Osteogenic Differentiation: Association With Acat3/Fatty Acid Oxidation-Related Metabolic Changes.

Periodontitis is a chronic inflammatory disease driven by dysregulated host immune responses. Macrophages, as key innate immune cells, play important roles in periodontal inflammation and bone homeostasis, and M2-like macrophage polarization has been associated with improved osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Curcumin possesses anti-inflammatory and bone-protective properties, but its application is limited by poor aqueous solubility, chemical instability, and low bioavailability. In this study, curcumin nanoparticles (CUR NPs) were prepared by a reprecip

Mass Spectrometry-Based Oral Meta-Omics Analysis-What We Know and What We Don't.

According to the World Health Organization, dental caries and periodontal disease are the most prevalent oral health conditions worldwide, usually linked with oral microbiome dysbiosis. It is assumed that oral microbiota alterations are also associated with immune challenges, beyond the oral cavity, thus having implications in the progression of various systemic diseases, but the mechanism by which the plaque biofilm microbiota contributes to the development of such diseases is not entirely clear. In this context, metaproteomics and mass spectrometry-based metabolomics can reveal essential inf

Schwann cell-derived extracellular vesicles promote periodontal tissue regeneration.

Periodontitis is a common chronic inflammatory disease worldwide, and conventional therapies control inflammation but fail to achieve predictable regeneration. The inferior alveolar nerve (IAN) is a key peripheral nerve that governs mandibular bone homeostasis, and its disruption precipitates bone metabolic imbalance and impaired repair. However, the dynamic behavior of Schwann cells (SCs), the principal glial cells of this neural axis, and their mechanistic influence on periodontal ligament stem cells (PDLSCs) remain elusive. We established a rat IAN transection model and found that denervati

Central nervous system, cranial, and cerebrovascular conditions beyond the Kimonis criteria in patients with Gorlin syndrome: a retrospective cohort study.

BACKGROUND: Gorlin syndrome is a multisystem developmental and cancer-predisposing disorder commonly caused by germline PTCH1 variants. The Kimonis clinical criteria capture the canonical phenotype; however, central nervous system (CNS), cranial, and cerebrovascular conditions documented beyond these criteria remain poorly defined. METHODS: We conducted a retrospective descriptive cohort study of 32 patients who fulfilled the Kimonis clinical criteria and received care at either or both hospitals in Japan. Clinical information through March 31, 2026, including sufficiently documented care els

Risk factor analysis and development of a prediction model for trismus after surgical treatment of buccal mucosa squamous cell carcinoma.

BACKGROUND: Buccal mucosa squamous cell carcinoma (BMSCC) arises in a functionally complex anatomical region adjacent to the buccinator muscle, the anterior border of the masseter muscle, the pterygomandibular raphe, the retromolar region, and the pterygomandibular space. Tumor infiltration, extended resection, post-reconstruction scarring, and postoperative radiotherapy may affect the masticatory muscle complex and mandibular movement. Compared with other oral subsites, such as tongue and floor-of-mouth cancers, postoperative trismus after BMSCC has more pronounced subsite-specific characteri

Salivary miR-21 and miR-31 in Oral Squamous Cell Carcinoma: Molecular Pathogenesis, Diagnostic Significance, and Translation to Isothermal RPA.

Oral Squamous Cell Carcinoma (OSCC) is often diagnosed at advanced stages despite the oral cavity's anatomical accessibility, highlighting the need for molecular adjuncts that can identify disease-associated changes before overt morphological progression. This review evaluates the biological roles and salivary diagnostic potential of microRNA-21 (miR-21) and miR-31, two miRNAs repeatedly associated with OSCC pathogenesis and progression. We examine current clinical evidence supporting their use as salivary biomarkers and discuss major sources of methodological variability, including difference
