Laser-assisted photodynamic therapy for oral leukoplakia with oral submucous fibrosis: A case series.

Oral submucous fibrosis (OSF) and oral leukoplakia (OLK) are both potentially malignant disorders. Patients with habits of betel quid chewing and smoking are frequently diagnosed with both conditions. Notably, the rate of malignant transformation is significantly higher in individuals with concurrent OSF and OLK than in those with either condition alone. Laser assisted photodynamic therapy (PDT) is a therapeutic modality that has been applied in the management of OLK, though its use in OSF remains unveiled. In this study, we report three patients with concurrent OSF and OLK who underwent laser

Evaluation of a multi-component mucoadhesive buccal patch: cytokine-associated inflammatory responses and tissue remodeling in experimental models.

INTRODUCTION: Dysregulated cytokine signaling is implicated in persistent inflammation and fibrosis in oral submucous fibrosis (OSMF) and has been reported in association with pathways observed in oral squamous cell carcinoma (OSCC). This study investigated cytokine associated inflammatory and fibrotic responses following treatment with a mucoadhesive buccal patch containing isotretinoin, bromelain, and limonene. METHODS: The formulation was evaluated using lipopolysaccharide (LPS) induced acute inflammatory in vitro models and a carbon tetrachloride (CCl₄) induced in vivo oral fibrotic model

Correlation of the frequency of micronucleated cells with the severity of disease process in oral submucous fibrosis and oral squamous cell carcinoma patients: A descriptive, cross-sectional study.

BACKGROUND: Exposure of a cell to carcinogens leads to an increase in the chromosomal aberrations, while such karyotypic anomalies and elevated deoxyribonucleic acid content have been observed in a plethora of oral precancerous lesions and conditions, and are expressed in the form of micronuclei. The aim of the present study was to correlate the frequency of these micronucleated cells (MNCs) with the severity of the disease process in oral submucous fibrosis (OSMF) and oral squamous cell carcinoma (OSCC) patients. METHODS: The present cross-sectional, hospital-based study consisted of 150 sub

Molecular and microenvironmental drivers of malignant transformation from oral potentially malignant disorders to oral squamous cell carcinoma.

Oral potentially malignant disorders (OPMDs) comprise a diverse group of oral mucosal lesions with a variable but significant risk of progression to oral squamous cell carcinoma (OSCC). However, predicting malignant transformation remains a major clinical challenge due to the limited reliability of conventional histopathological grading and the inherent biological heterogeneity of these lesions. Increasing evidence indicates that OPMD progression is a dynamic, multistep process driven by the convergence of molecular alterations and microenvironmental remodeling. This review synthesizes current

A Multifunctional Nucleic Acid Nanomedicine Coregulates Angiogenesis and Fibrosis to Treat Oral Submucous Fibrosis.

Oral submucous fibrosis (OSF) is a chronic progressive oral fibrotic disorder characterized by dysregulated immune inflammation, fibroblast activation, and vascular homeostasis disruption, yet synchronously regulating these axes remains challenging. We developed a multifunctional nucleic acid nanomedicine, Apt02-tFNAs-siTGF-β (A-T-S), in which tetrahedral framework nucleic acids co-deliver siRNA against TGF-β (siTGF-β) and the proangiogenic aptamer Apt02, enabling programmable integration and stable delivery. A-T-S was efficiently internalized by macrophages, human oral mucosal fibroblasts, an

Decoding the Dynamic Landscape of Sequential Disease Stages from Oral Submucous Fibrosis to Oral Squamous Cell Carcinoma.

Oral submucous fibrosis (OSF) is a chronic and progressive disease of the oral mucosa that is considered potentially malignant. However, the specific mechanism of OSF occurrence and malignant transformation into oral squamous cell carcinoma (OSCC) is still unclear. This study aimed to elucidate the spatiotemporal changes in cellular biological behaviour during the occurrence and malignant transformation of OSF, providing new insights for understanding the underlying mechanism and developing novel therapeutic strategies. Sets of normal, OSF, margin area, and OSCC tissue samples were collected,

Systematic review of antifibrotic effects of metformin: Insights from preclinical and clinical studies and potential relevance to oral submucous fibrosis.

Fibrosis involves excessive deposition of extracellular matrix, leading to ongoing tissue remodeling and organ failure. Metformin, a common antidiabetic medication, has shown potential antifibrotic effects by affecting AMP-activated protein kinase (AMPK) and transforming growth factor-beta (TGF-β) signaling pathways. This systematic review aimed to synthesize the current evidence on the antifibrotic effects of metformin and to explore its potential translational relevance to oral submucous fibrosis (OSF). The review was conducted in accordance with the PRISMA 2020 guidelines and prospectively

Comparative evaluation of salivary flow rate in patients with oral submucous fibrosis before and after transcutaneous electrical nerve stimulation.

BACKGROUND: Oral submucous fibrosis (OSMF) is a premalignant condition characterized by progressive fibrosis of the oral mucosa. It may extend to involve minor salivary glands, resulting in reduced salivary secretion and xerostomia. Pharmacological sialogogues which are traditionally used to manage hyposalivation are often associated with adverse effects, limiting their long-term use. Hence, safer non-pharmacological alternatives are warranted. AIM: To evaluate and compare salivary flow rate (SFR) in patients with OSMF before and after Transcutaneous electric nerve stimulation (TENS) therapy.

Decoding protein signatures and protein interactions in oral potentially malignant disorders: a systematic review and network analysis.

BACKGROUND: Proteomic profiling offers thorough insights into protein structure and function, as well as it acts as an essential approach for analyzing molecular changes at the tissue level. However, because of the proteome's diversity and dynamic nature, biomarker discovery remains challenging. By combining proteomics with bioinformatics, the level of understanding in relation to molecular interactions and disease processes can be improved. Through an integrative approach, few limitations can be addressed, thereby promoting proteomic profiling for the discovery of new therapeutic targets and

Evaluating Hearing Impairment in Different Histopathological Grades of Oral Submucous Fibrosis: An Audiometric Analysis.

Background Oral submucous fibrosis (OSMF) is a chronic, progressive, and potentially malignant condition primarily linked to areca nut and masala chewing. The fibrotic process not only affects the oral mucosa but may extend to adjacent structures, including the palatal and paratubal muscles, resulting in Eustachian tube dysfunction and consequent auditory impairment. Most individuals habitually place the quid on the left buccal mucosa due to right-handed dominance, which may explain the greater degree of fibrosis and more frequent auditory changes observed on the left side. Aim The present stu
