The Nox2 NADPH oxidase regulates neutrophilic inflammation in the oral cavity.

The leukocyte NADPH oxidase 2 (Nox2) is an important regulator of inflammatory responses, independent of its antimicrobial activity. Inactivating mutations in NOX2 cause chronic granulomatous disease (CGD), a severe immunodeficiency associated with recurrent infections and dysregulated neutrophilic inflammation. Recurrent oral ulcers, stomatitis, gingivitis, and other inflammatory issues affecting the oral mucosa have been observed in patients with CGD; however, the underlying mechanisms are not known. Here, we present evidence that the extensive inflammatory destruction of oral mucosal tissue

Nanocurcumin in Oral Disease Therapy: A Review of Innovations and Future Prospects.

Curcumin exhibits strong anti-inflammatory, antioxidant, and anticancer activities. However, its clinical use is limited due to poor solubility, low gastrointestinal absorption, and rapid systemic metabolism. Nanocurcumin offers enhanced solubility, bioavailability, and targeted delivery through systems such as nanoparticles, liposomes, micelles, and nanoemulsions. In oral health, nanocurcumin has shown significant therapeutic promise. In periodontitis models, it attenuates pro-inflammatory cytokines and oxidative stress markers. In radiation- and chemotherapy-induced oral mucositis, randomize

Early-stage pharmaceutical development of bilayer buccal films with a chitosan backing layer for local pain management in oral mucositis.

Oral mucositis is a painful inflammation and ulceration of the oral mucosa that frequently occurs as a side effect of radiation and chemotherapy that impairs patients' quality of life, making eating and drinking challenging. Local anesthetics applied directly to lesions can provide temporary pain relief without substantial systemic exposure. Buccal films are a promising delivery platform because they protect the sore area while releasing anesthetics locally to the inflamed mucosa. In this work, the mucoadhesive and swelling properties of chitosan were exploited as a backing layer in bilayer fi

A systematic review and meta-analysis of single-application topical doxycycline for recurrent aphthous stomatitis.

Recurrent aphthous stomatitis (RAS), an oral mucosal pathosis, can interfere with patients' quality of life, mainly due to its recurrent nature. The etiology of RAS remains unclear and its management is challenging. This systematic review and meta-analysis were performed to assess the efficacy of a single topical application of doxycycline to treat RAS. Online search of the Scopus, Medline (via PubMed), Embase and Google Scholar databases was performed from May to September 2025. Randomized controlled trials (RCTs) were selected, eligible studies were conducted for healthy patients with the pr

The natural history of paraquat-associated stomatitis: A four-stage model.

BACKGROUND: Paraquat-associated stomatitis (PAS) is a poorly characterized feature of paraquat poisoning and is presented as a binary finding. Despite its diagnostic potential, its evolution and prognostic value remain undefined. This pooled individual-patient analysis aimed to delineate the natural history of PAS and develop a structured, clinically applicable staging system. METHODS: Data from 170 published paraquat poisoning cases were aggregated at the individual-patient level. Variables included exposure-to-symptom interval, lesion morphology, topography, and survival outcome. Temporal a

Clinicopathological and biobehavioral factors influence the onset and severity of oral mucositis in patients with head and neck cancer.

Oral mucositis is a debilitating adverse effect of radiotherapy in head and neck cancer. It progresses from erythema to painful ulcerations, often causing infections, nutritional impairment, treatment interruption, and reduced survival. While treatment-related factors are well established, patient-related risk determinants remain insufficiently defined. Thus, this study investigated the association between the onset and peak severity of radiation-induced oral mucositis in patients with head and neck cancer and their clinicopathological, biobehavioral, and demographic characteristics. To carry

Salivary proteomics suggests persistent inflammatory signatures after clinical healing in recurrent aphthous stomatitis: a hypothesis-generating case-control study.

BACKGROUND: Recurrent aphthous stomatitis is a common inflammatory condition of the oral mucosa characterized by painful ulcers that heal spontaneously but recur unpredictably. Although lesions resolve clinically, the biological processes occurring during remission remain poorly understood. This exploratory study investigated whether salivary molecular profiles persist after clinical healing and may be associated with the recurrent nature of the disease. METHODS: In this hypothesis-generating case-control study, saliva samples were collected from patients with recurrent aphthous stomatitis du

Advances in microneedle therapeutics for recurrent aphthous ulcers.

Recurrent aphthous ulcer (RAU) is a common idiopathic oral mucosal disorder marked by painful, round ulcers on non-keratinized tissues such as the buccal, labial, and lingual mucosa. These recurrent lesions impair oral function and quality of life. However, systemic treatments, such as oral or injectable medications, often exhibit limited efficacy and are associated with notable adverse effects. The effectiveness of topical treatments is often limited by the barrier properties of the oral epithelium and the dynamic oral environment. Continuous salivary flow and mechanical forces from speaking

Cytokine Toxicity and Bacterial Dysbiosis in Chemotherapy- and/or Radiotherapy-Induced Oral Mucositis: Pathophysiological Mechanisms and Therapeutic Interventions.

Chemotherapy- and/or radiotherapy-induced oral mucositis (CRIOM) is a common complication in patients with head and neck cancer, driven largely by excessive proinflammatory cytokine signalling and treatment-associated bacterial dysbiosis. This narrative review synthesizes current mechanistic evidence and summarizes emerging therapeutic strategies targeting these pathways. Research indicates that elevated levels of IL-1β, IL-6, TNF, iNOS, and nitric oxide amplify tissue injury and ulceration, while disruption of oral and gut microbial communities, characterized by loss of beneficial commensals

Therapeutic Outcomes of Aloe vera Gel Compared with Triamcinolone Acetonide in Minor Aphthous Stomatitis.

BACKGROUND: Recurrent aphthous stomatitis (RAS) is a common oral mucosal condition characterized by painful, recurrent ulcers that affect quality of life. Topical corticosteroids such as triamcinolone acetonide are the standard treatment due to their anti-inflammatory effects, but concerns about side effects and patient preference have prompted interest in alternative therapies. Aloe vera, known for its anti-inflammatory and wound-healing properties, has emerged as a potential natural option. However, comparative evidence with standard treatments remains limited, necessitating further evaluati
