The orofacial cleft risk gene IRF6 is a target gene of SOX9 in cranial neural crest cells.

Orofacial clefts represent the most prevalent congenital anomalies affecting the craniofacial region. They can be evoked by a disturbed development of either oral epithelium or cranial neural crest-derived mesenchyme, or by disruptions of their interplay. IRF6 is a well-known risk gene associated with non-syndromic orofacial clefts and causative for Van der Woude syndrome, an autosomal dominant syndrome characterized by orofacial clefting and lower lip pits. Its expression in the oral epithelium depends on the transcription factor TFAP2A. We here show by immunofluorescence on mouse embryonic s

Cleft Lip and Palate is Common in PORCN-Related Focal Dermal Hypoplasia in Asians: Three New Case Reports and Literature Review.

Focal dermal hypoplasia is an X-linked dominant genetic disorder predominantly affecting females, caused by loss-of-function variants in the PORCN gene, characterized by ectodermal, skeletal, craniofacial and ocular structural abnormalities. We report three cases with de novo novel variants, and performed a comprehensive review of the clinical features of this disorder focusing on Asian patients, and identified significant differences compared with previous literature focusing on Caucasian patients. Cleft lip and palate is a frequently reported feature in Asian patients, but not in Caucasians.

Investigating global gender gap in lip and oral cavity cancer: A population-based study.

INTRODUCTION: Lip and oral cavity cancer (LOC) is typically male predominant, yet in several countries women experience a comparable or even higher burden. Quantifying gender differences in LOC and their social determinants is essential for equitable cancer control. MATERIALS AND METHODS: We analysed Global Burden of Disease 2021 estimates for LOC from 1990 to 2021 across 204 countries and territories. Gender disparities were assessed using the Female-to-Male disability-adjusted life years (DALYs) ratio. Temporal trends were evaluated using estimated annual percentage change (EAPC), and forec

Effectiveness of digitally- versus conventionally-fabricated nasoalveolar molding appliances for presurgical approximation of complete cleft lip and palate: A systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Presurgical nasoalveolar molding (NAM) has become an integral component of comprehensive cleft lip and palate (CLP) care. The aim of the present systematic review and meta-analysis is to assess the efficacy of digitally versus conventionally fabricated NAM appliances for presurgical approximation of complete CLP. METHODS: The focused research question is "In patients with CLP, does the use of digitally-fabricated NAM appliances yield superior presurgical cleft approximation outcomes than conventionally-fabricated NAM appliances?" Literature search was conducted in accordance with

Oral and maxillofacial manifestations associated with Kabuki syndrome: A systematic review.

BACKGROUND: Kabuki make-up syndrome (KMS) is a rare genetic disorder characterized by distinctive facial features and multiple congenital anomalies. Although the facial phenotype is central to diagnosis, the spectrum of oral and maxillofacial manifestations has not been comprehensively summarized. The aim of this study was to systematically review the craniofacial, oral, and dental features associated with KMS. METHODS: This systematic review was conducted in accordance with PRISMA guidelines. Searches were conducted in 5 databases supplemented by citation chasing and gray literature, using t

Complex Salvage Lower Lip Reconstruction Using Gracilis Free Flap.

BACKGROUND: The gracilis muscle microneurovascular free tissue transfer is a workhorse in dynamic facial reanimation. For patients with prior free flap reconstruction for large lower lip defects and in cases where local or regional tissue flap is inadequate for reconstruction, the gracilis free flap should be considered as a standard reconstructive option. OBJECTIVES: To define salvage lip reconstruction with gracilis muscle free flap. METHODS: We performed chart review of four patients with history of total lower lip defects with prior free flap reconstruction who underwent salvage lip reco

Associations Between Alcohol Dehydrogenase Mutations and Cleft Lip/Palate: A Scoping Review.

BackgroundThe incidence of cleft lip with or without cleft palate (CL ± P) varies significantly according to genetic ancestry. The alcohol dehydrogenase (ADH) family comprises several enzymes with polymorphisms that are also highly disparate between genetic backgrounds. A small number of heterogeneous studies have linked ADH variants with risk of CL ± P. Therefore, the authors conducted a scoping review to better understand these potential associations.MethodsThis review was conducted following PRISMA 2020 guidelines, screening for studies published in PubMed and Medline databases.
