Analysis of Cell Populations from Arbovirus-Infected Tissues by Flow Cytometry.

Arboviruses still represent a major challenge to public health in many parts of the world. Despite shared epidemiological characteristics, each arboviral disorder displays distinct cellular tropism and activation patterns, progressing to immunopathological outcomes that remain not fully elucidated. A deeper understanding of these processes requires methodologies capable of characterizing the often-complex cellular responses. In this context, flow cytometry emerges as a valuable tool, enabling the characterization of heterogeneous cell populations, the assessment of activation and proliferation

No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so they are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1-/-, and Ifnar1-/- Ifngr1-/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV), Sokuluk vi

Characterization of an Unusual Severe Yellow Fever Outbreak in the Colombian Andean Region.

BACKGROUND: Yellow fever (YF) reemerged in Colombia's Andean region during 2024-2025, outside the country's historically recognized endemic areas. We characterized critically ill adults admitted during this outbreak and described clinical features, organ dysfunction, supportive care, and in-hospital outcomes. METHODS: We conducted a prospective observational cohort study of consecutive adults with reverse transcription polymerase chain reaction (RT-PCR)-confirmed YF admitted to the national referral intensive care unit in Ibagué, Tolima, Colombia, from March through September 2025. Clinical,

Yellow Fever Outbreaks Are Not Associated With Long-Term Negative Human Perceptions of Wild Primates in Brazil.

Human perceptions of wildlife are fundamental to guiding effective strategies for conserving biodiversity and public health. Zoonotic diseases affecting non-human primates, such as yellow fever (YF), may trigger negative human behaviors toward these species. We conducted 474 semi-structured interviews across seven municipalities in Minas Gerais (n = 324 respondents) and Bahia (n = 150), Brazil, and used mixed-effects models to evaluate whether human perceptions of marmosets (Callithrix sp.) and howler monkeys (Alouatta sp.) differed according to previous YF outbreaks. YF occurrence was not con

[Bucket-list destinations from an infectious disease perspective].

Tropical countries are among the most popular destinations for Swiss travelers and pose a health challenge in terms of infectious disease risk. Arboviral diseases are one of the major infectious risks, along with rabies, hepatitis A, and malaria. Several resurgences of vaccine-preventable diseases, such as polio and yellow fever, have also been identified. In light of these data, it would appear that the safest travel destinations from an infectious disease perspective are Cuba in Latin America, Hong Kong in Asia, and the Seychelles in Africa. However, these data are subject to change and may

[Neurotropic disease associated with yellow fever vaccine: presentation as post-travel febrile syndrome].

The yellow fever vaccine is an essential tool for preventing yellow fever in people who live in or travel to areas at risk. It is highly effective, and serious adverse events -including hypersensitivity reactions, vaccine-associated viscerotropic disease, and vaccine-associated neurotropic disease (YEL-AND)-, are rare in relation to the millions of doses administered since their implementation. The incidence of YEL-AND ranges from 0.17 to 0.8 cases per 100 000 doses administered. The incidence is higher in males and in people over 60 years of age. YEL-AND can be manifested as meningoencephalit

Entomological investigations into yellow fever outbreak in northern Ghana.

Arboviruses are pathogens of emerging/re-emerging infectious diseases. Half the world's population is at risk. Recent African epidemics confirmed 19,569 cases with 820 deaths. In Ghana, Savannah Region had yellow fever (YF) outbreak in 2021. Reports revealed 35 deaths, 202 suspected cases (case fatality rate (CFR) = 17%), and 70 confirmed cases (CFR = 50%). This paper presents YF outbreak entomological investigations findings. A team with expertise in YF outbreak response were assembled and tasked to assess, contain and/or control the rapid spread of YF. Immature Aedes aegypti were collected i

NDM-Producing Escherichia coli subdural empyema mimicking chronic subdural hematoma: a case report.

BACKGROUND: Subdural empyema is a life-threatening intracranial infection that may mimic chronic subdural hematoma on non-contrast computed tomography (CT), delaying diagnosis and source control. Intracranial infection caused by New Delhi metallo-β-lactamase (NDM)-producing Escherichia coli is rare and may be overlooked when bloodstream infection appears microbiologically controlled. CASE PRESENTATION: An 82-year-old man presented with fever and progressive neurological deterioration. Chest computed tomography (CT) suggested right lower lobe pneumonia, whereas serial head CT scans showed a le

Yellow Fever Reemergence in Tolima, Colombia 2024-2025: An Eco-epidemiological Study.

BACKGROUND: Yellow fever virus is transmitted by mosquitoes among humans and non-human primates (NHPs) in South America. The 2024-2025 yellow fever (YF) outbreak was notable for its spread into new areas, including the department of Tolima in Colombia's Andean region. We investigated the eco-epidemiology of human and NHP YF cases to understand the patterns and factors associated with the Tolima outbreak. METHOD: We collected spatiotemporal, sociodemographic, and mortality data on human YF cases in Tolima, as well as the locations of deceased NHPs with YF. We then conducted exploratory, descri

Wesselsbron virus at the livestock-human interface: A practical guide to disease recognition and diagnosis.

Wesselsbron virus (WSLV) is a mosquito-borne Orthoflavivirus of the yellow fever virus (YFV) serocomplex, widely distributed across sub-Saharan Africa. It is neglected, and its true impact is likely underestimated. WSLV causes abortion, congenital malformations, and neonatal mortality in sheep, the primary recognized vertebrate host, but infections have also been documented in goats, cattle, camels, wildlife, and humans. The virus is maintained in complex cycles predominantly involving Aedes mosquitoes and incompletely defined domestic and wild vertebrate hosts. Direct infection via the placen

Temporal dissection of hepatitis C virus and orthoflavivirus infections by continuous luminescence monitoring.

Virus infection is a dynamic process consisting of rapidly evolving viral and host events, yet conventional endpoint assays provide only fragmented snapshots of these processes. Here, we established a unified temporal framework for dissecting virus-host interactions by integrating continuous bioluminescence monitoring with molecular and functional analyses. Real-time monitoring of hepatitis C virus (HCV) single-round infectious particles (SRIPs) revealed a biphasic replication profile consisting of an initial transient phase, followed by replication-dependent amplification. This transition was

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri
